Osteoclasten zijn een van de belangrijkste celtypen die betrokken zijn bij het optreden en de ontwikkeling van botziekten, evenals een van de primaire objecten van onderzoek naar botziekten20. Monocyten/macrofagen kunnen differentiëren in osteoclasten. Er is gemeld dat monocyten-macrofaag afstammingscellen in het beenmerg kunnen differentiëren in bloedmonocyten en weefselmacrofagen 1,2. De bovenstaande cellen, die kunnen differentiëren in osteoclasten om botgroei en -ontwikkeling in evenwicht te brengen, worden vaak gebruikt als een celmodel om osteoclasten in vivo te induceren 3,4. Beenmerg is een speciaal weefsel dat verschillende soorten cellen bevat, waaronder maar niet alleen beperkt tot mesenchymale stamcellen in het beenmerg, beenmergmonocyten-macrofaagcellen (BMM's), hematopoëtische stamcellen, endotheelcellen en immuuncellen. In feite suggereerden verschillende eerdere studies dat aanhankelijke cellen die uit de beenmergholte van het lange bot werden gehaast, konden differentiëren in osteoblasten, osteoclasten, chondrocyten of adipocyten 5,6,7,8. Hoewel verschillende isolatie- en kweekmethoden zijn gebruikt om verschillende homogene celpopulaties te produceren, zijn er nog steeds grote uitdagingen bij het isoleren en kweken van BMM's uit een verscheidenheid aan verschillende celtypen.
Er zijn verschillende methoden ontwikkeld om mononucleaire macrofagen (BMSC's) in het beenmerg te extraheren. De meeste van deze methoden zijn echter complex 9,10,11. Dichtheidsgradiëntcentrifugatie vereist bijvoorbeeld een gespecialiseerde kit en de bewerking is tijdrovend en omslachtig. Deze methode is geschikt voor de isolatie van BMM's uit bloedmonsters met een hoog volume, maar niet uit beenmergmonsters 9,12,13. Bovendien is het extraheren van weefselmonsters met behulp van collagenasevertering een complexe en tijdrovende procedure; deze methode wordt niet aanbevolen voor de isolatie van BMM's uit beenmergmonsters14,15. Bovendien, hoewel stroomscheiding kan resulteren in sterk gezuiverde monocyten/ macrofaagpopulaties, vereist het zeer grote steekproefgroottes en hoge instrument- en apparatuurvereisten10,16. Bovendien is de microbead-verrijkingsmethode extreem duur en niet haalbaar in een algemeen laboratorium17.
Daarom werd in de huidige studie een handige, snelle en goedkope methode voorgesteld voor de isolatie van mononucleaire macrofagen uit het beenmerg. Beenmergcellen die voor verschillende tijdspunten werden gehecht, werden gebruikt om BMM's te isoleren met behulp van een secundaire hechtingsmethode. BMM's geëxtraheerd met de bovenstaande methode kunnen de vorming van osteoclasten in vitro induceren, waardoor een eenvoudig en handig celmodel wordt geboden voor de toekomstige studie van osteoporose in vitro.

Met een afname van de botmineraaldichtheid hebben botten meer kans om te breken, waardoor de kwaliteit van leven van een patiënt negatief wordt beïnvloed. De groei en ontwikkeling van botten worden voornamelijk gereguleerd door osteoblasten en osteoclasten. Het is algemeen aanvaard dat osteoclasten zijn afgeleid van beenmergmonocyten-macrofaagcellen (BMM's). BMM's en andere hematopoëtische stamcellen bevinden zich in de beenmergholte. Daarom is het isoleren van enkele stabiele BMM's van verschillende en heterogene celpopulaties een enorme uitdaging. Hier presenteren we een protocol voor de isolatie van BMM's van SD-ratten, de secundaire therapietrouwmethode genoemd. Adherente cellen gekweekt gedurende 24-48 uur in primaire cultuur werden verzameld. Flowcytometrische analyse toonde aan dat ongeveer 37,94% van de cellen CD11b/c+ (monocyte-macrofaag oppervlakteantigeen) waren. Tartraatresistente zuurfosfatase (TRAP) kleuring en western blot analyse toonden aan dat BMM's in vitro konden differentiëren in osteoclasten. De bovenstaande bevindingen suggereerden dat de secundaire adherentiecellen kunnen worden beschouwd als een geschikt cellulair model voor onderzoek naar osteoclastendifferentiatie.

De secundaire aanhangende celpopulatie was stabiel en uniform. In dit protocol worden beenmergmonocyten-macrofaagcellen geëxtraheerd door middel van secundaire therapietrouwmethode. Beenmergmonocyten-macrofaagcellen kunnen met succes differentiëren tot osteoclasten, wat een stabiel celmodel biedt voor de studie van osteoclasten in vitro. Deze methode is geschikt voor kleine beenmergmonsters en kan beenmergmonocyten-macrofaagcellen eenvoudig en snel uit het beenmerg extraheren zonder dat er hightech laboratoriumapparatuur nodig is.De toename van osteoclasten kan de potentiële pathogenese van osteoporose zijn. Beenmergmonocyten-macrofaagcellen hebben een bepaalde onderzoekswaarde als osteoclastenvoorlopercellen. Om te beginnen dompel je de geëuthanaseerde ratten gedurende 10 minuten onder in 75% alcohol voor desinfectie.Na het zorgvuldig verwijderen van alle ledematen van de rat met een schaar en tang, aspirateer met PBS met behulp van een pipet en spoel het bloed dat zich aan de ledematen hecht. Dan bij twee milliliter van de 10%FBS DMEM in een buis van vijf milliliter. Western blot-analyse toonde aan dat de expressieniveaus van de osteoclastische specifieke eiwitten TRAP en cathepsine K significant verhoogd waren in behandelde cellen in vergelijking met de controlegroep.Het belangrijkste in deze procedure is de tijd om secundaire aanhangende cellen te verzamelen in 24 tot 48 uur primaire cultuur, beenmergmonocyten-macrofaagcellen beginnen zich volledig te hechten. Beenmergmonocyten-macrofaagcellen geïsoleerd door deze methode kunnen in vitro worden geïnduceerd in osteoclasten, wat een stabiel celmodel oplevert voor de studie van osteoporose.
