관상동맥 질환(CAD)은 전형적이고 대표적인 심혈관 질환으로 널리 인식되고 우려되고 있으며, 선진국과 개발도상국 모두에서 사망의 주요 원인이 되고 있다 1,2. 정상적인 심장 생리 기능을위한 혈액 및 산소 공급 경로로서, 순환 혈액은 심근 3,4의 표면에 두 가지 주요 관상 동맥과 혈액 혈관 네트워크를 통해 심장에 들어가고 영양을 공급합니다. 관상 동맥의 콜레스테롤과 지방 침착은 심장의 혈액 공급과 혈관 시스템의 폭력적인 염증 반응을 차단하여 죽상 동맥 경화증, 안정 협심증, 불안정 협심증, 심근 경색 또는 갑작스런 심장 사망을 유발합니다 5,6. 관상동맥의 병리학적 협착에 반응하여, 보상적 가속 생리적 심장 박동은 좌심실의 출력을 증가시킴으로써 심장 자체 또는 신체의 중요한 기관의 혈액 공급을 만족시킨다(7). 장기간의 관상 동맥 협착증이 제 시간에 완화되지 않으면 심장의 특정 영역에서 광범위한 새로운 혈관이 발생할 수 있습니다8. 현재, CAD의 임상 치료는 종종 약물 혈전 용해 또는 외과 적 기계적 혈전 용해 및 빈번한 약물 치료와 큰 외과 적 장애가있는 외인성 생체 공학 혈관 우회를 채택합니다 9. 따라서, 관상동맥 생리활성의 기능적 조사는 심혈관 질환(10)에 대한 여전히 시급한 돌파구이다.
생체 내 관상 동맥 압력, 혈관 긴장, 혈중 산소 포화도 및 pH 값11을 동적으로 기록 할 수있는 무선 원격 측정 시스템을 제외하고는 관상 동맥 생리 활성을 검출 할 수있는 기술적 수단이 없습니다. 따라서 관상 동맥의 텍스트 기밀성과 복잡성을 고려할 때 관상 동맥의 정확한 식별 및 분리는 의심 할 여지없이 시험관 내CAD의 여러 메커니즘을 탐구하기위한 최선의 선택입니다.
시리즈 다중 근그래프 시스템, 특히 와이어 현미경 사진 미세혈관 장력 검출기 ( 재료 표 참조)는 고정밀 및 연속 동적 기록의 특성을 갖는 작은 혈관, 림프관 및 기관지 관의 시험관내 조직 장력 변화를 기록하기 위한 매우 성숙한 시장성 장치이다(12). 상기 시스템은 직경이 60 μm 내지 10 mm인 공동구조체의 시험관내 조직 장력 특성을 기록하기 위해 광범위하게 이용되었다. 와이어 현미경 사진의 플랫폼의 연속 가열 특징은 불리한 외부 환경의 자극을 크게 상쇄합니다. 한편, 가스 혼합물 및 pH 값의 일정한 입력은 유사한 생리학적 상태(13)에서 보다 정확한 혈관 긴장 데이터를 얻을 수 있게 한다. 그러나 쥐 관상 동맥의 해부학 적 국소화의 복잡성을 고려할 때 (그림 1), 그 분리는 다양한 심혈관 질환 및 약물 개발에 대한 메커니즘의 탐구를 혼란스럽게하고 제한하고 있습니다. 따라서, 본 프로토콜은 래트 관상동맥의 해부학적 위치 및 분리 과정을 상세히 소개하고, 이어서 와이어 현미경 사진(14)의 플랫폼 상에서 장력 측정을 한다.
