1. 动物制备

</ol>2. 术前护理（由外科助理进行）

</ol>3.内侧半月板手术不稳定，随后出现软骨划痕
<ol><li>调整显微镜以聚焦于髌韧带。</li>
	<li>用锯齿状镊子捏住膝盖侧面的皮肤（见 材料表），用手术剪刀平行于髌腱远端做一个小切口，引入剪刀并将切口扩大到约1厘米。将皮肤移到内侧，露出髌韧带和胫骨平台近端（图1）。</li>
	<li>使用11号刀片，沿着髌韧带的内侧，从韧带的顶部到底部切开一个切口（图1A）。当到达髌韧带底部时，将刀片旋转 90°，并将切口从髌韧带向内侧延伸，以进入关节囊。 注意：此过程或后续步骤可能会出血。如果发生出血，请使用无菌棉签并施加压力几秒钟（5至30秒）。</li>
	<li>用钝尖镊子捏住髌韧带，然后旋转手腕将髌韧带移动到外侧，刚好暴露髌下脂肪垫（IFP）。 注意：为尽量减少对髌韧带的损伤，请勿将镊子握得太紧，刚好足以将韧带保持在一侧。</li>
	<li>在仍然轻轻握住髌韧带的同时，用微镊子捏住IFP（见 材料表）将其抬高并稍微向上移动。这允许可视化内侧半月板韧带。</li>
	<li>识别内侧半月板的内侧半月板韧带（MMTL），它将内侧半月板的颅角固定在胫骨前平台（图1B）。</li>
	<li>避免胫骨平台或股骨髁损伤和长时间接触软骨。</li>
	<li>用 2 毫米刀片 Vannas 弹簧剪刀小心地切断 MMTL，保持内侧半月板和其他韧带完好无损。此时，DMM模型的外科手术已完成（图1C）。</li>

	</ol></li>
	<li>用两个或三个小的7毫米伤口闭合金属夹或可吸收的6-0皮下手术缝合线关闭皮肤（见 材料表）。 注意：皮下手术缝合线更好，因为它们避免了进一步的干预，但它们延长了手术时间。外部缝合线增加了小鼠啃咬伤口的风险。</li>
	<li>识别内侧半月板的内侧半月板韧带以进行假手术，但不要切断。</li>
	<li>对于仅接受软骨划痕的小鼠，在不切断韧带的情况下进行三个浅表划痕。 注意：在每只鼠标之间，更换手套并通过高压灭菌器 对 仪器进行消毒。记得在重复使用前检查仪器是否已冷却。</li>
</ol>4. 术后护理
<ol><li>如果发生出血（>50uL），皮下注射500μL温热的无菌盐水（在小鼠背部）。 注意：根据我们的经验，虽然小鼠有轻微的出血，但它永远不会超过一小滴，因此不需要补充液体。</li>
	<li>手术后，将小鼠放在干净的纸巾上的恢复笼中，并从麻醉中恢复（5-10分钟）。</li>
	<li>手术后将完全清醒的小鼠转移到带有新鲜床上用品的干净笼子中。</li>
	<li>在手术干预后72小时内，监测任何疼痛或痛苦的迹象。注意：<ol><li>体重变化。虽然体重可能会在第一天和第二天下降，但这通常不超过术前体重的5%。</li>
		<li>切口周围普遍缺乏梳理或过度梳理。</li>

</ol>5.骨关节炎疾病的评估

MMTL非常靠近胫骨髁，因此在将弯曲弹簧剪刀的下刀片放置在MMTL下方时，必须注意不要伤害软骨。弯曲的叶片应指向内侧，略微向上，平行于髁突。为了最好地切割MMTL，请确保剪刀锋利。检查半月板在切断韧带后是否可以向内侧移动，因为有时会留下需要进一步切割的小附件。当引入微刀片刮擦髁突时，它必须垂直于髁突。使第一次划痕靠近关节中间，但注意不要损坏前交叉韧带。然后向内侧移动，然后在半月板后面移动。划痕可能在软骨上显示为微弱的白线。因为我们通常使用夹子，所以初始切口是在侧面进行的，因此在闭合伤口后将夹子定位在腿的一侧。这样可以避免夹子在鼠标恢复运动时摩擦膝盖。使用缝合线时，强烈建议使用皮下缝合。如果使用外部缝线，小鼠很可能会啃缝线并打开伤口，这将增加感染的机会。如果做得好，这种手术从切口到伤口闭合的时间不得超过5-10分钟，从而最大限度地减少软骨的暴露和可能发生的任何其他不受控制的损伤。手术后，小鼠恢复得非常快，几乎可以立即爬进笼子并正常走动。如果小鼠不活跃，应咨询该单位的适当专家。
