筋肉におけるエレクトロポレーション媒介遺伝子導入は、筋肉生理学の変化を調査するための簡単で効率的な技術である。エレクトロポレーションは筋肉の収縮性を損なわないことを発見しました。インビボでの筋肉への遺伝子構築物のエレクトロポレーションは、ウイルスベクターへの遺伝子構築物のより広範なパッケージングを必要としない簡単で効率的な手順である。EDLの位置とサイズが小さいため、EDLを視覚化、分離、および注入することは困難です。マウスの死体で練習し、インドインクなどの生物学的インクを注入することは、十分な注射の証拠を提供するのに役立つかもしれない。手順を実演するのは、私の研究室のポスドクであるブライアン・ヘインです。マウスを注射する前に、つま先ピンチ反射のない鉗子を使用して麻酔の手術面を確認し、残りの手順では、マウスを摂氏37度の循環水板の上に置いたノーズコーンに移します。マウスを仰臥位に置いたら、小さなバリカンを使用して両後肢から毛を取り除き、70%エタノールとベタジンを交互に注射領域を消毒します。注射のために、下肢の側方側の皮膚を通して見える前脛骨またはTA腱を見つけ、TA筋肉の上端に印を付ける。次に、50マイクロリットルのマイクロシリンジに取り付けられた30ゲージの針を、針が筋肉の上端に達するまで、筋腱接合部より1〜2ミリメートル優れた浅い5度の角度で筋肉に挿入する。注射経路に沿って針をゆっくりと引っ込めながらプランジャーを押し下げ、50マイクロリットルのプラスミド溶液を筋肉に送達する。筋肉は腫れるはずです。タイマーを1分間セットし、TA筋で脚の太さを測定し、ノギス電極を測定太さに設定します。次に、エレクトロポレーター電圧を1ミリメートルあたり12.5ボルトに設定します。1分後、キャリパー電極を過度にきつくすることなく下肢の周りにしっかりと置き、20ミリ秒の持続時間と200ミリ秒間隔で5つの方形波パルスを送達します。筋肉は脈拍ごとに痙攣するはずです。コントロールプラスミドベクターを用いて他の四肢でこの手順を繰り返す。完了したら、マウスをノーズコーンから取り外し、37°Cに設定した加熱パッドでマウスを回復させます。回復したら、マウスをケージに戻します。マウスを仰臥位に置いた状態で、視覚的および穏やかな動悸を通して脛骨前紋を見つける。メスを使用して、膝より5ミリメートル下からTA筋腱接合部より2ミリメートル優れた脛骨前頂部の側方側の皮膚を通して浅い切開を行う。小さなはさみの助けを借りて、筋膜を鈍く解剖し、TA筋肉を露出させ、筋肉を引っ張って伸筋桁ロンガスまたはEDLを明らかにすることによってTA筋肉を脛骨から穏やかに分離する。小さな湾曲した鉗子を使用して、手順中にTAをEDLから離してください。針が筋肉の上端に達するまで30ゲージ針を縦方向にEDLに挿入し、前述のように10マイクロリットルのプラスミド溶液を注入する。EDL筋肉は腫れます。EDLで脚の厚さを測定し、ノギス電極を測定された厚さに設定して、前述のように5つの方形波パルスを送達します。完了したら、使い捨ての4-0非吸収性ナイロン縫合糸を使用して切開部を閉じます。その後、マウスをノーズコーンから取り外し、摂氏37度に設定した加熱パッドで回復させます。手術直後および手術後12〜24時間に適切な皮下鎮痛剤を投与する。回収したら、マウスをホームケージに戻します。TAおよびEDL筋へのpcDNA3-EGFPプラスミドの注入およびエレクトロポレーション後、筋線維におけるDNA取り込みを示すGFP発現が観察された。より低い倍率で画像化した場合、EDL筋トランスフェクション効率は、緑色の繊維対黒色の繊維の出現によって視覚化することができた。注入およびエレクトロポレーションされたEDLは、非注入またはエレクトロポレーションされた対照の筋肉と比較して、100ヘルツで同様の破傷風応答を有した。注入およびエレクトロポレーションEDLでは、破傷風筋力、特定の破傷風筋力、緊張をピークとする時間、および半分の弛緩時間は、手つかずの対照と比較して損なわれなかった。注入する筋肉を正確に特定し、プラスミド溶液を効率的に送達することが重要です。このプロトコルを使用して、多くの組織学的および生化学的測定を行うことができます。標的タンパク質の細胞局在化、標的遺伝子ノックダウン、転写レポーター、および組織染色のアレイを実施することができる。この技術は、生理学的および病態生理学的状態における筋肉における分子シグナル伝達の膨大な配列の変化を同定するために使用されてきた。遺伝子導入後の筋収縮性の研究は、この技術を用いて実現可能である。
