著者らは以前、ほぼ完全に市販の部品11で構成された低コストの灌流バイオリアクターシステムの設計について報告した。このシステムのベースバージョンでは、ウェルプレート内の複数の培養物をCO2 インキュベーターに保持し、シリンジポンプからの培地を連続的に灌流することができ、培養物からの流出培地ストリームは、カスタム修正を施したフラクションコレクターを使用して、時間の経過とともに自動的にサンプルに分画されます。したがって、このシステムは、培養培地上清の自動サンプリングおよび経時的な培養物への連続溶質入力を可能にする。このシステムはマクロ流体でモジュール式で、新しい実験計画のニーズを満たすように簡単に変更できます。

流れ内の分散と拡散により、溶質信号は流れシステムを通過するときに歪んだり「塗りつぶされたり」します。この歪みは、滞留時間分布(RTD)13を使用することで定量化できます。この記事では、灌流システムのコンポーネントに対してトレーサー実験を実行する方法(図2)を説明し、測定データからRTDを生成するためのMATLABスクリプトを提供します。この分析の詳細な説明は、著者の以前の論文11に記載されています。追加のMATLABスクリプトは、適切な機能をRTDに適合させ、物理パラメータを抽出し、RTDを使用して信号畳み込みを実行して、ユーザによって入力された溶質信号が灌流システム14を介してどのように伝播および歪むかを予測する。

multi-stream perfusion bioreactor integrated with outlet fractionation for dynamic cell culture
