골격근은 성인 체중의 ~40%를 차지하는 신체에서 가장 큰 조직으로 자세 유지, 움직임 생성, 기초 에너지 대사 조절 및 체온1을 담당합니다. 골격근은 매우 역동적인 조직이며 기계적 스트레스, 대사 변화 및 일상적인 환경 요인과 같은 다양한 자극에 적응하는 놀라운 능력을 가지고 있습니다. 또한 골격근은 급성 손상에 반응하여 재생되어 형태와 기능을 완전히 회복시킵니다2. 골격근 가소성은 주로 근섬유 원형질막과 기저층 2,3 사이에 위치한 위성 세포라고도 하는 상주 근육 줄기 세포(MuSC) 집단에 의존합니다. 정상적인 조건에서 MuSC는 정지 상태로 근육 틈새에 상주하며 세포 회전율을 보상하고 줄기 세포 풀을 보충하기 위해 몇 개의 분할 만 있습니다4. 부상에 대한 반응으로 MuSC는 세포주기에 들어가 증식하며 새로운 근육 섬유의 형성에 기여하거나자가 재생 과정에서 틈새로 돌아갑니다 2,3. MuSC 외에도 골격근의 항상성 유지 및 재생은 섬유-지방형성 전구세포(FAP)5,6,7라고 하는 근육 상주 세포 집단의 지원에 의존합니다. FAP는 근육 결합 조직에 내재된 중간엽 기질 세포이며 섬유원성, 지방원성, 골인성 또는 연골성 계통 5,8,9,10을 따라 분화할 수 있습니다. FAP는 근육 줄기 세포 틈새에서 세포 외 기질 단백질의 공급원이기 때문에 MuSC에 대한 구조적 지원을 제공합니다. 다분화능과 재생 능력으로 인해 FAP와 MuSC는 골격근 장애 치료를 위한 재생 의학의 잠재적 표적으로 확인되었습니다. 따라서, 이들의 기능 및 치료 잠재력을 조사하기 위해서는 FAP 및 MuSC의 분리 및 배양을 위한 효율적이고 재현 가능한 프로토콜을 수립하는 것이 중요하다.
형광 활성화 세포 분류(FACS)는 크기 및 입도와 같은 형태학적 특성을 기반으로 다양한 세포 집단을 식별할 수 있으며 세포 표면 마커에 대한 항체의 사용을 기반으로 세포 특이적 분리를 허용합니다. 성체 마우스에서 MuSC는 혈관 세포 접착 분자 1 (VCAM-1, CD106이라고도 함) 14,15 및 α7- 인테그린 15를 발현하는 반면, FAP는 혈소판 유래 성장 인자 수용체 α (PDGFRα) 및 줄기 세포 항원 1 (Sca1 또는 Ly6A / E) 5,6,9,12,16,17 . 여기에 설명된 프로토콜에서 MuSC는 CD31-/CD45-/Sca1-/VCAM-1+/α7-인테그린+로 식별된 반면, FAP는 CD31-/CD45-/Sca1+/VCAM-1-/α7-인테그린-으로 식별되었습니다. 대안적으로, PDGFRαEGFP 마우스를 사용하여 FAP를 CD31-/CD45-/PDGFRα+/VCAM-1-/α7-인테그린-이벤트18,19로 단리하였다. 또한 PDGFRα-GFP+ 세포의 형광 신호 간의 중첩을 표면 마커 Sca1로 식별된 세포와 비교했습니다. 우리의 분석은 모든 GFP-발현 세포가 Sca1에 대해서도 양성임을 보여주었으며, 이는 FAP의 식별 및 분리를 위해 두 접근법 중 하나를 사용할 수 있음을 나타냅니다. 마지막으로, 특이적 마커 항체로 염색하여 각 세포군의 순도를 확인하였다.
