過去10年間の直接電子検出器の出現1,2,3は、単一粒子クライオEM4,5,6を使用して解決された高分子および高分子集合体の高分解能構造の数が指数関数的に増加しました。ほとんどすべての精製高分子種は、サイズが~10 kDa以下の最小のタンパク質を除いて、クライオEMを使用した構造決定に適していると予想されます。グリッド調製および構造決定に必要な出発物質の量は、核磁気共鳴分光法およびX線結晶構造解析などの他の構造決定技術よりも少なくとも桁違いに少ない4,5,6。
