EV's zijn in opkomst om een belangrijke rol te spelen in diverse biologische activiteiten, waaronder antigeenpresentatie, genetisch materiaaltransport, celmicro-omgevingsmodificatie en andere. Bovendien brengt hun brede toepassing nieuwe benaderingen en mogelijkheden voor het diagnosticeren en behandelen van ziekten21. Stamcellen zijn ongedifferentieerde pluripotente cellen met zelfvernieuwingsvermogen en translatiepotentieel1. Mesenchymale stamcellen (MSC's) worden gemakkelijk geïsoleerd, gekweekt, uitgebreid en gezuiverd in het laboratorium, wat kenmerkend blijft voor stamcellen na meerdere passages. In de afgelopen jaren is er steeds meer bewijs dat de opvatting ondersteunt dat MSC's in een paracriene modus werken bij therapeutisch gebruik 2,3. Vooral de afscheiding van extracellulaire blaasjes (EV's) speelt een cruciale rol bij het bemiddelen van de biologische functies van MSC's. Als heterogene vliezige nanodeeltjes die vrijkomen uit de meeste celtypen, bestaan EV's uit subcategorieën die exosomen (Exos), microvesicles (MV's) en nog grotere apoptotische lichamenworden genoemd 4,5. Onder hen is Exos de meest bestudeerde EV met een grootte van 40-150 nm, die van endosomale oorsprong is en actief wordt uitgescheiden in fysiologische omstandigheden. MV's worden gevormd door rechtstreeks van het oppervlak van het celplasmamembraan af te stoten met een diameter van 100-1.000 nm, die worden gekenmerkt door een hoge expressie van fosfatidylserine en expressie van oppervlaktemarkers van donorcellen6. EV's bevatten RNA, eiwitten en andere bioactieve moleculen, die vergelijkbare functies hebben als de oudercellen en een belangrijke rol spelen bij celcommunicatie, immuunrespons en herstel van weefselschade7. MSC-EV's zijn op grote schaal onderzocht als een krachtig celvrij therapeutisch hulpmiddel in regeneratieve geneeskunde8.
Isolatie en zuivering van MSC-afgeleide EV's is een veel voorkomend probleem op het gebied van onderzoek en toepassing. Op dit moment zijn differentiële en dichtheidsgradiënt ultracentrifugatie9, ultrafiltratieproces 10, immunomagnetische scheiding11, moleculaire uitsluitingschromatograaf 12 en microfluïdische chip13 veelgebruikte methoden bij de isolatie en zuivering van EV's. Met de voor- en nadelen van elke aanpak kan niet tegelijkertijd worden voldaan aan de hoeveelheid, zuiverheid en activiteit van ingezamelde EV's14,15. In de huidige studie wordt het differentiële centrifugatieprotocol van isolatie en karakterisering van EV's uit gekweekte MSC's in detail getoond, wat efficiënt therapeutisch gebruik heeft ondersteund 16,17,18,19,20. Het aanpassingsvermogen van deze methode voor een reeks downstream-benaderingen, zoals fluorescerende etikettering, lokale transplantatie en systemische injectie, is verder geïllustreerd. De implementatie van deze procedure zal tegemoetkomen aan de behoefte aan eenvoudige en betrouwbare verzameling en toepassing van MSC-EV's in translationeel onderzoek.

Extracellulaire blaasjes (EV's) zijn heterogene membraannanodeeltjes die door de meeste celtypen worden vrijgegeven en ze worden steeds meer erkend als fysiologische regulatoren van organismale homeostase en belangrijke indicatoren van pathologieën; In de tussentijd ontstaat hun immense potentieel om toegankelijke en controleerbare ziektetherapieën op te zetten. Mesenchymale stamcellen (MSC's) kunnen grote hoeveelheden EV's in cultuur vrijgeven, die veelbelovend zijn gebleken om effectieve weefselregeneratie op gang te brengen en uitgebreide therapeutische toepassingen met goede schaalbaarheid en reproduceerbaarheid mogelijk te maken. Er is een groeiende vraag naar eenvoudige en effectieve protocollen voor het verzamelen en toepassen van MSC-EV's. Hier wordt een gedetailleerd protocol geleverd op basis van differentiële centrifugatie om representatieve EV's te isoleren en te karakteriseren uit gekweekte menselijke MSC's, exosomen en microvesicles voor verdere toepassingen. Het aanpassingsvermogen van deze methode wordt aangetoond voor een reeks downstream-benaderingen, zoals etikettering, lokale transplantatie en systemische injectie. De implementatie van deze procedure zal tegemoetkomen aan de behoefte aan eenvoudige en betrouwbare MSC-EV's die worden verzameld en toegepast in translationeel onderzoek.

MV's en Exo's van gekweekte menselijke UCMSC's worden geïsoleerd volgens de experimentele workflow (figuur 1). Deze procedure voorzag in de behoefte aan eenvoudige en betrouwbare mesenchymale stamcel-afgeleide extracellulaire blaasjesverzameling, en van toepassing bij de vertaling van onderzoek. Het verzamelen van extracellulaire blaasjes door differentiële centrifugatie is eenvoudig, gemakkelijk schaalbaar en reproduceerbaar, wat nuttig zal zijn voor het veld. Extracellulaire blaasjes onduidelijk}van onduidelijk}mesenchymale stamcel is een onduidelijke}toepassing in moderne ziektebehandelingen.Ik zal niet bang zijn voor fouten. Deze bediening is eenvoudig en gemakkelijk te leren. Er zijn veel details in dit eenvoudige programma die beter kunnen worden gepresenteerd via video.Om te beginnen, kweek de mesenchymale stamcellen tot meer dan 90% samenvloeiing. Vervang het kweekmedium in elk gerecht door 8 ml aangevulde alfa-MEM. Na 48 uur kweek moeten de cellen gelijkmatig worden gegroeid om het kweekmedium volledig te verzamelen in centrifugebuizen van 50 ml.Centrifugeer vervolgens het kweekmedium op 800 x g gedurende 10 minuten bij vier graden Celsius. Breng het supernatant over naar 1,5 ml conische buizen om de celfragmenten en cellulair puin te verwijderen. Centrifugeer het supernatant op 16.000 x g gedurende 30 minuten bij vier graden Celsius.Gebruik een pipet om de suspensie over te brengen naar ultracentrifugebuizen. Centrifugeer vervolgens op 150.000 x g gedurende twee uur bij vier graden Celsius. Resuspendeer de microvesicle pellet in 50 uL PBS per gerecht.Gooi het supernatant weg en verzamel het residu, dat exosomen zijn. Resuspendeer de exosomen uit zeven schotels van cellen in 50 uL PBS. Verdun in een buis van 15 ml 1 uL extracellulaire blaasjes in 1.499 uL PBS en meng.Start vervolgens de compatibele software voor de trackinganalyzer. Vul de monstercel met gedestilleerd water en laat het instrument vervolgens de celcontrole starten. Kalibreer het instrument met een voorbereide standaardoplossing.Zorg ervoor dat het deeltjesaantal dat wordt weergegeven op de softwaredetectie-interface tussen 50 en 400 ligt, bij voorkeur rond de 200, en klik op OK. Spoel vervolgens de monstercel af met 5 ml gedestilleerd water. Spoel vóór de monsteranalyse het kanaal met 1 ml gedestilleerd water.Zorg ervoor dat het deeltjesaantal dat wordt weergegeven op de softwaredetectie-interface kleiner is dan 10. In een conische buis van 1,5 ml resuspendeert u de extracellulaire blaasjes met 250 uL PBS. Bereid in een andere conische buis van 1,5 ml de werkoplossing van de PKH26-kleurstof.Meng de geresuspendeerde extracellulaire blaasjes met de werkoplossing. Laat het vijf minuten op kamertemperatuur staan en voeg vervolgens 500 uL EV-uitgeputte FBS toe om de reactie te stoppen. Voor microvesicles, centrifugeer op 16.000 x g gedurende 30 minuten bij vier graden Celsius.Gooi het supernatant weg. Voeg 1 ml PBS toe om het residu af te spoelen. Centrifugeer opnieuw en gooi het supernatant weg om de ongebonden kleurstof te verwijderen.Voor microvesicles, centrifugeer op 16.000 x g gedurende 30 minuten bij vier graden Celsius. Resuspensie van het residu met 200 uL PBS. Laat 20 uL suspensie op de dia vallen en observeer deze onder een fluorescentiemicroscoop.Resuspendeerde extracellulaire blaasjes in 200 uL PBS en meng vervolgens met heparine-oplossing met een volumeverhouding van 10: 1. Plaats de muis in het beeldvormingssysteem van de caudadale ader. Druk op de schakelaar om de hendel op te tillen.Voer met behulp van een staartaderillustrator een systematische injectie van de extracellulaire blaasjessuspensie door de staartader uit. Analyse van de deeltjesgrootteverdeling met behulp van NTA toonde aan dat de grootte van exosomen uit menselijke mesenchymale stamcellen varieerde van 40 tot 335 nm, met een piekgrootte van ongeveer 100 nm. Ook varieerde de grootte van microvesicles van 50 tot 445 nm, met een piekgrootte van 150 nm.Morfologische karakterisering van exosomen toonde aan dat ze een typische bekervorm vertoonden. Extracellulaire blaasjes werden efficiënt gelabeld door PKH26, wat werd waargenomen door de bruto weergave als gelabelde pellets, evenals door fluorescerende microscopie. De operator moet opletten tijdens het verzamelen van bovennatuurlijke middelen, omdat cellen gelijkmatig moeten worden geblazen om de vrijgekomen EV's die in de onduidelijke mesenchymale stamcellen worden vastgehouden, volledig te verzamelen.Om het succes van de injectie te verifiëren, kan in vivo beeldvorming van biodistributie van fluorescentie-gelabelde EV's of bevroren secties van specifieke weefselplaatsen van transplantatie worden uitgevoerd.
