黄龍峰(HLB)は、世界で最も問題のある柑橘類の病気の1つです1。HLBは、師部コロニー形成および潔癖性細菌カン ディダトゥ ス・リベリバクター属( Candidatus Liberibacter asiaticus(CLas)、 Ca. L. africanus、 Ca. L. americanus2を含む。中国と米国で最も一般的なHLB関連種はCLasであり、これはアジアの柑橘類のオオバコ(Diaphorina citri)または接ぎ木によって伝染します3。CLasに感染した後、柑橘類の木は死ぬまで成長の低下を示します2。CLasに感染した柑橘類の葉の一般的な症状は、しみのあるまだら、緑色の島(小さな円形の濃い緑色の点)、厚くて革のような葉の隆起したコルク状の静脈、および不均一な黄変芽です2。さらに、CLasに感染した果物は小さくて偏っているように見えます2。

Cas12a単独では、低レベルの核酸を検出するのに十分な感度は理論的にありません。したがって、その検出感度を向上させるために、CRISPR−Cas12a検出は、典型的には等温増幅ステップ14、15と組み合わされる。リコンビナーゼポリメラーゼ増幅(RPA)は、37°Cから42°Cの温度範囲で高感度かつ迅速な等温DNA増幅を可能にします19。
Cas12aのDNase活性とRPAおよび蛍光読み出しを組み合わせたDETECTR(DNAエンドヌクレアーゼ標的CRISPR トランス レポーター)と呼ばれる検出プラットフォームが最近考案され12 、より高感度で核酸を検出することが示されています20。さらに、陽性サンプルから放出された蛍光シグナルは、現場でハンドヘルド蛍光検出装置を介して観察することができる。
RPAでDNAを増幅し、CLas21に特異的な5コピーnrdB(リボヌクレオチド還元酵素β-サブユニット)遺伝子を標的とするcrRNAを設計し、Cas12aタンパク質のDNase活性を利用したことから、このCLas検出法をCLas-DETECTRと名付けました。既存のCLas検出方法と比較して、CLas-DETECTRは高速、正確、高感度、および展開可能です。

Field-Deployable Candidatus Liberibacter asiaticus Detection Using Recombinase Polymerase Amplification Combined with CRISPR-Cas12a

黄龍弓、HLBと略される。それは世界中で壊滅的な柑橘類の病気です。中国や米国などのほとんどの柑橘類生産国では、HLBは師部制限細菌によって引き起こされます カンディダトゥス・リベリバクター・アジアティカス (CLasと略される)。CLasの早期発見は、早期介入を促進し、病気の蔓延を防ぐ生産者にとって重要でした。そこで、ここでは、リコンビナーゼポリメラーゼ増幅およびCRISPR-Cas12aシステムと組み合わせて、迅速でポータブルなHLB診断のための新しい方法を開発しました。私たちはそれをCLas-DETECTRと呼んでいました。私たちのプラットフォームの感度はPCRよりもはるかに高いです。さらに、葉サンプルを使用した場合のqPCRでも同様の結果を示します。従来のCLas検出と比較して、私たちの方法は90分で終了でき、等温条件で動作します。結果は、現場でハンドヘルド蛍光検出装置を介して視覚化することもできます。CLas-DETECTRには、溶液調製、柑橘類の全DNA単離、等温DNA増幅、および可視化結果の4つのステップが含まれています。CLas-DETECTRアッセイの概略図をここに示す。バッファーA、溶液B、および溶液Cは、プロトコルに記載されているように調製されます。時間を節約し、この方法をフィールドCLas検出に適したものにするために、大腸菌ポリエチレングリコールベースのアプローチを使用して、DNA増幅用の粗植物抽出物を取得しました。柑橘類の葉がツールに使用されています。葉から葉のディスクのいくつかの部分を打ち抜きます。リーフディスクを1.5ミリリットルのエッペンドルフチューブに入れます。次に、200マイクロリットルのバッファーAをチューブに追加します。手動でプラスチック棒で滑らかになるまでリーフディスクを挽きます。チューブを邪魔せずに10分間放置します。その後、上清をDNA増幅に応用する。等温DNA増幅のために、ステップ2.3の上清1マイクロリットルと酢酸マグネシウム1マイクロリットルを溶液Bに加え、よく混合します。実験室で、37度の単純なインキュベーターで15分間インキュベートします。結果を視覚化するには、ステップ3.2の混合物10マイクロリットルを溶液Cに加え、よく混合します。37度のインキュベーターで60分間インキュベートします。ゴーグルを着用し、ハンドヘルド蛍光検出装置を通して緑色の蛍光シグナルを観察します。CLas-DETECTRの特異性を試験するために、まずアグロバクテリウム・ツメファシエンスGV3101、キサントモナス・シトリ亜種を純粋なコロニーとした。私たちの研究室で分離されたカンキツ潰瘍の病原体であるシトリとバークホルデリアスタビリス株1440を使用して、それらのゲノムDNAを抽出しました。次にこの3つの細菌ゲノムDNAおよびCLas陽性ニューホール葉ゲノムDNAをCLas-DETECTR試験に供した。CLas-DETECTRの感度をテストするために、プロトコルに記載されているように、PCR、CLas-DETECTR、およびqPCRを使用して一連のCLas DNAフラグメント希釈を検出しました特異性と感度テストの後、CLas-DETECTR法を使用して、フィールド内のCLasの存在を検出しました。中国江西省の甘南師範大学のキャンパスにある生殖質資源保育園で育てられたニューホールスイートオレンジの木から収集された葉のサンプル。ラボで通常使用される説得力のあるCLas検出方法であるqPCRも適用されました。HLB感染およびHLB非感染ニューホールツリーの葉サンプルで、CLasの存在が私たちの研究室で確認され、CLas-DETECTRテストに供されました。緑色の蛍光シグナルは、HLBに感染したサンプルでは捕捉されましたが、HLB非感染および陰性対照では捕捉されませんでした。CLas-DETECTRの特異性を他の細菌から抽出した核酸を用いて決定した。CLas-DETECTRアッセイでは、CLas陽性のニューホール葉から抽出されたgDNAのみが緑色の蛍光シグナルを示しました。アグロバクテリウム・ツメファシエンスGV3101、キサントモナス・シトリ亜種シトリおよびバークホルデリア・スタビリス1440株から抽出されたgDNAは抽出されなかったため、CLas-DETECTRはCLasを特異的に検出できることを意味します。また、プロトコルに記載されている一連のCLas DNA希釈液を使用して、CLas-DETECTRの感度をテストしました。PCRはメガリットルあたり201コピーを検出できます。一方、CLas-DETECTRはメガリットルあたり2.01コピーを検出でき、高感度フィールド診断として利用可能であることを示唆しています。qPCRはメガリットルあたり0.201コピーを検出でき、Ct値は36を超えています。したがって、CLas-DETECTRは、希釈されたCLas特異的アンプリコンを使用した場合、従来のPCRよりも2桁感度が高く、qPCRよりも1桁感度が低くなります。最後に、フィールドサンプルに対するCLas-DETECTRの実現可能性を再検討し、その感度をqPCRと比較しました。数字で見る。CLas濃度がメガリットルあたり0.2コピー未満の場合、Ct値は36よりも高く、非常に低い濃度であることがわかります。15サンプルのうち、qPCRで検出されたサンプル1、2、3、4、8、10、13、14のCt値は36未満であった。明らかな緑色の蛍光シグナルが観察された。一方、qPCRで検出されたサンプル6、7、9、12、15のCt値が未定の場合、緑色の蛍光シグナルは観測されなかった。また、qPCRで検出したサンプル5,11のCT値が36より高い場合には、弱い緑色の蛍光シグナルが認められた。結果は、私たちのプラットフォームが現場でのHLB診断のための迅速で堅牢かつ高感度なツールであることを示唆しています。
