1. 食谱

</ol>2.同步C的制备。 秀丽隐杆线虫
<ol><li>使用标准方法26，在6厘米NGM板上培养动物（步骤1.1）。通过在LB培养基中散布0.2mL大 肠杆菌 OP50液体培养物（参见步骤1.9.）并在室温下孵育不超过24小时（旧细菌会影响预处理结果）来制备NGM板。 注意： 秀丽隐杆线虫的 学习和记忆对机械、化学和温度应力极其敏感。因此，强烈建议培养动物，保持包括水在内的所有试剂，并在17°C至20°C之间的室温下进行所有测定。 必须避免物理和机械刺激，如涡旋、粗移液和离心。新鲜的1-丙醇应最多每3个月使用一次，原因不明。重要的是，动物必须用大量的食物来饲养，因为饥饿会严重影响条件反射的结果。</li>
	<li>在第1天，用铂蠕虫采摘器将五只喂食良好的妊娠动物（放更多缓慢产卵的突变动物）放在四个6厘米NGM板上，并让它们在室温下产~50个卵3小时以获得同步的成年动物种群。通过用铂金蠕虫采摘器从盘子中取出亲本动物来停止产卵。 注意：种子板应保持在室温下，以尽量减少对动物的压力。</li>
	<li>在室温下培养动物约5天，这是动物达到成熟成年阶段所需的时间，而不是年轻成年阶段。 注意：4.5天至5.5天的培养期应根据条件进行调整，因为年轻的成年动物比成熟的成年动物对用于调理的化学物质更敏感（补充图1）。经过调理后，较年轻的成年动物可能表现出较低的趋化指数（CI）值。</li>
	<li>用1mL的0.25%明胶水溶液洗涤每个板（步骤1.4），在动物收集器中收集~200只成年动物（参见步骤1.4）。这种水性明胶可防止动物粘附在移液器吸头等塑料表面。</li>
	<li>用ddH 2 O（步骤1.8.）洗涤收集器中的动物，方法是在~10 mLddH2 O中非常轻轻地上下移动收集器2倍。 注意：细菌污染严重影响动物的趋化性。</li>
</ol>3. 短期联想学习和记忆的大规模训练
注意：有关大规模训练工作流，请参阅 图 1 。
<ol><li>将含有~200只动物的动物收集器轻轻浸入40mL的1%1-丙醇和HCl（与1-丙醇混合后pH 4.0;参见步骤1.7）的混合物中~1秒。 注意：对于对照动物，做同样的事情，但只浸入1%的1-丙醇水溶液中。最好只用HCl（pH 4.0）治疗动物作为另一种对照。</li>

</ol>5. 趋化性测定
<ol><li>在6cm塑料培养皿中制备琼脂平板用于趋化性测定（参见步骤1.5）。</li>
	<li>将动物转移到收集器（步骤2.5.）中，将其放置在塑料培养皿盖的平坦表面上，使用锯掉的移液器尖端与1mL0.25%水性明胶使用锯掉的移液管尖端具有>1mm（内径）开口。 注意：使用锯掉的移液器吸头以最大程度地减少对动物的剪切应力非常重要。</li>
	<li>在动物通过重力沉淀在管底部~1分钟后，从管中取出上清液（不要离心）。</li>
	<li>轻轻地将动物重悬于1mL趋化性测定缓冲液中（参见步骤1.6.），并让它们在重力作用下沉降到管底部~1分钟（不要离心）。通过移液尽可能多地去除上清液。</li>
	<li>同时，以相同的方式在两个地方对角点4 μL的5%水性1-丙醇，并在另外两个地方点4 μL的ddH2O，如图 3A所示。对于对1-丙醇敏感性较低的突变体的趋化性测定，发现较高浓度的1-丙醇水溶液，导致幼稚突变体的趋化指数（C.I.）值~0.6，如 补充表1所示。 注意：尽快完成发现程序很重要。点缀5%水溶液1-丙醇，因为1%水溶液1-丙醇太弱而无法在趋化性测定中吸引动物。相反，使用1%的1-丙醇水溶液进行预处理，因为用浓度高于1%的1-丙醇水溶液处理的动物显示出较低的CI值。</li>
	<li>在趋化性测定缓冲液（步骤5.4.）中点缀含有~60只动物的动物悬浮液的6μL部分，用于趋化性测定，使用锯掉的移液器吸头具有~1.0mm（内径）开口。用实验室组织芯尽可能多地去除液体，不要接触动物，并在盘子上盖上盖子。 注意：尽快完成这些过程非常重要。</li>
