Als potentieel vaccinplatform hebben buitenmembraanblaasjes (OMV's) de afgelopen jaren steeds meer aandacht getrokken 1,2. OMV's, voornamelijk van nature uitgescheiden door gramnegatieve bacteriën3, zijn bolvormige deeltjes op nanoschaal die bestaan uit een lipide bilayer, meestal in de grootte van 20-300 nm4. OMV's bevatten verschillende ouderlijke bacteriële componenten, waaronder bacteriële antigenen en pathogeen-geassocieerde moleculaire patronen (PAMPs), die dienen als solide immuunpotentiators5. Profiterend van hun unieke componenten, natuurlijke blaasjesstructuur en geweldige genetische manipulatiemodificatielocaties, zijn OMV's ontwikkeld voor gebruik op vele biomedische gebieden, waaronder bacteriële vaccins6, adjuvantia7, kankerimmunotherapiegeneesmiddelen8, medicijnafgiftevectoren9 en antibacteriële kleefstoffen10.
De SARS-CoV-2-pandemie, die zich sinds 2020 wereldwijd heeft verspreid, heeft een zware tol geëist van de wereldwijde samenleving. Het receptorbindende domein (RBD) in spike-eiwit (S-eiwit) kan binden met humaan angiotensine-converterend enzym 2 (ACE2), dat vervolgens de binnenkomst van het virus in de cel bemiddelt 11,12,13. RBD lijkt dus een belangrijk doelwit te zijn voor vaccinontdekking 14,15,16. Monomere RBD is echter slecht immunogeen en het kleine molecuulgewicht maakt het moeilijk voor het immuunsysteem om te herkennen, dus adjuvantia zijn vaak nodig17.
Om de immunogeniciteit van RBD te verhogen, werden OMV's met polyvalente RBD's geconstrueerd. Bestaande studies die OMV gebruiken om RBD weer te geven, fuseren RBD meestal met OMV om tot expressie te komen in bacteriën18. RBD is echter een van het virus afgeleid eiwit en prokaryote expressie heeft waarschijnlijk invloed op de activiteit ervan. Om dit probleem op te lossen, werd het SpyTag (ST)/SpyCatcher (SC) systeem, afgeleid van Streptococcus pyogenes, gebruikt om een covalent isopeptide met OMV en RBD te vormen in een conventioneel buffersysteem19. Het SC-domein werd uitgedrukt met Cytolysine A (ClyA) als een fusie-eiwit door bio-engineered Escherichia coli, en ST werd uitgedrukt met RBD via het HEK293F cellulaire expressiesysteem. OMV-SC en RBD-ST werden 's nachts gemengd en geïncubeerd. Na zuivering door ultracentrifugatie of grootte-uitsluitingschromatografie (SEC) werd OMV-RBD verkregen.

a "plug-and-display" nanoparticle vaccine platform based on outer membrane vesicles displaying sars-cov-2 receptor-binding domain

Biomimetische nanodeeltjes verkregen uit bacteriën of virussen hebben aanzienlijke belangstelling getrokken voor vaccinonderzoek en -ontwikkeling. Buitenmembraanblaasjes (OMV's) worden voornamelijk uitgescheiden door gramnegatieve bacteriën tijdens de gemiddelde groei, met een nanodiameter en zelfadjuvante activiteit, wat ideaal kan zijn voor de levering van vaccins. OMV's hebben gefunctioneerd als een veelzijdig afgiftesysteem voor eiwitten, nucleïnezuren en kleine moleculen. Om ten volle te profiteren van de biologische kenmerken van OMV's, werden bio-technische Escherichia coli-afgeleide OMV's gebruikt als drager en SARS-CoV-2 receptorbindingsdomein (RBD) als een antigeen om een "Plug-and-Display" vaccinplatform te bouwen. De SpyCatcher (SC) en SpyTag (ST) domeinen in Streptococcus pyogenes werden toegepast om OMV's en RBD te conjugeren. Het Cytolysine A (ClyA) gen werd vertaald met het SC gen als een fusie-eiwit na plasmide transfectie, waardoor een reactieve plaats op het oppervlak van de OMV's achterbleef. Na het 's nachts mengen van RBD-ST in een conventioneel buffersysteem werd een covalente binding gevormd tussen de OMV's en RBD. Zo werd een multivalent omv-vaccin bereikt. Door te vervangen door diverse antigenen, kan het OMV-vaccinplatform efficiënt een verscheidenheid aan heterogene antigenen weergeven, waardoor epidemieën van infectieziekten mogelijk snel worden voorkomen. Dit protocol beschrijft een nauwkeurige methode voor het bouwen van het OMV-vaccinplatform, inclusief productie, zuivering, bioconjugatie en karakterisering.

Het stroomdiagram voor dit protocol is weergegeven in figuur 1. Dit protocol zou een algemene benadering kunnen zijn voor het gebruik van OMV's als vaccinplatform; men hoeft alleen de juiste expressiesystemen te kiezen op basis van het type antigenen.

Voeg het mengsel toe aan 280 milliliter celkweekmedium en kweek met 130 rotaties per minuut bij 37 graden Celsius gedurende vijf dagen. Voer vervolgens RBD spytag-zuivering uit.Begin met centri fuseren van de cellen op 6.000 keer G bij 25 graden Celsius gedurende 20 minuten en gebruik een pipet om het supernatant te verzamelen. Vul de kolom met twee milliliter nikkel NTA agarose en was deze drie keer met PBS. Voeg verdunde eiwitoplossing toe en meng met BCA-werkoplossing. Incubeer op 37 graden gedurende 30 minuten.Meet de absorptie op 562 nanometer van elke put en bereken de eiwitconcentratie uit de standaardcurve. Om bioconjugatie van OMV spycatcher en RBD spy tag uit te voeren, mengt u OMV spycatcher en RBD spytag en PBS in een verhouding van 40 op één. Verticaal draaien om het mengsel een nacht te mengen met 15 rotaties per minuut bij vier graden Celsius.Voer zuivering van OMV RBD uit met behulp van PBS-oplossing met behulp van een procedure die vergelijkbaar is met OMV spycatcher-zuivering. Het plasmaontwerpschema laat zien dat het spycatcher-gen via flexibele linker verbonden is met het cly A-gen, terwijl de spytag verbinding maakt met de vijf priemterminal van het RBD-gen met een hys tag-gen voor zuivering en verificatie. Na ultracentrifugatie bleven bijna alle niet-gereageerde RBD-spytags in het supernatant achter.De tweede ultracentrifugatie leverde geen verder significant voordeel op ten opzichte van de eerste keer. De gemiddelde hydrodynamische diameter van OMV spycatcher was 133 nanometer terwijl het 152,6 nanometer was voor OMV-RBD. Deze verschillen kunnen zijn omdat RBD de OMV-deeltjesgrootte verhoogt na intensieve weergave.De resultaten van TEM komen overeen met de deeltjesgrootteverdeling die wordt verkregen uit de DLS-resultaten. Rotatie voor het mengen van het mengsel is erg belangrijk. De efficiëntie van de reactie kan veel lager zijn zonder rotatie.
