データ収集と分析のためのソフトウェアは、IGOR Pro(完全にインタラクティブなソフトウェア環境)でカスタム作成されました。アナログ-デジタルコンバータ(A/D)システムは、コンピュータ制御下で指令電圧を発行し、カスタムアンプからデータを集録しました。アンプは、特定のユニークな機能を持っていました。これらには、4つの集録チャンネルのそれぞれに対応する帰還抵抗(切り替え可能)が含まれており、1 MOhmまたは10 MOhmのフィードバック電圧クランプ回路を選択して電極の変動性を統合することができます。また、1つのヘッドと4つの集録チャンネルすべてに対して相互のグランド/リファレンス回路を備えたステージユニットも構築され、デバイスを1つの物理モジュールに胸部の近くに配置しました。1 MOhmの帰還抵抗設定を使用して、報告されたすべてのデータを収集しました。

インビボでのペプチド伝達物質レベルの時間分解測定のための方法が開発されたのは今回が初めてです。ほぼリアルタイムの解析により、単一の被験者および離散的な場所のペプチド伝達物質レベルを測定できます。まず、25センチメートルの長さのパーフルオロアルコキシ被覆白金線を切断する。次いで、メスを用いて、一端から約5ミリメートルのパーフルオロアルコキシ被膜を剥ぎ取り、白金線に切り込まないように注意する。次に、白金線の剥がれた端を金メッキの1ミリメートルのオスコネクタピンに挿入します。次に、ニードルノーズペンチを用いて、白金線の剥がれた端の周りのコネクタピンの歯を圧着し、白金線を金メッキコネクタピンにはんだ付けします。次に、pH6の10ミリモルPBS50ミリリットルにドーパミン塩酸塩50ミリグラムを加えてドーパミン溶液を調製し、攪拌混合する。ドーパミンを完全に溶解させた後、白金線の先端を、作りたてのドーパミン添加PBSの入った容器に入れる。塩化銀ディスク電極をヘッドステージのグランドチャネルに接続します。次いで、ドーパミン添加PBS及び白金線の入った容器に塩化銀ディスクを入れる。先に進む前に、ワイヤシャントをヘッドステージのリファレンスチャンネルに接続します。インタラクティブなデータ収集ソフトウェアを開き、開始電位をマイナス0.6ボルト、終了電位を0.65ボルト、スキャンレートを毎秒0.04ボルト、蒸着期間を420秒に設定して、鋸歯状電解析出コマンド電位プロトコルを設定します。ポリドーパミン析出を完了した後、塩化銀粉砕ペレットおよび白金線の先端を容器から取り外す。ワイヤチップをpH 7.4のPBSを含むマイクロチューブに2〜5分間置き、ワイヤチップがマイクロチューブの側面または底部に接触していないことを確認します。次いで、適当な大きさの容器内で目的の抗体とpH7.4のPBSとを1:20の比率で結合させて抗体溶液を調製する。次に、白金電極のポリドーパミン析出先端部を抗体溶液に室温で2時間浸し、白金線先端部が溶液中に浮遊し、マイクロチューブの内面に載っていないことを確認する。容量性免疫プローブの機能的先端を流しチャンバ内に置き、電極の先端をいかなる方法でも乱さないように注意し、そうすることでプローブの感覚先端を損傷する可能性がある。最初の実験試験の前に、TBS標準ランを実行して容量性免疫プローブをコンディショニングし、電圧プロトコルについては、正のステップ電位を110ミリボルト、負のステップ電位をマイナス5ミリボルト、ステップ持続時間を20ミリ秒、および取得期間を600秒に設定します。次に、同じTBSを使用して目的のペプチド溶液を作成し、過fusateの組成を維持する。超fusateをTBSとペプチド補足TBSの間で切り替えることができるマニホールドシステムを設定します。TBS標準パラメータを使用し、集録時間を360秒に設定してペプチドセンシングデータ収集プロトコルを設定します。ポリドーパミン沈着の前に、白金線電極の露出した先端を22ゲージの皮下注射針に通し、針の先端から約2ミリメートル離れたところに残す。鉗子を使用し、白金線電極の先端を優しく曲げて、皮下注射針の端から垂れ下がるとげを作ります。有刺鉄線の先端から針を静かに引き抜き、針が流体に接触することなく容器に入れるのに十分なワイヤーを残し、前述のようにドーパミン沈着を進行させる。次に、有刺鉄線プローブチップをPBSですすぎ、抗体溶液に入れます。次いで、PBSから官能化チップを静かに取り外す。皮下注射針を有刺鉄線容量性免疫プローブに移し、関心領域にそっと埋め込み、金色のコネクタピンをヘッドステージに差し込みます。移植したら、皮下注射針を抜き取り、電極を所定の位置に残します。コマンド電位の正のステップは、プローブ電圧をクランプするために必要な注入電流を測定し、容量性電流の測定を可能にします。負のコマンド電位ステップは、静電反発を介して抗体から結合したペプチドをクリアする。ステップ関数コマンド波形には、ペプチドの時間分解反復検出および定量が描かれた。下部のトレースは、TBSにおけるプローブのスーパーフュージョン下でのコマンド電位とその結果生じる電流を表していました。平衡化された容量性免疫プローブは、より小さな初期崩壊および6分間にわたって安定したベースラインを示した。ニューロペプチドYの流れチャンバへの流れは、ベースライン上の容量性電流を増加させた。容量性免疫プローブ電流をニューロペプチドY抗体官能化プローブで測定し、低ピコモールニューロペプチドYに感受性の濃度依存的シグナルを示した較正のために、試験した全ての濃度について標準曲線を測定した。両側星状神経節の刺激は、ネガティブコントロール容量性電流と共プロットしたときに上昇したニューロペプチドYを示した。星状刺激下で測定されたノルエピネフリン放出は、神経ペプチドYとの同期放出を示し、誘発された交感神経応答と一致する。これらの技術の開発は、心臓活動の主要なモジュレーターの術中読み出しを提供し、したがって、潜在的な迅速な治療的または介入的ガイダンスを提供する可能性がある。ここで提示される技術は、心血管展開に特異的であるが、技術自体は、尿、骨格筋、腎臓、脂肪組織などの他の生理学的設定に適している。
