私たちのプロトコルは、破骨細胞再吸収アッセイのための簡単で信頼性の高い方法を説明しています。リン酸カルシウムコーティングされた細胞培養プレートの使用は、ラボで利用可能な材料を使用して容易に調製および視覚化することができる。不透明な骨や象牙質のスライスとは対照的に、リン酸カルシウムコーティングは、再吸収ピットと細胞を同時に視覚化することを可能にします。このプロトコルでは、破骨細胞の分化にPBMCを使用します。しかし、骨髄由来単球またはRaw 264.7細胞のような他の細胞型にも適用することができる。まず平底96ウェル細胞培養プレートと予め調製したリン酸カルシウム溶液300マイクロリットルのピペットを各ウェルに取り込む。その後、プレートを蓋で覆い、プレートを摂氏37度で3日間インキュベートする。3日後、予め石灰化した96ウェル細胞培養プレートから予備石灰化溶液を吸引し、各ウェルにリン酸カルシウム溶液300マイクロリットルを加える。プレートを蓋で覆い、摂氏37度で1日間インキュベートする。その後、プレートを反転させて溶液を注ぎ出し、プレートを脱イオン水で3回徹底的に洗浄します。ヘアドライヤーまたは圧縮二酸化炭素または窒素ガスを使用してプレートを直ちに乾燥させ、均一な表面を維持します。さて、クリーンベンチの上の塗装板に紫外線を1時間照射します。コーティングされたプレートをすぐに使用するか、パラフィルムでプレートを密封し、室温で保管してください。15ミリリットルの淡血を等量のPBSで希釈し、数回反転またはピペッティングして混合する。次いで、50ミリリットルの円錐形チューブに15ミリリットルの密度勾配溶液を採取し、チューブを傾け、15ミリリットルの密度勾配溶液上に30ミリリットルの希釈血液サンプルを慎重に重ねる。その後、チューブをスイングバケットローターで810倍Gで休憩なく20°Cで20分間遠心分離します。次いで、単核球層を界面に乱れずに残して上層を吸引する。単核球細胞層を新しい50ミリリットルの円錐管に慎重に移す。チューブをPBSで満たし、混合し、300倍Gで20°Cで10分間遠心分離する。PBMCsを1ミリリットル当たり20ナノグラムのマクロファージコロニー刺激因子を含む完全なα−MEMに再懸濁し、フラスコ内のPBMCsをセンチメートル平方あたり10〜5番目の細胞に2.5回播種する。リン酸カルシウムコーティングプレートを50マイクロリットルのウシ胎児血清とともに摂氏37度で1時間インキュベートする。次に、フラスコをPBSで2回洗浄して破骨細胞前駆体を死細胞または非接着細胞から剥離し、次いで75平方センチメートルフラスコ当たり4ミリリットルのトリプシンを加える。30分後、4ミリリットルの完全α-MEMを加えて消化を止め、細胞スクレーパーを用いて細胞を慎重に剥離する。細胞を50ミリリットルのチューブに移し、計数チャンバを用いて細胞の総数を計数した。チューブを350倍Gで7分間遠心分離して細胞をペレット化し、ペレットを蘇生させ、M-CSFおよびRANKLを含むアルファMEMを完成させて、1ミリリットルあたり10〜6個の細胞を1回得る。リン酸カルシウムコートプレートからウシ胎児血清を吸引し、1ウェルあたり200マイクロリットルの細胞懸濁液をピペットした。インキュベーション後、細胞をPBSで2回洗浄する。細胞を4%パラホルムアルデヒドで10分間固定し、PBSで再度洗浄する。染色のために、固定細胞に透過処理緩衝液を加え、それらを5分間インキュベートする。アクチンフィラメントを染色するには、細胞を100マイクロリットルのAlexa Fluor 546標識ファロイジン溶液と共にPBS中で30分間インキュベートし、次いで染色溶液を吸引し、100マイクロリットルのヘキストを用いて核を10分間染色する。リン酸カルシウムコーティングを100マイクロリットルの10マイクロモルカルセインおよびPBSで10分間染色する。PBSで3回洗浄した後、画像をキャプチャします。リン酸カルシウムコーティングのフォンコッサ染色のために、50マイクロリットルの5%硝酸銀および脱イオン水を各ウェルに加え、ウェルの底部のコーティングが茶色になるまでプレートを紫外線照射下で1時間インキュベートする。96ウェルプレートの底部にあるリン酸カルシウムコーティングは、均一でよく接着しているように見えた。リン酸カルシウムコーティングプレート上のM−CSFおよびRANKLでの9日間の培養後、破骨細胞前駆体は再吸収ピットおよび大きな破骨細胞を形成した。ピット内に位置する多核破骨細胞は、高いTRAPレベルを発現した。成熟破骨細胞は、3つ以上の核および別個のアクチン環の存在を示した。緑色のリン酸カルシウムコーティングは、黒い再吸収ピットの存在を示した。破骨細胞の数とサイズはROIマネージャーを使用して計算され、RANKLの非存在下で増殖した画像J.破骨細胞の前駆体の多角形選択ツールを使用して破骨細胞の数とサイズは再吸収ピットを形成できなかったが、M-CSFおよびRANKLは骨殻形成ピットの形成を促進した。画像J.を用いてピット面積を定量化し、塗布板を気流で直ちに乾燥させることで、均一なリン酸カルシウム被膜を作るのに役立つ。さもなければ、それは井戸の真ん中でより厚いコーティングを形成する傾向があります。
