ニワトリ胚の聴覚脳幹は、回路およびシナプスの発生におけるFMRP調節の詳細な機能解析のための独自の有利なモデルを提供します。ニワトリの胚の脳に簡単にアクセスできることと、遺伝子操作のための確立された卵エレクトロポレーション技術は、初期の胚期における脳の発達の理解に大きく貢献しています。最近発表された研究では、この技術は、FMRPの誤発現の時間的制御を可能にする高度な分子ツールと組み合わされました20,21。ここでは、シナプス前ニューロンとシナプス後ニューロンの選択的操作を別々に誘導する方法論が進められています。この方法は、聴覚脳幹回路で開発されました。音響信号は、聴覚内耳の有毛細胞によって検出され、聴覚神経節(AG、哺乳類ではらせん神経節とも呼ばれます)に伝達されます。AGの双極性ニューロンは、末梢突起で有毛細胞を神経支配し、次に中枢投影(聴神経)を脳幹に送り、そこで2つの一次蝸牛核、マグノセルラリス核(NM)と角核(NA)で終わります。NMのニューロンは、構造的および機能的に、哺乳類の前腹側蝸牛核の球状のふさふさした細胞に匹敵します。NM内では、聴神経線維(ANF)は、Held端子22の大きな終末球を介してNMニューロンの体細胞上にシナプスする。発生中、NMニューロンは後脳23の菱形5および6(r5/6)から生じ、AGニューロンは耳嚢胞24に存在する神経芽細胞に由来する。ここでは、シナプス前AGニューロンとシナプス後NMニューロンにおけるFMRP発現を選択的にノックダウンする手順を別々に記載する。
