Periaortisk anvendelse af CaPO4 er en robust tilgang til at inducere AAA hos mus. En abdominal aortaaneurisme (AAA) er en dødelig hjerte-kar-sygdom karakteriseret ved permanent udvidelse af abdominal aorta, med høj dødelighed, når brud opstår. AAA er forbundet med aldring, rygning, mandligt køn, hypertension og hyperlipidæmi1. Flere patologiske processer har vist sig at bidrage til AAA-dannelse, herunder ekstracellulær matrixfiberproteolyse, immuncelleinfiltration og tab af vaskulære glatte muskelceller. I øjeblikket forbliver de patologiske mekanismer i AAA undvigende, og der er ingen dokumenterede lægemidler til behandling af AAA1. Forskning i human AAA er begrænset på grund af eksistensen af få humane aortaprøver; således er flere kemiske modifikationsinducerede AAA-modeller for dyr blevet etableret og bredt vedtaget, herunder subkutan angiotensin II (AngII) infusion, perivaskulær eller intraluminal elastaseinkubation og perivaskulær calciumphosphatanvendelse2. En almindeligt anvendt musemodel er anvendelsen af calciumphosphat (CaPO4) til adventitia af den infrarenale abdominale aorta, hvilket er omkostningseffektivt og ikke kræver genetisk modifikation.
Direkte periaortisk anvendelse af CaCl2 på halspulsåren hos kaniner for at fremkalde aneurismeændring blev oprindeligt rapporteret af Gertz et al.3 og blev senere anvendt på abdominal aortas hos mus. Modellen blev udviklet af Yamanouchi et al. for at fremskynde aortaudvidelse ved hjælp af CaPO 4-krystaller i mus4. Infiltration af CaPO4 i mus aortas rekapitulerer mange patologiske træk observeret i humane AAA'er, herunder dyb makrofaginfiltration, ekstracellulær matrixnedbrydning og calciumaflejring. Risikofaktorerne for human AAA, såsom hyperlipidæmi, øger også CaPO 4-induceret AAA hos mus5. I modsætning til AngII perfusionsinduceret AAA i ApoE-/- eller LDLR-/- mus forekommer CaPO 4-induceret AAA i det infrarenale aortaområde, som efterligner human AAA. En abdominal aortaaneurisme (AAA) er en livstruende hjerte-kar-sygdom, der forekommer over hele verden og er karakteriseret ved irreversibel udvidelse af abdominal aorta. I øjeblikket anvendes flere kemisk inducerede murine AAA-modeller, der hver simulerer et andet aspekt af patogenesen af AAA. Den calciumphosphatinducerede AAA-model er en hurtig og omkostningseffektiv model sammenlignet med angiotensin II- og elastaseinducerede AAA-modeller. 14 dage efter CaPO4-påføring blev C57BL/6J-hanmusene aflivet, og deres aortaer blev høstet og renset. Aortaernes morfologi blev afbildet for at visualisere AAA-dannelsen. Denne protokol beskriver calciumphosphatinduceret abdominal aortaaneurisme triple A musemodel for at studere de patologiske træk i de molekylære mekanismer i abdominal aortaaneurisme. Den calciumphosphatinducerede abdominale aortaaneurismemodel er en hurtig og omkostningseffektiv model sammenlignet med angiotensinrøret og modellerne for elastaseinduceret abdominal aortaaneurisme. Demonstrerer proceduren vil være Zeyu Cai, postlæge her, en fyr fra vores laboratorium.For at begynde at skære 0,3 centimeter brede strimler af pulverfri gummihandsker og gasbind. Køb derefter otte til ti uger gamle C57BL6J Mayo-mus, og hus dyrene i et klimatiseret miljø med en 12 timers lys mørk cyklus og fri adgang til mad og vand. Overdådig musene med PBS-buffer gennem hjertets venstre ventrikel for at fjerne blod i aorta og derefter perfuse med 4% paraformaldehyd som beskrevet i manuskriptet. Fjern utilsigtet væv og fedt forsigtigt under et stereomikroskop.Høst derefter aorta og læg den i et rør indeholdende fem milliliter 4% paraformaldehyd i 48 timer. Fastgør derefter aorta på en sort voksplade med insektnåle og fortsæt med at erhverve aortabilleder.
