En tant que plate-forme vaccinale potentielle, les vésicules de membrane externe (OMV) ont attiré de plus en plus d’attention ces dernières années 1,2. Les OMV, principalement sécrétées naturellement par les bactéries à Gram négatif3, sont des particules sphériques à l’échelle nanométrique composées d’une bicouche lipidique, généralement de la taille de 20-300nm4. Les OMV contiennent divers composants bactériens parentaux, y compris des antigènes bactériens et des profils moléculaires associés à des agents pathogènes (PAMP), qui servent de potentialisateurs immunitaires solides5. Bénéficiant de leurs composants uniques, de la structure naturelle des vésicules et des grands sites de modification du génie génétique, les OMV ont été développés pour être utilisés dans de nombreux domaines biomédicaux, notamment les vaccins bactériens6, les adjuvants7, les médicaments d’immunothérapieanticancéreuse 8, les vecteurs d’administration de médicaments9 et les adhésifs antibactériens10.
La pandémie de SRAS-CoV-2, qui s’est propagée dans le monde entier depuis 2020, a fait payer un lourd tribut à la société mondiale. Le domaine de liaison au récepteur (RBD) dans la protéine de pointe (protéine S) peut se lier à l’enzyme 2 de conversion de l’angiotensine humaine (ACE2), qui intervient ensuite dans l’entrée du virus dans la cellule11,12,13. Ainsi, le TCSP semble être une cible de choix pour la découverte de vaccins14,15,16. Cependant, le TCSP monomère est peu immunogène et son faible poids moléculaire le rend difficile à reconnaître par le système immunitaire, de sorte que des adjuvants sont souvent nécessaires17.
Afin d’augmenter l’immunogénicité des TCSP, des OMV présentant des RBD polyvalents ont été construits. Les études existantes utilisant OMV pour afficher RBD fusionnent généralement RBD avec OMV pour être exprimé dans les bactéries18. Cependant, le TCSP est une protéine dérivée d’un virus, et l’expression procaryote est susceptible d’affecter son activité. Pour résoudre ce problème, le système SpyTag (ST)/SpyCatcher (SC), dérivé de Streptococcus pyogenes, a été utilisé pour former un isopeptide covalent avec OMV et RBD dans un système tampon conventionnel19. Le domaine SC a été exprimé avec la cytolysine A (ClyA) en tant que protéine de fusion par Escherichia coli issu de la bio-ingénierie, et ST a été exprimé avec RBD via le système d’expression cellulaire HEK293F. OMV-SC et RBD-ST ont été mélangés et incubés pendant la nuit. Après purification par ultracentrifugation ou chromatographie d’exclusion de taille (SEC), OMV-RBD a été obtenu.

a "plug-and-display" nanoparticle vaccine platform based on outer membrane vesicles displaying sars-cov-2 receptor-binding domain

Les nanoparticules biomimétiques obtenues à partir de bactéries ou de virus ont suscité un intérêt considérable dans la recherche et le développement de vaccins. Les vésicules de la membrane externe (OMV) sont principalement sécrétées par les bactéries à Gram négatif pendant la croissance moyenne, avec un diamètre nanométrique et une activité auto-adjuvante, ce qui peut être idéal pour l’administration du vaccin. Les OMV ont fonctionné comme un système d’administration à multiples facettes pour les protéines, les acides nucléiques et les petites molécules. Pour tirer pleinement parti des caractéristiques biologiques des OMV, les OMV dérivés d’Escherichia coli issus du bio-ingénierie ont été utilisés comme porteur et le domaine de liaison au récepteur du SARS-CoV-2 (RBD) comme antigène pour construire une plate-forme vaccinale « Plug-and-Display ». Les domaines SpyCatcher (SC) et SpyTag (ST) de Streptococcus pyogenes ont été appliqués aux OMV et RBD conjugués. Le gène de la cytolysine A (ClyA) a été traduit avec le gène SC en protéine de fusion après transfection plasmidique, laissant un site réactif à la surface des OMV. Après avoir mélangé RBD-ST dans un système tampon conventionnel pendant la nuit, une liaison covalente s’est formée entre les OMV et les RBD. Ainsi, un vaccin multivalent OMV a été obtenu. En les remplaçant par divers antigènes, la plateforme vaccinale OMVs peut afficher efficacement une variété d’antigènes hétérogènes, prévenant ainsi potentiellement rapidement les épidémies de maladies infectieuses. Ce protocole décrit une méthode précise pour construire la plateforme vaccinale OMV, y compris la production, la purification, la bioconjugaison et la caractérisation.

L’organigramme de ce protocole est illustré à la figure 1. Ce protocole pourrait être une approche générale de l’utilisation des OMV comme plate-forme vaccinale; Il suffit de choisir les systèmes d’expression appropriés en fonction du type d’antigènes.

Incuber la solution bactérienne dans un milieu de deux litres et cultiver à 220 rotations par minute à 37 degrés Celsius pendant cinq heures jusqu’à ce que le stade de croissance logarithmique soit atteint.Ajouter l’isopropyl bêta D-IPTG pour obtenir la concentration finale de la solution bactérienne à 0,5 millimolaire, puis la culture pendant la nuit à 220 rotations par minute à 25 degrés Celsius. Ajouter le mélange à 280 millilitres de milieu de culture cellulaire et cultiver à 130 rotations par minute à 37 degrés Celsius pendant cinq jours. Ensuite, effectuez la purification du spytag RBD.Commencez par centri fusionnant les cellules à 6 000 fois G à 25 degrés Celsius pendant 20 minutes et utilisez une pipette pour recueillir le surnageant. Remplissez la colonne avec deux millilitres d’agarose NTA de nickel et lavez-la trois fois avec du PBS. Ajouter l’imidazole dans le surnageant cellulaire pour obtenir la concentration finale de 20 millimolaires, et charger le surnageant cellulaire.Ajouter trois volumes de colonne de PBS contenant 20 millimolaires d’imidazole pour le lavage et recueillir la fraction de lavage. Pour déterminer la concentration en protéines par la méthode BCA, diluez en série la solution de protéine BSA standard de deux à 0,0625 milligramme par millilitre, diluez 10 fois le spycatcher OMV purifié et l’étiquette espion RBD.Ensuite, mélangez les solutions de travail BCA A et B dans un rapport de 50 pour un. Ajouter la solution de protéines diluées et mélanger avec la solution de travail BCA. Incuber à 37 degrés pendant 30 minutes.Mesurez l’absorbance à 562 nanomètres de chaque puits et calculez la concentration en protéines à partir de la courbe standard. Pour effectuer la bioconjugaison d’OMV spycatcher et RBD spy tag, mélangez OMV spycatcher et RBD spytag et PBS à un ratio de 40 pour un. Tourner verticalement pour mélanger le mélange pendant la nuit à 15 rotations par minute à quatre degrés Celsius.Effectuer la purification d’OMV RBD à l’aide d’une solution PBS en utilisant une procédure similaire à la purification OMV spycatcher. Le schéma de conception du plasma montre que le gène spycatcher est connecté au gène cly A via un linker flexible tandis que le spytag se connecte aux cinq terminaux premiers du gène RBD avec un gène hys tag pour la purification et la vérification. Après l’ultracentrifugation, presque tout le spytag RBD n’ayant pas réagi est resté dans le surnageant.La deuxième ultracentrifugation n’a pas fourni d’autre avantage significatif par rapport à la première fois. Le diamètre hydrodynamique moyen z de l’OMV spycatcher était de 133 nanomètres alors qu’il était de 152,6 nanomètres pour OMV-RBD. Ces différences peuvent être dues au fait que le RBD augmente la taille des particules OMV après un affichage intensif.Les résultats de la TEM concordent avec la distribution granulométrique obtenue à partir des résultats de la DLS. La rotation pour mélanger le mélange est très importante. L’efficacité de la réaction peut être beaucoup plus faible sans rotation.
