교모세포종(GBM)은 성인에서 가장 흔하고 공격적인 원발성 뇌종양입니다. 최근 신경외과, 표적 약물 개발 및 방사선 요법의 발전에도 불구하고 GBM 환자의 5년 생존율은 5% 미만으로 30년 이상 크게 개선되지 않은 통계입니다1. 따라서보다 효과적인 치료 전략이 필요합니다.
새로운 치료법을 개발하기 위해 연구 프로토콜은 (1) 종양 이질성과 미세 환경을 정확하게 요약하는 번역 가능한 전임상 모델을 활용하고, (2) 현재 수술, 방사선 요법 및 화학 요법을 포함하는 GBM 환자에게 사용되는 표준 치료 요법을 반영하고, (3) 절제 된 코어와 잔여 코어의 차이를 설명해야한다는 것이 점점 더 분명 해지고 있습니다. 침윤성 종양 조직 2,3,4,5. 그러나 현재 이용 가능한 대부분의 전임상 뇌종양 모델은 외과적 절제를 시행하지 않거나 상대적으로 시간이 많이 소요되는 외과적 절제모델을 활용하여 상당한 양의 출혈을 초래하거나 표준화가 부족합니다. 또한, 설치류 뇌종양의 절제를 수행하는 것은 임상적으로 비교 가능한 수술 도구 또는 프로토콜이 부족하고 체계적인 조직 수집을 위한 확립된 플랫폼6의 부재로 인해 어려울 수 있습니다(표 1).
본 프로토콜은 다기능 비절제 최소 침습 절제 시스템(MIRS) 및 통합 조직 보존 시스템(TPS)을 사용하여 설치류 뇌종양 절제 및 조직 보존을 위한 표준화된 패러다임을 설명하는 것을 목표로 합니다(그림 1). 이 독특한 기술은 GBM 및 기타 유형의 뇌종양 모델에 대한 전임상 연구의 다양한 연구에 활용할 수있는 표준화 된 플랫폼을 제공 할 것으로 기대됩니다. 뇌종양에 대한 치료 또는 진단 양식을 조사하는 연구원은 이 프로토콜을 구현하여 연구에서 표준화된 절제를 달성할 수 있습니다.
