1 차 생산성은 전체 시스템 먹이 그물을 형성하는 수생 시스템으로 자동 탄소가 들어가는 유일한 항목입니다1. 따라서 1 차 생산성의 정확한 추정은 수생 생태계의 기능을 이해하는 데 필수적인 단계입니다. 연안 지역은 1 차 생산성과 생물 다양성이 높은 지역입니다. 식물성 플랑크톤 외에도 페리피톤(이하 미생물 매트라고 함)과 거대조류는 연안 지역2의 1차 생산성에 크게 기여하는 것으로 추정됩니다. 그들의 정착 생활 방식과 상당한 공간적 이질성으로 인해 1 차 생산성의 정량화는 쉽지 않습니다. 
1 차 생산성은 주로 광도, 온도, 영양소의 가용성 및 유포 영역 3,4의 각 깊이에서 탄산염 시스템의 이온 종의 분포에 의해 주도됩니다. 깊이는 미생물 매트의 공간 분포에 현저한 영향을 미칩니다. 미생물 군집은 얕은 깊이에서 높은 조사와 뚜렷한 계절적 온도 변화의 부작용에 대처하고 더 깊은 깊이에서 더 낮은 광도에 대처해야 합니다. 깊이 구배 외에도 동적 영양 상호 작용은 서로 다른 스케일5에서 여러 개의 복잡한 공간 패턴을 생성합니다. 이 복잡한 시스템은 실험실에서 시뮬레이션하기가 복잡합니다. 연안 지역에서 개별 1 차 생산자의 대사 활동을 추론하는 가장 정확한 방법은 현장 실험을 설정하는 것입니다.

온전한 미생물 매트의 샘플을 수집하여 스쿠버 다이버가 지정된 깊이의 기밀 배양 병에 넣습니다. 각 병에는 시간 경과에 따른 O2 생산성 / 소비를 모니터링하는 비 침습적 광학 산소 마이크로 센서가 장착되어 있습니다. 모든 측정은 각 깊이에서 5개의 반복 어두운/밝은 쌍으로 수행됩니다. 온도 및 광합성 활성 방사선(PHAR) 강도는 배양 전반에 걸쳐 각각의 깊이에서 측정됩니다. 6시간의 계내 배양(일광 시간) 후, 미생물 매트를 병으로부터 수확하고 건조시킨다.
