1. 组织解剖和清洁
注意：视频协议中使用10至12周龄的C57BL / 6J小鼠。有关组织解剖和清洁动脉的示意图和预期结果，请参阅 图1 ，有关耗材和制造商信息的列表，请参阅 材料表 。

		<li>使用#55镊子小心地从动脉中取出实质脂肪。一次去除小块脂肪，并在立体镜下仔细观察动脉/静脉对，以识别并避免损坏感兴趣的脉管系统。 注意： 相应地调整放大倍数和光源以清洁动脉。增加放大倍数和光强度，以便在去除脂肪时更好地观察动脉。动脉的精细清洁应在4x-5x放大镜下进行。区分结缔组织（胶原蛋白）、静脉和动脉很重要。结缔组织和胶原纤维具有无分支的弦状外观，有条纹，颜色发白。与胶原纤维和结缔组织不同，动脉和静脉是透明的，并且具有多个分支和明确的管腔。小动脉和静脉在无实质组织时可能看起来相似。管腔中血液的存在可以进一步帮助人们从静脉中识别动脉。与同等大小的动脉相比，静脉具有更薄的血管壁和更大的管腔，并且含有更多的血液。</li>
		<li>重复步骤 1.5.3。直到动脉完全没有实质脂肪组织。</li>
		<li>使用#5镊子将清洁的动脉转移到带有新鲜HEPES缓冲液的新管中，并保持在冰上直到准备好消化。</li>
	</ol></li>
</ol>2.消化分离动脉以分离内皮细胞
注意：请参阅 图2 ，了解组织消化和内皮细胞分离的示意图，以及材料 表 ，了解该方案所需的耗材的完整列表。
<ol><li>向每个 2 mL 微量离心管中分别加入 1 mg 分散酶和弹性蛋白酶。向每个试管中加入 2 mL 解离溶液。涡旋并在37°C孵育直至完全溶解。使用#5镊子将清洁的动脉（步骤1.5.5.）转移到相应的样品管中。每种酶的最终浓度为0.5mg/mL。</li>
	<li>在37°C下孵育1小时，每10-15分钟通过倒置轻轻搅拌，使得动脉悬浮在溶液中以保持酶和动脉之间的一致和均匀接触。</li>
	<li>称取每个样品1mg的I型胶原酶，并将其添加到新管中。</li>
	<li>让动脉沉淀在管子底部。在不干扰动脉的情况下从顶部取出 500 μL 缓冲液，并将其转移到含有胶原酶的指定管中。涡旋并将溶液返回到原始样品管中的相应样品中。I型胶原酶的最终浓度为0.5毫克/毫升。</li>
	<li>在37°C孵育15分钟。确保在样品管短暂摇晃后动脉分离成碎片。如果动脉在扰动后没有分离成碎片，则以 5 分钟为增量孵育，直到可见动脉破裂。</li>
	<li>使用玻璃移液管，用力研磨消化的组织10x-15x以机械方式解离细胞。</li>
	<li>将溶液和消化的组织通过70μm细胞过滤器或流式细胞术管过滤器以获得单细胞悬液。</li>
</ol>3. 流式细胞术前内皮细胞染色
注意：除非有指示，否则内皮细胞的染色和处理在冰上进行以提高细胞活力。将 5 mL 聚苯乙烯圆底管在室温 （RT） 下以 1，163 x g 离心 3 分钟。

isolation and identification of vascular endothelial cells from distinct adipose depots for downstream applications
