사구체는 사구체 여과 장벽 1,2를 통과하는 큰 분자의 통과를 제한함으로써 혈액 여과에 중요합니다. 사구체는 정수리 상피 세포, 족 세포 (내장 상피 세포), 사구체 내피 세포 (GEC) 및 간질 세포3의 네 가지 세포 유형을 포함합니다. 사구체 내피는 큰 여과 부피에 필요한 fenestrae의 존재에 따라 독특한 혈관 구조를 특징으로합니다4. 사구체 내피의 정점 표면은 음전하를 띤 글리코칼릭스 층과 내피와 혈액 사이에 공간을 만드는 내피 표면층이라고 하는 코트로 덮여 있습니다. 이 구조는 알부민과 같은 음전하를 띤 분자의 통과를 제한하고 백혈구 및 혈소판 부착을 방지하는 높은 전하 선택성을 제공합니다5.
GEC는 당뇨병 환경과 관련된 고혈당증과 같은 대사 변화에 매우 민감합니다. 실제로 당뇨병은 유해 물질의 순환 증가, 포도당 대사 경로의 포화 및 세포 산화 환원 균형 방해로 이어집니다 3,6. 또한, 활성 산소 종의 증가는 내피 기능에 영향을 미치는 미토콘드리아 기능 장애를 유도합니다7.
현재 프로토콜의 전반적인 목표는 형광 미토콘드리아 특징을 가진 불멸화 된 사구체 내피 세포를 분리하는 것입니다. 실제로, 일차 GEC의 세포 배양은 제한된 증식 주기 및 초기 노화8를 갖는다. 또한, 형광 미토콘드리아의 존재는 고혈당증 또는 다른 치료에 대한 반응으로 핵분열 및 융합 사건을 검사하는 데 도움이됩니다. 대안으로 다른 실험실에서는 h-TERT를 사용하여 시험관 내9에서 세포를 불멸화했습니다.
여기에 설명 된 방법은 4-6 주 된 동물에서 조건부로 불멸화 된 mitoDendra2 사구체 내피 세포를 분리 할 수 있습니다 (그림 1). 이 상세한 프로토콜은 유인원 바이러스 40 대형 종양 항원 (SV40 TAg) 유전자10,11을 보유하고있는 트랜스 제닉 마우스 (H-2K b-tsA58)의 사용을 설명하여 열 안정성 조건부 불멸화 세포를 생성합니다. tsA58 TAg 유전자 생성물은 마우스 H-2Kb 유전자의 유도성 5' 플랭킹 프로모터의 제어 하에 33°C의 허용 온도에서 기능하며, 이는 인터페론 감마(IFNγ)에 노출시 기저 수준 이상으로 증가하여, 조건부 증식 표현형(12)을 유지한다. H-2Kb-tsA58 형질전환 마우스를 미토콘드리아 특이적(시토크롬 c 산화효소의 서브유닛 VIII) Dendra2-green을 발현하는 PhAM 마우스와 임의로 교차시켜 형광 미토콘드리아16의 실시간 검출을 가능하게 합니다. Dendra2 녹색 형광은 405nm 레이저16에 노출된 후 적색 형광으로 전환됩니다. 미토콘드리아가 광 전환 후 융합되면 녹색과 노란색 물질의 교환으로 노란색으로 나타나거나 핵분열 7,17을 겪을 때 빨간색으로 나타나는 길쭉한 모양을 형성합니다. mitoDendra2-GEC는 다양한 자극에 대한 GEC 미토콘드리아의 세포 반응을 연구하기위한 훌륭한 도구입니다.
