一次生産性は、主に光強度、温度、栄養素の利用可能性、および共光帯3,4のそれぞれの深さにおける炭酸塩系のイオン種の分布によって駆動されます。深さは微生物マットの空間分布に著しく影響します。微生物群集は、浅い深さでの高照射と顕著な季節温度変化の悪影響に対処し、より深い深さではより低い光強度に対処する必要があります。深さ勾配に加えて、動的栄養相互作用は異なるスケールで複数の複雑な空間パターンを生成します5。この複雑なシステムは、実験室でシミュレートするのが複雑です。沿岸域から個々の一次生産者の代謝活性を推測する最も正確な方法は、in situ実験を設定することです。
本稿で紹介する方法論は、従来のチャンバー法2,6,7と、輸送可能で建造が容易な低コストのフローティングバージに基づいています。これにより、自然光スペクトル、温度、および深さに伴う炭酸塩系のイオン種の異なる分布の下で、異なる深さでの一次生産性の測定が可能になります。この方法は、植物プランクトンの光合成を測定するために最初に採用され6、現在でも一般的に使用されている明るいボトル酸素と暗いボトル酸素の原理に基づいています6,7。光の中に保たれたボトル内の酸素の変化率(一次生産性と呼吸の影響を含む)と暗所に保たれたもの(呼吸のみ)を比較します8。この方法は、一次生産性の代理として酸素発生(光合成)を使用します。測定された変数は、正味の生態系生産性(NEP、光条件下での経時的なO2濃度の変化として)および生態系呼吸(RE、暗所での経時的なO2濃度の変化として)である。総生態系生産性(GEP)は、2つの差の計算です(表1)。ここでは「生態系」という用語は、ペリフィトンが独立栄養生物と従属栄養生物で構成されていることを示すために使用されます。この従来のチャンバー法の最も重要な改善点は、非侵襲的酸素光学センサーを使用し、周辺一次生産性を測定するためのこの主にプランクトンの方法を最適化することです。

適切な場所におけるペリフィトンまたは任意の水微生物の総生産性の詳細なin-situ測定は、レンティック水中の一次生産性の動的を制御するプロセスに関する現在の知識を向上させることができます。提案手法の主な改善点は、非侵襲性、費用対効果、およびさまざまな場所での同時測定の可能性です。また、追加費用なしで大規模なデータ資産を収集できます。実験を行う際にバージを交換する場合は、持ち運びが簡単なインフレータブルカヤックを使用してください。フロートを固定するのに理想的な深さの場所を選択します。次に、組み立てられたはしけをボートの船尾の後ろに取り付け、ボートの側面に沿ってアンカーを慎重に下げます。アンカーを水面の少し下に吊るして位置合わせし、フロートをアンカーで必要な深さのスポットに簡単に牽引できるようにします。その場所に着いたら、ボートからアンカーをほどいて底まで下げ、はしけをアンカーチェーンに固定し、インキュベーションボトルをバージに取り付けるためのチェーンを固定します。インキュベーションボトルを準備するには、気密シール付きの透明でワイドネックの0.5リットルボトルの内壁に酸素光学センサースポットを取り付け、黒い電気テープで包んで暗いトリートメントボトルの不透明な層を作ります。配置した光学センサーのスポットに小さな穴を開け、カット穴がセンサーの直径よりわずかに小さいことを確認して、光がボトルに入らないようにします。インキュベーションボトルをポータブルボックスに入れ、周囲の水の沈殿物を乱すことなく、ボックスと一緒にそれぞれの深さまで潜ります。次に、長いピンセットを使用して、サンプルのバイオマスを乱しすぎないように注意しながら、サンプルをインキュベーションボトルに慎重に充填します。微生物マットが小さな石などの固い表面に成長する場合は、無傷のバイオマスを含む石全体をボトルに慎重に移します。明るいボトルと暗いボトルの1組に、沈殿物のないそれぞれの深さからのきれいな水を入れて、空白のコントロールとして機能します。すべてのインキュベーションボトルの水がきれいで、邪魔な沈殿物が含まれていないことを確認した後、閉じたボトルをフローティングバージに固定されたボートに持ってきます。最初の2組のインキュベーションボトルを最初のチェーンのスナップフックに取り付け、光ファイバー酸素計を使用して各ボトルの初期酸素濃度を測定します。メーターの光ケーブルをボトル内に取り付けられた酸素センサーに接続し、数秒以内にメーター内の酸素濃度を読み出して測定値を記録します。測定後すぐに、付属のボトルでチェーンを慎重に水中に戻し、インキュベーションボトルがそれらの中に入れられたバイオマスがサンプリングされたのと同じ深さに配置されていることを確認します。1時間後、ボトルの入った各チェーンをボートに慎重に引っ張って、酸素濃度を再度測定します。酸素値の読み取りは以前に実証され、サンプルを再び水中に下げました。すべての測定が完了したら、インキュベーションボトルからサンプルを取り出し、石などの硬い物質の表面に成長した微生物マットを歯ブラシや小さなナイフでこすり、内容物をプラスチックフラスコに移します。時間の経過に伴う酸素濃度の増加は、対照と光にさらされたサンプルボトルの両方で明らかであり、正味の生態系生産性を示しています。それにもかかわらず、増加の傾きは、微生物マットサンプルを含むボトルで有意に高くなります。暗いボトル内の時間の経過に伴う酸素濃度の変化は、独立栄養呼吸と従属栄養呼吸の合計です。この場合、コントロールボトル内の酸素濃度の傾きはゼロと有意に異ならない。生態系の正味生産性から呼吸率を差し引くと、生態系の総生産性が得られ、データは定義によく準拠しています。この方法の実用化は、植生期における周生生物群集の総生態系生産性を示す3つの採掘後の湖の畑で達成されました。水中でのバイオマスサンプリングプロセスでは、サンプルを追加した後に閉じた後、インキュベーションボトル内に気泡が閉じ込められないようにすることが重要です。この手順を使用して、水域と適切なサイズの任意の生物または生物群集との間の酸素交換を定量化することができる。この方法では、自然条件を可能な限り作り出すことで、選択した生物の年間を通じてその場で一次生産性を研究することができ、生涯の炭素代謝に対するその相対的な重要性をよりよく理解できます。
