1. 仪器和动物的准备

	</ol></li>
	<li>用干热灭菌器对所有手术器械进行消毒。</li>
	<li>为窗户植入手术准备小鼠。 注意：小鼠应达到适当的年龄。优选的是，它们经过基因工程改造以表达CA1和齿状回（DG）中的荧光蛋白。这些要点将在讨论部分进一步解释。</li>
</ol>2.麻醉和头部固定
<ol><li>通过腹膜内注射氯胺酮 - 甲苯噻嗪（100mg / kg氯胺酮和10mg / kg甲苯噻嗪）麻醉小鼠。皮下注射美洛昔康（2mg / kg）用于术前镇痛。确认对疼痛刺激的反应消失，例如尾部捏，以确保足够的麻醉深度。每次麻醉在手术过程中消失时加入半剂量的氯胺酮 - 甲苯噻嗪，通常在初始给药后1小时，此后每30分钟一次。在身体下方使用加热垫提供热支持，并在眼睛上涂抹眼药膏以防止麻醉下干燥。 注意：手术麻醉的选择至关重要。这一点在讨论部分有详细讨论。</li>
	<li>涂抹脱毛膏去除头皮上的毛发。用聚维酮碘磨砂膏和酒精以圆周运动对头皮进行几次消毒。将鼠标放在用无菌防水垫准备的手术平台上，并应用无菌布以确保手术部位。然后，用手术刀圆形切割并去除头皮，使顶骨和枕骨完全暴露在手术侧。</li>

	<li>将牙科蚀刻材料涂在整个表面上，等待数十秒，然后用足够的无菌盐水冲洗。</li>
	<li>使用防水墨水标记要去除的骨骼区域（直径为3.0毫米的圆形区域，中心距后方约2.5毫米，后部约2.5毫米，后侧约2.5毫米）。标记后，彻底擦干颅骨表面。 注意：开颅术的位置取决于海马体的大小，应针对不同的年龄进行优化。下面将介绍开颅术位置的有用指标（见步骤3.4.5，注）。</li>
	<li>根据制造商的说明，使用牙科树脂水泥将厚度为1.0毫米，中心有一个直径为5.0毫米的孔的矩形铝板连接到头骨上。<ol><li>小心地将板中心的孔与用笔标记的圆形区域对齐（见步骤2.5），使铝板平行于头骨的标记表面（图1B）。确保水泥紧紧密封板和骨头之间的所有间隙。等待约5分钟，使水泥充分硬化。</li>
	</ol></li>

</ol>3.观察窗的植入
注意：以下外科手术应在体视显微镜下进行。
<ol><li>使用直径为3.0毫米的真皮冲头在头骨上做一个圆形凹槽。在步骤 2.5 中对齐圆形凹槽和用笔标记的区域。用轻轻的压力扭转真皮冲头，使凹槽深度逐渐增加。经常检查凹槽深度在整个圆周上是否恒定。</li>

	<li>使用硬脑膜拾取器取出硬脑膜。 注意：完全去除颅窗内的硬脑膜是必要的，并且需要使用细镊子小心地切除外围的残余硬脑膜。</li>
	<li>轻轻吸出组织，通过连接到吸引器的 23 G 或 25 G 钝针暴露海马肺泡。 注：此步骤对于成功映像最为关键。组织抽吸的关键技术要点在讨论部分有详细说明。<ol><li>吸出软脑膜和软脑膜血管。</li>
		<li>从表面吸出皮质组织。在整个颅窗上保持暴露组织表面的深度均匀。逐渐加深抽吸，直到外囊的纤维暴露出来。</li>
		<li>小心地将吸头放在颅窗前外侧端的侧脑室 （LV） 附近的外囊上。去除沿尾状向前外侧方向运行的外囊表层，然后去除沿中外侧方向运行的内层。 注意：LV附近的外部话筒头可以很容易地从底层结构中移除。</li>
		<li>通过检查肺泡表面和背海马连合（DHC）的完全暴露来确认整个外部胶囊的去除。 注意：DHC 覆盖 CA1 的尾部。肺泡和DHC可以通过它们的纤维取向与外部胶囊区分开来。也就是说，肺泡内的纤维和DHC沿前内侧到尾外侧方向运行，并且可以与外部胶囊中的纤维区分开来。肺泡和DHC紧紧地附着在CA1上，不应损坏。应去除外部胶囊的所有碎片，因为任何剩余的碎片都会阻止玻璃盖玻片与肺泡的直接接触。</li>
		<li>确认出血已彻底停止，并用无菌盐水填充孔至颅骨表面的水平。 注意：此处的视野有助于判断孔是否处于适合鼠标特定年龄的位置。理想情况下，肺泡和下面的CA1应占据中心区域的大部分。DHC的一部分在尾状区域可见。左心室的开口可在前外侧边缘检测到（图3A）。</li>
	</ol></li>
	<li>将玻璃底金属管（见步骤1.1）垂直插入孔中，同时吸出从管子侧面溢出的多余水，直到玻璃底部轻轻压在肺泡上并部分压平下面的CA1。 注意：此压力需要适当调整。如果太强，CA1会损坏。如果太弱，由于CA1曲率引起的球差将损害深层结构的成像分辨率。</li>
	<li>使用牙科树脂水泥，将插入管的壁固定在周围的颅骨上（图1C）。确保整个圆周密封严密，没有空气或脑脊液泄漏。等待约5分钟，让水泥硬化。 注意：如果水泥在其粘度低时放置，它可能会通过管子和颅骨之间的间隙泄漏到脑实质中并损害大脑。因此，水泥应在聚合引发后应用，这样可以增加粘度并减少通过间隙的泄漏。</li>
	<li>用无菌盐水清洗整个板、颅骨和管内以去除碎屑。</li>
</ol>4. 手术后立即进行双光子显微镜检查，以检查手术质量
<ol><li>将鼠标放在双光子显微镜物镜下的头部固定装置中，并放在电动XY扫描载物台上。使用加热垫进行热支撑。如步骤2.1中所述监测和调整麻醉深度，因为此质量检查可能需要长达30分钟。 注意：建议使用工作距离（WD）大于3 mm且数值孔径（NA）较高的水浸物镜。物镜的校正环可以通过补偿玻璃和生物材料之间的折射率不匹配来提高分辨率。</li>
	<li>用水填充玻璃和物镜之间的空间，特别注意避免气泡。如有必要，使用可容纳更多水的塑料薄膜扩大金属板的面积，以覆盖物镜的整个前透镜。</li>
	<li>打开920nm波长的飞秒脉冲激光器以激发大脑中表达的荧光探针并启动图像采集软件。</li>
	<li>使用电动载物台和电动对焦系统将焦点调整到 CA1。在脉冲激光的连续照明下，在脑实质发出的荧光和金属管边缘的反射光的引导下定位目标结构。</li>
	<li>通过使用电动对焦系统调整焦点来测量接触玻璃底部的脑实质的深度。比较不同水平位置的深度，以判断玻璃底部和成像平面的对齐情况。<ol><li>通过倾斜头部固定装置来调整鼠标头的角度，直到将玻璃底部设置为平行于成像平面。</li>
	</ol></li>
	<li>调整物镜的校正环，以在CA1中目标结构的深度处实现最高分辨率。</li>
	<li>确认DG上叶片分子层（ML）中的荧光细胞可以在距玻璃底部500μm的深度在整个视野中成像。 注意：此步骤对于判断手术质量很重要。如果CA1受损，局灶性脑水肿会阻止距离表面超过200μm深度的荧光检测。只有当CA1没有损坏，并且CA1层的曲率被上覆的盖玻片适当地压平时，才能在手术后立即检测到DG信号（图2A-D）。代表性结果部分提供了一种确定CA1和DG之间边界的简单方法。</li>
</ol>5. 术后护理
<ol><li>吸出管内的水并用塑料密封盖住，以防止碎屑进入。</li>
	<li>保持鼠标温暖并等待从麻醉中恢复。</li>
	<li>每24小时皮下注射美洛昔康（2mg / kg），只要小鼠表现出疼痛相关行为。</li>
	<li>在单个外壳中举起术后小鼠数周。</li>
</ol>6. 使用 CX3CR1-GFP 小鼠对小胶质细胞进行体内慢性双光子成像
<ol><li>诱导麻醉后，用无菌盐水清洗金属管内部以去除碎屑。确保通过玻璃可以看到海马体的白色肺泡（图3A），并且没有术后出血等并发症。</li>
	<li>按照步骤4.1至4.6将鼠标置于双光子显微镜下。</li>

这种方法包含复杂的外科手术，但如果准备得当，它可以在整个CA1上长时间提供高分辨率图像。几只小鼠的实验证实，慢性术后阶段的图像质量优于急性术后成像。更好的图像质量保持了2个多月。失败的最常见原因是手术过程中CA1的意外损坏，这在术后质量检查中确定（见步骤4）。这可能是由于误吸直接伤害、大脑干燥、玻璃压力过大或最后一步牙科水泥的毒性。失败的另一个原因是术后出血，可以在手术后1周内通过玻璃底窗确认（见步骤6.1）。仔细阅读协议并采取一切预防措施以避免此类麻烦。
即使目前在实验室中使用带有GaAsP探测器的双光子显微镜，由于不可忽视的光散射，试图通过皮层对背侧CA1进行成像也会导致分辨率的显着损失。因此，为了获得足够的分辨率来观察CA1中小胶质细胞过程的运动或神经元树突棘的形成，去除覆盖皮层的一部分是不可避免的，就像在这种方法中一样。在保持高图像分辨率的同时减少皮质去除程度的唯一替代方法是嵌入中继透镜，例如梯度折射率（GRIN）透镜。在这种方法中，GRIN透镜被放置在直接植入CA1上方的导管内，用于慢性CA1成像28，29。导管的外径为1.8毫米，小于本文中装置的3.0毫米，同时产生与此处系统相当的高分辨率（NA;0.82）（有效NA;0.88）。然而，使用GRIN镜头的方法具有用于高分辨率观察的窄视野和受GRIN镜头WD限制的短成像深度。因此，以高分辨率在宽视场内对所有海马层进行成像是本文该方法的一个具体优点。
该程序的局限性是皮层的部分去除和炎症可能对海马体产生一些不利影响。但由于被移除的皮层没有直接进入海马体，内嗅皮层没有损伤，海马体本身也没有直接损伤，总体评价是海马体的功能障碍不显著。先前的几项研究已经证实，海马窗口植入后保留了海马功能，包括学习和记忆25，26，27，30，31，32，33，34。因此，这里描述的成像方法有望在足够的术后时间后忠实地报告海马功能。

本文侧重于小胶质细胞成像，但相同的成像程序适用于CA1中的其他神经元和神经胶质细胞类型。此外，该方案仅限于麻醉小鼠的成像，但该技术也可以扩展到监测清醒小鼠的海马小胶质细胞。由呼吸、心跳和身体运动引起的运动伪影，特别是在远离玻璃窗的深层海马层中，可能存在于清醒小鼠的实验中。因此，可能需要适当的运动校正系统来捕获小胶质细胞形态和动力学。
与《议定书》有关的讨论 建议选择具有适当年龄和遗传修饰的小鼠来表达具有时间和空间特异性的荧光探针（参见步骤1.3）。1个月大的小鼠可用于实验，但2个月以上的小鼠更容易处理，它们的体型较大，抗手术压力。此外，在年轻小鼠中，海马体的外部囊和肺泡更难分离。因此，在年轻小鼠中去除外部胶囊需要手术技巧（参见步骤3.4.3-4）。对于在CA1和DG中可重复且均匀地表达荧光蛋白的小鼠，手术评估和随后的成像将更加直接（参见步骤4.7）。因此，在手术的初始试验中，建议使用具有稀疏标记神经元的转基因小鼠，例如Thy1-YFP或Thy1-GFP小鼠39，或具有标记小胶质细胞的小鼠，例如CX3CR1-GFP小鼠24。
对于成功的手术，麻醉的选择很重要（见步骤2.1）。不同类型的麻醉可引起不同程度的术中脑水肿，影响手术难度、植入金属管的相对位置以及玻璃窗对脑的压迫程度。这些因素也可能影响手术后海马神经元的存活。
在暴露海马体的抽吸过程中（见步骤3.4），应注意以下几点，以尽量减少对大脑的损害，确保成像成功。通过在颅窗一侧放置一个出口，不断将无菌盐水供应到组织表面。防止组织表面在抽吸过程中暴露在空气中。组织抽吸的基本原理是通过液体流入吸头来去除组织表面。应避免吸头与组织直接接触。将施加在吸入管上的负压调整为最小值。逐步吸出组织，并确认每一步出血都得到控制。当出血时间延长时，请等到出血自发停止。保持抽吸距离出血部位较短的距离，并持续流动无菌盐水。吸出组织以创建一个圆柱形空间，其大小适合玻璃底金属管（参见步骤1.1）。选择 23 G 针头吸出厚血管或大组织碎片，选择 25 G 针头吸吮小组织碎片。

手术后立即在CX3CR1-GFP小鼠中小胶质细胞的代表性图像如图 2所示。如果CA1没有明显损伤，并且CA1层的曲率被上覆的盖玻片适当地压平，则小胶质细胞的双光子成像深度超过整个CA1层，从手术表面达到500μm，其中存在DG的ML或颗粒细胞层（GCL）（图2A;见步骤4.7和 图4E).小胶质细胞的分支略少，特别是在靠近手术表面的层中，与完整海马体中的小胶质细胞相比。然而，它们没有表现出明显的激活，表明适当的外科手术（图2B）。另一方面，由组织损伤引起的CA1水肿会将成像深度限制在200μm（图2C）。组织损伤也会引起异常的小胶质细胞活化，例如向游离组织表面积聚凸起的过程（图2D;与图 2B的上图相比）。这些是不适当手术的迹象。
术后3周以上慢性期小胶质细胞的代表性图像如图3所示。小胶质细胞可以在CA1之外再次成像到DG的更深层，具有更高的荧光强度和分辨率以及更均匀的分布（参见步骤6.3，图3B）。成像质量优于手术后立即（图2A）。这种差异可以通过减少水肿和炎症来解释。所有层中的小胶质细胞已经恢复了其分支形态（图3C），与术后外观不同（图2B）。CA1中小胶质细胞的延时成像揭示了用于监测的固定细胞体和高度运动的过程（视频1-2）。它们的运动行为类似于之前关于皮层9中小胶质细胞过程动力学的报告。
