원자적으로 얇은 그래핀 또는 실리콘 기반 마이크로칩과 같은 재료 생산의 최근 발전은 투과 전자 현미경(TEM)을 사용한 실시간 구조 기능 분석을 위한 새로운 기회를 제공합니다. 이러한 재료는 라이브 EM 이미징 6,7,8,9,10,11을 위해 밀폐된 챔버를 생성할 수 있습니다. 방법론적 발전이 Cryo-EM 분야의 분해능 혁명에 박차를 가한 것처럼, 과학계를 위한 고처리량 도구로서 액체-EM의 사용을 확장하기 위해서는 새로운 기술과 방법이 필요합니다. 여기에 제시된 방법의 전반적인 목표는 액체-EM 시편 준비 프로토콜을 간소화하는 것입니다. 개발된 기술의 이면에 있는 이론적 근거는 액체 및 극저온 EM 데이터 수집 모두에 적합한 새로운 마이크로칩 설계 및 오토로더 장치를 사용하는 것입니다(그림 1)7,14,15,16,17. 어셈블리는 세션당 여러 샘플을 수용할 수 있는 Krios 또는 F200C TEM과 같은 자동화 기기용 표준 그리드 클립을 사용하여 기계적으로 밀봉됩니다(그림 2). 이 방법론은 표준 Cryo-EM 애플리케이션을 넘어 고해상도 이미징의 사용을 확장하여 실시간 재료 분석의 더 넓은 목적을 보여줍니다.
