Mycobacterium tuberculosis extracellulaire blaasjes zijn zeer antigene reservoirs, die ze presenteren als een aantrekkelijke manier voor het ontwikkelen van diagnostische hulpmiddelen en toekomstige vaccins 4,19,20. Extracellulaire vesicle (EV) afgifte door pathogenen kan de sleutel zijn tot het ontsluiten van nieuwe technologieën om infectieziekten te beheersen1. Mycobacterium tuberculosis (Mtb) is een ziekteverwekker van hoge betekenis, infecteert ongeveer een derde van de wereldbevolking en eist elk jaar het leven van miljoenen mensen2. Ev-productie door Mtb is goed gedocumenteerd maar ongrijpbaar in de biogenese en gevarieerde rollen (d.w.z. immunostimulerend, immunosuppressief, ijzer- en nutriëntenacquisitie) van deze EV's in de context van infectie 3,4,5. Inspanningen om de samenstelling van Mtb EV's te begrijpen onthulden 50-150 nm lipidemembraan-ingesloten bollen afgeleid van het plasmamembraan met lipiden en eiwitten van immunologische betekenis 3,6. Onderzoek naar de rol van MTB EV's in de bacteriële fysiologie heeft het belang van bacteriële EV-modulatie als reactie op omgevingsstress voor overleving aan het lichtgebracht 5. Gastheer-pathogeen interactiestudies zijn ingewikkelder om te interpreteren, maar er zijn aanwijzingen dat Mtb EV's de immuunrespons van de gastheer kunnen beïnvloeden en mogelijk kunnen dienen als een effectieve vaccinatiecomponent 3,4,7.
De meeste studies van Mtb EV's tot nu toe hebben vertrouwd op dichtheidsgradiënt ultracentrifugatie voor blaasjesverrijking8. Dit is effectief geweest voor kleinschalige studies; deze techniek heeft echter verschillende technische en logistieke uitdagingen. Alternatieve workflows koppelen meerstaps centrifugatie, voor het verwijderen van hele cellen en groot puin, met een laatste ultracentrifugatiestap naar pellet-EV's. Deze methodologie kan variëren in efficiëntie en resulteert vaak in een lage opbrengst en co-zuivering van oplosbare niet-blaasjes geassocieerde biomoleculen, terwijl het ook de integriteit van blaasjes beïnvloedt9. Bovendien is dit proces tijdrovend, handmatig intensief en zeer beperkt in doorvoer vanwege apparatuurbeperkingen.
Het huidige protocol beschrijft een alternatieve techniek voor ultracentrifugatie van dichtheidsgradiënt: grootte-exclusiechromatografie (SEC). Deze methode is gedemonstreerd voor omgevingsmycobacteriën en in het huidige werk is het geëxtrapoleerd naar Mtb10. Een in de handel verkrijgbare kolom en automatische fractiecollector kunnen de consistentie in de vesische bereiding verbeteren en de noodzaak voor specifieke, dure apparatuur verminderen. Het is ook mogelijk om dit protocol in een fractie van de tijd te voltooien in vergelijking met ultracentrifugatie van de dichtheidsgradiënt, waardoor de doorvoer toeneemt. Deze techniek is technisch minder uitdagend, waardoor het gemakkelijker te beheersen is en kan de reproduceerbaarheid tussen / intra-laboratoria vergroten. Ten slotte heeft SEC een hoge scheidingsefficiëntie en is zachtaardig, waardoor de integriteit van de blaasjes behouden blijft.

De rol van extracellulaire blaasjes (EV's) in de context van bacteriële infectie is naar voren gekomen als een nieuwe weg voor het begrijpen van microbiële fysiologie. Specifiek spelen Mycobacterium tuberculosis (Mtb) EV's een rol in de gastheer-pathogeen interactie en reactie op omgevingsstress. Mtb EV's zijn ook zeer antigeen en hebben potentieel als vaccincomponenten. De meest gebruikelijke methode voor het zuiveren van Mtb EV's is ultracentrifugatie van de dichtheidsgradiënt. Dit proces heeft verschillende beperkingen, waaronder een lage doorvoer, lage opbrengst, afhankelijkheid van dure apparatuur, technische uitdagingen en het kan een negatieve invloed hebben op de resulterende voorbereiding. Size exclusion chromatography (SEC) is een zachtere alternatieve methode die veel van de beperkingen van ultracentrifugatie bestrijdt. Dit protocol toont aan dat SEC effectief is voor Mtb EV-verrijking en produceert hoogwaardige Mtb EV-preparaten met een verhoogde opbrengst op een snelle en schaalbare manier. Bovendien toont een vergelijking met ultracentrifugatie van dichtheidsgradiënt door kwantificerings- en kwalificatieprocedures de voordelen van SEC aan. Terwijl de evaluatie van ev-kwantiteit (nanodeeltjestrackinganalyse), fenotype (transmissie-elektronenmicroscopie) en inhoud (Western blotting) is afgestemd op Mtb EV's, kan de geleverde workflow worden toegepast op andere mycobacteriën.

Kweekfiltraateiwit (CFP) van Mycobacterium tuberculosis (Mtb) werd geconcentreerd, gekwantificeerd en vervolgens werd 3 mg materiaal aangebracht op een sec-kolom (size exclusion chromatography). De eiwit- en deeltjesconcentraties werden opgesomd door respectievelijk BCA en NTA. Het belangrijkste voordeel van deze techniek is dat het gemakkelijk uit te voeren is, waardoor het beter reproduceerbaar is.Het biedt een hoge opbrengst van Mtb EV's met minimale niet-EV eiwit co-verrijking. De procedure wordt gedemonstreerd door Joanie Ryan, een voormalige afgestudeerde student van ons lab. Bereid om te beginnen een centrifugaalfilter met moleculair gewichtsafsluiting van 100 kilodalton voor door de volledige volumecapaciteit van PBS toe te voegen.Centrifugeer gedurende vijf minuten op 2.800 maal G bij 4 graden Celsius. Gooi de flowthrough en eventuele resterende PBS weg en vul de monsterkamer met Mtb CFP tot de maximale capaciteit. Centrifugeer bij 2.800 maal G bij 4 graden Celsius totdat het volume is teruggebracht tot het minimumvolume van het ultrafiltratieapparaat.Herhaal dit zo nodig totdat het gehele monster voldoende is verkleind. Voeg 1X PBS toe aan de filtereenheid die het concentraat bevat tot de totale apparaatcapaciteit. Verminder het volume zoals eerder beschreven en herhaal deze stap vijf keer om volledig wassen en bufferuitwisseling te garanderen.Herstel vervolgens het geconcentreerde CFP-retentaat van 100 kilodalton of 100R, volgens de specificaties van het ultrafiltratieapparaat. Was het filter minstens drie keer met een minimaal volume van 1X PBS en trek de was met de 100R om het herstel te maximaliseren. Gebruik verschillende verdunning van monsters in PBS en kwantificeer het 100R-materiaal met een bicinchoninische assay.Test het monster in drievoud. Laat de grootte-uitsluitingschromatografie, of SEC-kolom, in evenwicht brengen met kamertemperatuur. Schakel de AFC in met de aan/uit-schakelaar aan de achterkant van de toreneenheid en druk indien nodig op Setup op het aanraakscherm van het hoofdmenu om de instellingen voor de loadcel aan te passen.Lijn in het installatiescherm de carrousel uit door Carrousel te selecteren, gevolgd door Kalibreren. Plaats de carrousel met de 13 kleine gaatjes naar boven in de AFC-toren en pas de carrousel aan door op de min- of plusknoppen te drukken, zodat het vloeistofmondstuk zich direct boven de spoelpositie bevindt. Verwijder de doppen uit de gebalanceerde SEC-kolom en schuif de SEC-kolom in de juiste kolombevestiging.Installeer het zorgvuldig op de AFC-toren. Zorg ervoor dat het radiofrequentie-identificatielabel op de kolom naar de AFC is gericht en controleer of de verbinding tussen de kolom en de klep veilig is. Plaats de afvaluitlaatbuis in een opvangbak.Selecteer in het installatiescherm Collectieschema en stel het aantal in op 13 en de grootte op 0,5 milliliter door op de min- of plusknoppen te drukken. Laat de instelling voor het buffervolume staan als de standaardinstelling van 2,7 milliliter en sluit vervolgens het venster met het verzamelschema. Selecteer Verzameling starten in het startscherm en bevestig de collectieparameters door Ja te selecteren.Laad 13 gelabelde microcentrifugebuizen van 1,7 milliliter met de deksels open en gericht naar het carrouselcentrum. Ga verder met de AFC door OK te selecteren, monteer vervolgens het kolomreservoir en ga opnieuw verder door op OK te drukken. Selecteer de optie om de kolom leeg te maken door op Ja te drukken en voeg één kolomvolume van 0,2 micrometer gefilterd PBS toe om eventuele opslagbuffers te verwijderen. Zodra de spoeling is voltooid, gaat u verder met de AFC door op OK te drukken. Plaats het carrouseldeksel over de AFC en druk op OK. Gebruik een pipet om overtollige buffer uit de kolom te verwijderen.Breng drie milligram 100R monster naar 500 microliter met 1X PBS en voeg het monster toe aan de bovenkant van de kolom. Bereid één kolomvolume van 0,2 micrometer gefilterd PBS voor. Zet de AFC vooruit en laat het monster in de fret lopen.Zodra het monster volledig in de kolom is gekomen, voegt u de eerder voorbereide PBS toe aan het reservoir. Controleer de uitvoering van de AFC terwijl deze eerst het leegtevolume en vervolgens de opgegeven fracties verzamelt. Nadat de run is voltooid en de carrousel terugkeert naar de afvalpositie, verwijdert u de afdekking en de fractiebuizen uit de carrousel.Voor 0,5 millimeter fracties verzameld uit drie milligram 100R Mtb CFP, meet de eiwitconcentratie met behulp van de micro-BCA met een 1: 3 verdunning voor fracties genummerd 1 tot 7 van elk monster in drievoud. Gebruik voor 0,5 milliliter fracties genummerd 5 tot 13 de BMA zonder verdunning voor elk monster in drievoud. Voeg sds-monsterbuffer toe aan 10 microliter van elke fractie en vijf microgram CFP en 100R.Kook vijf minuten op 100 graden Celsius. Laad vervolgens een 4 tot 12% Bis-Tris-gel met een moleculaire gewichtsladder voor polyacrylamidegelelektroforese of PAGE. Voer een SDS-PAGE-gel uit met behulp van 1X MES-loopbuffer en een 200-volt stroom gedurende 35 minuten.Verwijder de gel van de cassette en breng gedurende minimaal een uur een stroom van 50 volt aan om de opgeloste eiwitten over te brengen naar een nitrocellulosemembraan van 0,2 micrometer. Voer ten slotte de Western blot-analyse uit om de eiwitten te evalueren. Een SDS-PAGE gel gekleurd met zilver toont het algemene eiwitprofiel voor elke fractie.Uit de gegevens blijkt dat de latere fracties meer eiwit bevatten en lijken op het 100R-materiaal. Western blots die LAM, LpqH en GroES detecteren, tonen aan dat eiwitmarker verandert over de fracties. Transmissie-elektronenmicroscopie, of TEM-beelden, van SEC-fracties 1 tot 3 getrokken voorafgaand aan fixatie worden hier weergegeven.De ultracentrifugatie van de dichtheidsgradiënt van Mtb EV's was negatief gekleurd voor TEM-beeldvorming. De nanodeeltjes tracking analyse, of NTA-verdeling, van SEC-fracties 1 tot 3 wordt hier weergegeven. De dichtheidsgradiënt ultracentrifugatie MTB EV's werden geanalyseerd met nanodeeltjes tracking analyse, en de grootteverdeling wordt hier weergegeven.Dit protocol is geoptimaliseerd voor EV-zuivering vanaf drie milligram 100R. Als de eiwitbelasting varieert, kan de aanbevolen verdunnings- en BCA-strategie voor resulterende fracties moeten worden aangepast. Mtb EV's die met deze techniek zijn bereid, kunnen worden gebruikt voor downstream compositionele en op omics gebaseerde studies.Ze zijn ook geschikt voor in vitro en in vivo experimenten. Deze techniek biedt een gemakkelijke en efficiënte manier om hoogwaardige Mtb EV-preparaten te produceren, waardoor de weg wordt vrijgemaakt voor een verhoogde reproduceerbaarheid in toekomstige Mtb EV-experimenten.
