私たちのプロトコルは、脳機能のより良い理解に不可欠な回路からコンポーネントスケールまでのニューロン構造の調査を容易にします。当社のクリアリング技術であるScaleSFは、強力なクリアリング能力と、大規模および小規模の両方の構造を同時に可視化するために必要な高レベルの組織保存性能を発揮します。私たちのプロトコルは、神経細胞が精巧なプロセス、途方もないリンクを示し、特殊な微細な細胞内構造を配置する脳において特に効果的です。実験を開始する前に、この表に示すように Sca/eS 溶液を調製し、サンプルをホイルで覆います。6ウェル細胞培養プレートの2つの別々のウェルにそれぞれ8ミリリットルのScaleS0およびScaleS4溶液を加え、インキュベーター内でプレートを摂氏37度に予備温することによって手順を開始する。脳スライスをヘラで予め加温したScaleS0溶液に移し、90RPMの振とうインキュベーター内で37°Cで2時間スライスをインキュベートする。次に、透過処理した脳スライスをPBSマイナス8ミリリットルで6ウェル細胞培養プレートに移し、40〜60RPMのオービタルシェーカーに保持して15分間洗浄する。この手順を 2 回繰り返します。次に、脳スライスを8ミリリットルの予め加温したScaleS4溶液に移し、摂氏37度で90RPMで8〜12時間インキュベートする。チャンバーの準備のために、チャンバーフレームと顕微鏡ステージアダプターを3Dプリントします。感圧接着剤を使用してチャンバーフレームをカバースリップに取り付けます。ScaleS4D25(0)溶液中に1.5%アガロースを調製する。溶液を混合し、アガロースが完全に溶解するまで電子レンジで送る。その後、溶液を摂氏37度まで冷却します。クリアした脳スライスをヘラでイメージングチャンバの底部カバースリップに取り付けます。清浄な組織を用いて透明化したスライスから余分な溶液を除去する。ScaleS4ゲルをマイクロピペットを用いて脳スライスに加え、イメージングチャンバを充填する。鉗子で別のカバースリップを上に置き、その後にティッシュペーパーとスライドガラスを置きます。撮像チャンバを摂氏4度の冷蔵庫に移す。スライドガラスに金属製のおもりを置き、30分間放置します。インキュベーション後、イメージングチャンバから材料を取り出し、余分なゲルを拭き取ります。イメージングチャンバを60ミリメートルのガラスシャーレに置き、感圧接着剤で複数のポイントでイメージングチャンバのリムをディッシュに取り付けます。ScaleS4溶液を皿に注ぎ、摂氏20〜25度で40〜60RPMで1時間静かに振る。新鮮な溶液で代用し、ピペットチップを使用して表面を優しく掻き取ってゲル表面の気泡を除去します。イメージングチャンバーを顕微鏡ステージに取り付けます。作動距離2.5mm、倍率16倍、開口数0.60の多液浸対物レンズを搭載した共焦点レーザー走査顕微鏡を作製する。ワークステーション、顕微鏡、スキャナー、レーザー、水銀灯など、関連するすべてのイメージング機器の電源を入れ、イメージングソフトウェアを起動します。マルチ液浸対物レンズの補正カラーを1.47に設定します。対物レンズを溶液に浸し、ゆっくりとスライスに近づけます。対物レンズの先端に付着した気泡をすべて取り除きます。エピ蛍光を使用して、クリアされた組織の関心領域を見つけます。画像取得パラメータを設定するには、まず、画像のサイズがビットとともに大きくなるにつれて、画像取得のビット深度を決定します。次に、発光スペクトルに応じて検出波長を設定します。XY解像度、スキャン速度、ピンホールサイズを設定します。また、適切な画像が得られるまで、レーザー出力、検出器アンプのゲイン、およびオフセットを調整します。ROIのサイズに基づいてタイリングサイズを決定し、ROIの全長と幅がキャプチャされるようにします。すべての平面で組織内を移動し、スタックの開始点と終点を設定します。Z ステップ サイズを、目的の Z 解像度に応じて設定します。画像を収集し、画像解析ソフトウェアを使用して処理します。このプロトコルを用いて、厚さ1ミリメートルのマウス脳スライスの光学的クリアリングが達成された。PV-FGLマウスの大脳皮質におけるEGFP発現は、組織の蛍光および構造的完全性が保存されていることを示した。屈折率の不一致誘発収差は、EGFP陽性ニューロンの画像輝度および解像度の顕著な損失を引き起こした。これらのニューロンは、このScaleS4溶液と一致するように補正カラーを調整したときに、共焦点レーザー走査顕微鏡で明確に視覚化された。マウス原発性体性感覚皮質におけるEGFP標識ニューロンの3D再構成は、厚さ1ミリメートルの脳スライスで行った。樹状突起棘で装飾された個々の樹状樹木は、より高い倍率で見られた。軸索終末樹木化および軸索ブートンもスライスにおいて観察された。イメージング流体とオブジェクトレンズ間の反射インデックスマッチングは、クリアされた組織における3Dイメージングにとって重要です。我々の技術は、回路から構成要素スケールへのニューロン構造の統合を容易にし、ニューロン機構、脳内の処理に関する情報への洞察を提供する。
