1. 生物样本的先决条件

</ol>2. 生物样本库软件
<ol><li>使用实验室信息管理系统 （LIMS） 软件跟踪所有生物样品。确保良好的操作系统的可用性，该系统可在患者注册时自动获取个人和临床信息，并且可以与病历、行政案例、RPA 和患者病理数据集成，如图 2 所示。</li>

3. 样品采集
<ol><li>日常手术计划准备<ol><li>确定患者是否已签署 RPA 以及他们是否符合条件。</li>
		<li>检查以下条件：<ol><li>验证患者是否符合纳入标准：在适当的感染风险“RI”字段中报告任何 HIV、HBV、HCV 和 COVID-19 阳性病例，以避免收集生物标本。</li>
			<li>通过正确填写每个患者档案中的 研究/ 项目字段，验证患者是否参加了临床试验或特定批准的研究项目。</li>

4. 血液样本采集
<ol><li>将患者的血液收集在含有抗凝剂Na2 EDTA（7.2mg，喷雾干燥）的6 mL真空容器中。注册标有医疗记录编号和剧集代码的真空容器，并以两种不同的方式处理它们：新鲜或冷冻。</li>
	<li>对于新鲜样本，请在生物样本库软件中登记血液样本。用生物样本库ID代码标记真空容器，并将其交付给授权用户。</li>
	<li>对于储存在生物库中的冷冻样品，准备两个冷冻库 2D 编码管，每个管 900 μL 血液（图 4）。在生物样本库软件中注册等分试样，将它们放在特定的条形码板上，并将其储存在冰箱（-80°C）中以确保恒温。</li>

5. 血清样本采集
<ol><li>将患者的血液收集在含有喷雾涂层二氧化硅的 6 mL 真空容器“塑料血清管”中以诱导凝血。将真空容器在室温（RT）下放置3小时以诱导凝血，然后在20°C下以828× g 离心10分钟。</li>
	<li>将血清 （450 μL） 储存在 0.5 mL Cryobank 2D 编码管中，每个样品总共 3-4 个等分试样。如果最后一个等分试样的血清体积小于 450 μL，请在注册期间将其指定为“剩余”，以跟踪冷冻血清的正确量。</li>
	<li>在生物样本库软件中注册等分试样，将它们放在特定的条形码板上，并将其储存在冰箱（-80°C）中以确保恒温。 注意：等分试样的数量取决于采集的全血的初始量和获得的血清量。</li>
</ol>6.外周血单核细胞分离
<ol><li>打开层流罩，用70%乙醇清洁。准备装有生物废物的袋子、无菌 1x 磷酸盐缓冲盐水 （PBS） 和一个空的无菌 50 mL 锥形管。</li>
	<li>将血液（来自EDTA收集管）倒入空的无菌50 mL锥形管中，并使用无菌的1x PBS（例如，15 mL血液+ 15 mL 1x PBS）1：1稀释。使用PBS冲洗血管。</li>
	<li>在20°C下以400× g 离心管30分钟，并在生物危害罩下处理管。使用巴斯德移液管回收含有PBMC的中间白色层，并将其置于新的无菌50 mL锥形管中。加入多达45mL的PBS洗涤PBMC，混合并在4°C下以400× g 离心10分钟。 回收沉淀，将其重悬于PBS中，并使用一次性伯克室计数细胞。 注意：PBS的体积取决于颗粒。从 15 mL 血液中，确保重悬体积为 2-3 mL，然后稀释 1：10 进行计数。使用公式 （1）： 重悬体积 10，000 × × × mL 稀释因子 3 平方的平均值 = 总细胞数 （1）</li>
	<li>用PBS再次洗涤PBMC，混合并在4°C下以400× g 离心10分钟。 在冷冻培养基（胎牛血清 （FBS） + 10% 二甲基亚砜 [DMSO]）中以 2-3 x 106 个细胞 /mL 稀释 PBMC。通过将 1 mL 重悬细胞转移到每个冷冻管中来制备 Cryobank 2D 编码管，将它们放入特定的冷冻盒中，并尽快将其储存在 −80 °C。 注意：冷冻培养基由具有无菌10%DMSO的FBS组成，并在-20°C下等分储存长达6个月。一旦在4°C缓慢解冻，必须在2周内使用。</li>
</ol>7. 血浆样本采集
<ol><li>将患者的血液收集在含有抗凝剂Na2 EDTA（7.2mg，喷雾干燥）的6 mL真空容器中。将装有全血的真空容器在4°C下以2，000× g 离心10分钟以分离血浆。</li>
	<li>使用3mL巴斯德移液管去除血浆的上层，转移到无菌的15mL锥形无菌管中，并在4°C下以16，000× g 离心10分钟以消除污染的血细胞。将血浆转移到 1 mL 冷冻银行二维码管中。</li>
	<li>在生物样本库软件中注册等分试样，将它们放在特定的条形码板上，并将其储存在冰箱（-80°C）中以确保恒温。</li>
</ol>8. 粪便和口腔拭子样本采集
<ol><li>在含有以下特定溶液的 15 mL 管中收集粪便和口腔拭子：50 mM Tris-HCl、10 mM NaCl 和 10 mM EDTA，pH 7.5。</li>
	<li>在生物样本库软件中注册试管。用生物样本库代码标记试管。</li>
	<li>将粪便和口腔拭子储存在生物库冰箱中的4°C。</li>
</ol>9. 组织处理
<ol><li>让病理学家检查组织样本，以确定诊断程序不需要的材料是否足以用于研究目的。 注意：当组织体积小于1.5 mm3时，将其存储在OCT中或作为与特定研究请求相关的新鲜样品（A）提供。</li>
	<li>只要有可能，甚至收集病理组织的非病理对应物（NP）（P）。将样品放入标记为P和NP的无菌细胞培养培养皿中（图5）。将组织保持在4°C的冰上，如果有足够的材料，则将其分成三部分（A，B和C）。<ol><li>新鲜组织样品（A）：将P和NP组织的新鲜等分试样放入具有每个特定方案中定义的适当培养基的管中，并将其发送到外部研究单位（例如，研究实验室）。</li>
		<li>OCT组织样品（B）：将P和NP组织的新鲜等分试样放入冷冻模具中，用OCT树脂（10.24%聚乙烯醇，4.26%聚乙二醇和85.5%非反应性成分）填充，并立即将它们置于-80°C的速冻装置中。 注意：在-80°C下，OCT包埋的组织需要60-150秒才能冷冻。</li>
		<li>组织 样本（C）：将剩余的组织样本放入闪速冷冻装置的冷冻银行二维编码管中。</li>
	</ol></li>
	<li>将板储存在-80°C。 注意：对于每个冷冻的OCT等分试样，在分发及其使用之前进行质量检查测试以进行组织学评估，如第11节所述。</li>

10. 组织冷冻
<ol><li>使用快速冷冻设备在异戊烷蒸气中快速冷冻组织。将组织样品置于-120°C的异戊烷中3分钟，并将其储存在-80°C的冷冻保存室中。 注意：使用快速冷冻冷冻两种方法冷冻肿瘤和健康组织样本，以保持核酸的完整性。</li>
</ol>11. 组织质量控制

12. 为研究目的索取和检索标本

biobank for translational medicine: standard operating procedures for optimal sample management
