该协议提供有关载体系统如何与细胞相互作用的定量数据。定量数据是工程和优化的关键。它允许我们从是或否的生物学问题转向多少问题。该技术可用于将临床前药物载体性能的结果（定性）转换为定量结果。即使您在与表征细胞不同的系统上表征载体，这也有效。可能需要几次迭代才能找到落在纳米侧线性或定量范围内的最佳载流子稀释度。花点时间寻找合适的条件。首先，将流通池安装到激光模块上。用一毫升蒸馏水以每秒0.1毫升的速度缓慢冲洗流通池。如果流通池内形成气泡，请部分缩回悬浮液以将气泡与气液界面合并，然后再继续。然后将整个激光模块锁定在仪器内部。大约在刷新中途启动相机。请务必确认载体碎屑是否已冲洗掉。选择拍摄以打开拍摄设置选项卡，然后单击启动相机。用一毫升空气驱动系统。如果屏幕上可见任何静电载体，请按照制造商的说明清洁流通池。根据所涉及的载体系统，确保载体通过超声或涡旋很好地悬浮，为纳米颗粒跟踪分析做好准备。在水中稀释载体，以制备至少0.6至1毫升的每个样品，载体浓度在每毫升1x10至第七个和1x10至第九个载流器之间。取出激光模块并将其直立放置。将第一个载体样品放入一毫升注射器中，并将注射器连接到管入口。然后小心地将样品装入流通池中。如果在流通池内形成气泡，请部分缩回悬浮液以将气泡与气液界面合并，然后再继续。确保整个流通池充满液体，然后暂停加载。如果需要，调整相机对焦以可视化各个载体。使用仪器右侧的旋转旋钮进行粗焦调整。通过选择硬件选项卡进行更精细的调整。通过调整焦点滑块来更改焦点。为确保没有过饱和，请通过调整捕获选项卡中的滑块来选择最佳相机级别。如果仪器配备了此附件，请将装有载体样品的注射器装入注射泵中。在 SOP 选项卡下，选择标准测量以拍摄五次，每次 30 秒。输入基本文件名，如果需要，通过单击高级按钮添加其他示例信息，该按钮将打开包含各种选项的模式对话框。按创建并运行脚本，然后等待出现一个弹出窗口，要求请提前采样。如果使用注射泵，请选择硬件选项卡，然后选择注射泵选项卡。设置所需的输注速率并按输注。如果不使用注射泵，请手动推进样品。在弹出窗口中，选择“确定”开始捕获。在五次捕获中的每一次捕获后，当请高级样品弹出窗口再次出现时，检查样品是否仍在流通池中移动。然后选择“确定”以继续下一次捕获。在“过程”选项卡中，在 4 到 8 之间调整检测阈值滑块，以正确识别屏幕上可见的不同载体。您可以调整屏幕增益以帮助可视化，它不会影响下游分析。在弹出窗口中，按确定启动跟踪分析。通过单击单个分析选项卡中的分析来监视分析进度。分析完成后，查找要显示的导出设置提示。确认已选择“包括 PDF”和“包括实验摘要”。根据需要选择任何其他导出格式。在PDF数据导出的结果部分中，为确保测量的浓度可靠，请验证测量的载体浓度是否在每毫升1x10至第七个载流子和1x10至第九个载流子之间，并检查浓度测量结果下方是否有任何错误消息或警告消息。使用原液中的不同稀释度重复该过程两次或更多次，并确保每个样品的浓度在仪器的线性范围内。按照文本手稿中的说明计算库存载体浓度。按照文本手稿中的说明制备游离或抗体偶联荧光染料溶液。根据浓度、分子量和阿伏伽德罗数计算储备溶液的浓度，如公式所示。然后在载体稀释剂中对染料进行连续稀释，以生成标准曲线样品。然后将载体样品添加到测量板中，并使用酶标仪测量荧光。按照文本手稿中的说明生成标准曲线。通过将体积荧光除以载体浓度来计算每个载体的绝对荧光强度。通过确定相关通道中的最佳PMT电压设置，为最终的载体细胞实验设置流式细胞仪。运行阴性对照样品，其中细胞未与载体孵育以确定背景荧光。制备并重新悬浮流式细胞术定量微球。使用与细胞样品相同的缓冲液。如果单独提供珠群，请将它们拉到一起。运行流式细胞术定量微球和载体细胞样品以确定每个细胞的荧光强度。运行流式细胞仪定量珠以生成标准曲线，将绝对荧光强度转换为MFI。使用它来计算文本手稿中所述的载体的理论MFI。运行细胞载体样品以确定每个样品的每个细胞的荧光强度。最后，通过从样品的测量荧光中减去背景荧光，然后除以每个颗粒的荧光来计算每个样品的每个细胞的载体数。对于633纳米聚甲基丙烯酸核壳颗粒，每个载体的浓度和荧光强度可以使用细胞仪流直接定量。相比之下，100纳米超顺磁性氧化铁纳米颗粒太小，无法单独检测，并使用大流进行分析。标记的定量微球在流式细胞仪上生成标准曲线。测量的MFI与定量微球的MESF值之间的相关性是线性的，并且在测量日期之间大致相似。使用荧光标记的HeLa细胞和235纳米聚甲基丙烯酸胶囊从0到24小时的时间实验显示，随着时间的推移，MFI增加，表明胶囊与HeLa细胞相关。细胞对载体的表观反应差异是明显的，这取决于相对或绝对定量。绝对定量与标记强度无关，因此更具可比性。这些技术是深入分析和比较颗粒性能的基础。我们很高兴使用这些技术来参数化数学模型，这将使我们能够独立于任何特定实验来表征粒子性能。
