L. 아메리카누스2를 포함하는 체관부 집락화 및 까다로운 박테리아 칸디다투스 리베리박터 종에 의해 발생합니다. 중국과 미국에서 가장 널리 퍼진 HLB 관련 종은 아시아 감귤류 프실라(Diaphorina citri) 또는 접목을 통해 전염되는 CLas입니다.3. CLas에 감염된 후 감귤 나무는 죽을 때까지 성장 감소를 보여줍니다2. CLas에 감염된 감귤류 잎의 일반적인 증상은 얼룩덜룩 한 얼룩, 녹색 섬 (작은 원형 짙은 녹색 점), 두껍고 가죽 같은 잎에 융기 된 코르키 정맥, 불균일 한 황변 싹2입니다. 또한 CLas에 감염된 과일은 작고 편향된 것처럼 보입니다2.
감귤류 품종은 HLB에 내성이 없고 HLB에 대한 치료법이 없기 때문에 HLB를 예방하려면 CLas 양성 감귤류 나무의 격리 및 분리가 필요합니다 2,3. 따라서 CLas의 확산을 방지하고 경제적 손실을 최소화하기 위해 모니터링 및 격리에 조기 발견이 중요합니다3. 또한, 감염 초기 단계에서 식물에서 CLas의 낮은 역가로 인해 민감한 CLas 검출이 필요합니다3. 중국에서 CLas 검출은 일반적으로 인증된 특정 테스트 센터에서 수행됩니다. 그러나 탐지 과정은 일반적으로 최소 1 주일이 걸리며 탐지 수수료가 비쌉니다. 따라서 HLB 발생률을 모니터링하는 데 도움이 됩니다.
HLB 4,5,6,7,8,9를 진단하기 위해 다양한 기술이 적용되었습니다. 중합효소연쇄반응(PCR) 및 정량적 PCR(qPCR)은 높은 감도와특이성으로 인해 CLas 검출에 가장 많이 사용되는 도구입니다4,5. 그러나 이러한 기술은 값비싼 기기와 고도로 숙련된 인력에 크게 의존합니다. 또한, 루프 매개 등온 증폭 (LAMP)과 같은 여러 등온 증폭 방법은 단순성, 신속성 및 저렴한 비용으로 인해 기존 PCR 방법에 대한 매력적인 대안으로 개발되었습니다 8,9,10. 그러나 비특이적 증폭 신호로 인해 CLas를 정확하게 검출하기 위해 이를 적용하는 것은 어렵고 위양성 결과를 유발할 수 있습니다.
RNA 유도 CRISPR/CAS(클러스터링된 규칙적인 간격의 짧은 회문 반복/CRISPR 관련) 엔도뉴클레아제 기반 핵산 검출은 높은 감도, 특이성 및 신뢰성 11,12,13,14로 인해 차세대 분자 진단 기술로 개발되었습니다. 이러한 CRISPR/Cas 진단 기술은 Cas 단백질의 부수적인 뉴클레아제 활성에 의존하여 올리고뉴클레오티드의 각 말단에 있는 형광 리포터와 형광 소광제로 변형된 단일 가닥 DNA(ssDNA)를 절단하고 방출된 형광 리포터11,12를 캡처하는 형광 검출 장치에 의존합니다. . Cas12a의 DNase 활성과 RPA 및 형광 판독을 결합한 DETECTR(DNA Endonuclease Targeted CRISPR Trans Reporter)라는 검출 플랫폼이 최근에 고안되었으며12 더 높은 감도의 핵산을 검출하는 것으로 나타났습니다20. 또한 양성 샘플에서 방출된 형광 신호는 현장에서 휴대용 형광 검출 장치를 통해 관찰할 수 있습니다.
RPA로 DNA를 증폭하고, CLas21에 특이적인 5카피 nrdB(리보뉴클레오티드 환원효소 β-서브유닛) 유전자를 표적으로 하는 crRNA를 설계하고, Cas12a 단백질의 DNase 활성을 사용했기 때문에 이를 CLas 검출 방법을 CLas-DETECTR라고 불렀습니다. 기존 CLas 검출 방법과 비교하여 CLas-DETECTR는 빠르고 정확하며 민감하고 배포가 가능합니다.
