このプロトコルは、キナーゼの分子サイズのリン酸化を直接評価することにより、KCC2の制御に関与するメカニズムを理解するのに役立ち、その生理学的機能と活性に光を当てます。タンパク質発現が非常に微量であっても、関連分子の複雑な混合物から目的のタンパク質などの分子を同定し、特性評価するために使用できます。この技術は、複雑なシステムのタンパク質分析と、異常な組織機能や疾患の根底にある潜在的なメカニズムを特定するために不可欠なツールです。手順を実演するのは、私の研究室の博士課程の学生であるサンデーソロモンジョサイアです。摂氏37度のビーズバスですべての試薬を事前に武装させ、安定に切断されたラットKCC2bヒト胚性腎臓293細胞をビーズバスで完全に解凍することから始めます。次に、細胞をクライオバイアルチューブから5ミリリットルの新鮮な培地を含む遠心分離管に移し、細胞を1, 200 Gで3〜5分間遠心分離します。真空ポンプに固定したアスピレーターピペットを用いて上清を吸引した後、細胞を10ミリリットルの新鮮な培地に再懸濁する。懸濁液を10センチメートルの皿プレートに移し、摂氏37度の温度と5%の二酸化炭素を有するインキュベーター内で48時間増殖させる。細胞が90%コンフルエントに達したら、古い培地を吸引し、2ミリリットルのPBSですすぐことによって細胞を分割します。2ミリリットルのトリプシンを加え、室温で約1〜2分間インキュベートします。2ミリリットルの完全培地を使用してトリプシン処理細胞を洗浄します。新しい皿に9ミリリットルの新鮮な培地を加えます。次に、古い皿から新しい皿に1ミリリットルの溶液を加えます。スプリットセルディッシュをインキュベーターに48時間移し、90%以上のコンフルエンシーを達成します。細胞をジメチルスルホキシド、8マイクロモルのスタウロスポリン、または0.5ミリモルのN-エチルマレイミドで15分間処理してから、溶解バッファーで回収します。トランスセクトした培養皿から培地を吸引し、培養皿を氷上に置き、氷冷PBSで細胞を洗浄します。PBSを吸引し、1.0ミリリットルの氷冷溶解バッファーを皿に加えます。冷たいプラスチックセルスクレーパーを使用して皿の底から細胞をこすった後、細胞懸濁液を氷上に置いた微量遠心チューブに移し、続いて摂氏4度で30分間絶えず攪拌した。細胞溶解物を冷たい遠心分離機で遠心分離した後、氷の上に置きます。上清を予冷した新鮮なチューブに集め、ペレットを廃棄します。300マイクロリットルのプロテインGセファロースをマイクロ遠心チューブにピペットで入れます。次に、溶液を500 Gで2分間遠心分離します。上清を捨てた後、500マイクロリットルのPBSとボルテックスをよく加えます。遠心分離後の上清を捨て、工程を繰り返す。次に、1ミリグラムの抗KCC2スレオニン906および抗KCC2スレオニン1007抗体を200マイクロリットルのプロテインGセファロースビーズと混合してから、PBSで容量を500マイクロリットルにします。振動するプラットフォームまたは回転ホイールで摂氏4度で2時間振った後、PBSで2回の洗浄を繰り返します。細胞ライセートでタンパク質定量を行い、洗浄したビーズに1ミリグラムの細胞ライセートを加え、スピンダウンする前にエンドツーエンドのローテーターでインキュベートします。150ミリモルの塩化ナトリウムを含むPBSで3回洗浄し、続いて200マイクロリットルのPBSで3回洗浄してから、最終ペレットを100マイクロリットルのLDSサンプルバッファーに再懸濁します。ローターシェーカーでチューブを室温で5分間振とうします。それらを加熱ブロックでインキュベートし、遠心分離してから、上清をゲルローディングに使用します。ウェスタンブロットキャスティング装置を組み立て、準備したばかりの8%分離ゲルを注ぎ、キャスティンググラスの上部から約2センチメートルのスペースを確保してゲルをキャストします。200マイクロリットルの絶対イソプロパノールをセットアップに追加し、室温で60分間放置します。ピペットを使用してイソプロパノールを除去し、約200マイクロリットルの蒸留水でゲルを注意深くすすいでください。新しく調製した6%スタッキングゲルをキャスティングセットアップに加えた後、ウェルコームにそっとはめ込み、室温で30分間放置します。次に、キャストしたゲルを電気泳動タンクに固定します。ランニングバッファーをタンクに注いだ後、5マイクロリットルの分子量マーカーを最初のウェルにロードし、等量のタンパク質をSDS-PAGEゲルの各ウェルにロードします。空のウェルをLDSで満たし、120ボルトで約90〜120分間ゲルを実行します。ニトロセルロースメンブレンを20%メタノールを含むトランスファーバッファーで再水和します。ゲルとメンブレンをトランスファーバッファーですすぎ、準備スタックに静かに広げます。負極、サンドイッチフォーム、ろ紙すすぎSDS-PAGEゲル、すすぎ硝化硝化膜、ろ紙、サンドイッチフォーム、正極の順に転写するサンドイッチを配置します。完了したら、組み立てたサンドイッチをトランスファータンクに積み重ね、90ボルトで90分間、または30ボルトで360分間運転します。ブロッキングバッファーを使用して室温で1時間乾燥膜をブロッキングします。一次抗体とβ-アクチンをブロッキングバッファーで適切な希釈液でメンブレンを室温で1時間、または摂氏4度で一晩インキュベートします。次に、TBSTをそれぞれ5分間3回洗浄します。洗浄したメンブレンをブロッキングバッファーで希釈した二次抗体で60分間インキュベートし、TBSTの洗浄を3回繰り返します。完了したら、洗浄したメンブレンをイメージングボードに置きます。シグナルを発生させるには、イメージングボードをイメージングシステムに移してイメージングする前に、各増強化学発光試薬を等量の混合した溶液をメンブレン上に広げてイメージングします。HEK293細胞をスタウロスポリンとN-エチルマレイミドまたはNEMで処理すると、SPAK標的部位、Tループキナーゼドメインに位置するスレオニン233、および内因性発現SPAKのSループリン酸化部位セリン373でリン酸化が低下しました。スタウロスポリンは、KCC2bを安定に発現するHEK294細胞においてセリン940のリン酸化を低下させたが、NEMはそのリン酸化を有意に増加させた。さらに、スタウロスポリンはスレオニン505部位でのリン酸化を減少させるが、NEMは同じ部位でのリン酸化のわずかな増加を引き起こした。スレオニン505部位のプロテインキナーゼCとセリン940部位のKCC2bのリン酸化に対する両化合物の異なる効果はよく相関しています。代表的な結果はまた、NEMが総KCC2量の発現の有意な増加を引き起こしたのに対し、両方の化合物で処理した場合、総NKCC1およびSPAKの発現は有意に変化しないことを示しました。さらに、2つの化合物はスレオニン233およびセリン373部位でSPAKのリン酸化の低下を引き起こし、これはスレオニン906/1007およびスレオニン203/207/212およびNKCC1の内在的に発現する短縮リン酸化と相関した。さらに、スタウロスポリンおよびNEMは、それぞれセリン940部位のプロテインキナーゼCのリン酸化を減少および増加させ、KCC2bを安定に発現し、スタウロスポリンおよびNEM処理時にそれぞれスレオニン505部位のプロテインキナーゼCデルタのリン酸化の減少および増加と相関した。不適切な一次抗体または二次抗体を使用した場合、イメージング中にバンドは見られません。また、低濃度の抗体を使用するとシグナルが見えない場合があります。この技術は、タンパク質の定量分析に関連するツールであり、効果的な早期診断ツールを含む、多くの科学的およびリソース目的での使用を可能にしています。
