腫瘍細胞の運動性は転移に重要な役割を果たします1,2。腫瘍細胞が近隣や遠隔地の健康な組織に広がると、がん治療が困難になり、再発の一因となります3,4。したがって、腫瘍細胞の運動性のメカニズムを理解し、関連する治療戦略を開発することが不可欠です。多くの腫瘍細胞は遺伝子発現プロファイルを変化させているため、遺伝子発現プロファイルのどの変化が腫瘍細胞の運動性の変化につながるかを理解することは非常に重要です5,6。
in vitroで細胞遊走を測定するために、いくつかのアッセイが開発されています。一部のアッセイでは、特定の時点でしか測定できないため、限られた情報しか提供されませんが、他のアッセイでは、腫瘍細胞の運動性に関する包括的な情報がリアルタイムで提供されます7。これらの細胞運動性アッセイの多くは、特定の時間またはエンドポイントで定量的な結果を得ることができますが、実験期間中の細胞遊走速度の動的変化に関する十分な詳細情報を提供することができません。また、細胞遊走速度の潜在的な変化は、実験計画、細胞の種類、細胞数によっては検討が困難な場合があります。さらに、単純な治療の効果は、従来の運動性アッセイの単純な定量化によって調査することができるが、様々な組み合わせ治療の複雑な効果を研究するには、より高度な定量化が必要になるかもしれない8。
微小電極で覆われたマイクロタイタープレートウェル底の電流をモニターする装置が開発された9。ウェルの表面への細胞の接着は電子の流れを妨げ、インピーダンスは細胞の量的および定性的結合と相関します。坑井底に微小電極が存在することで、細胞の接着、拡散、増殖を測定できます。上部チャンバーの微多孔膜の下に微小電極が存在するため、下部チャンバーへの細胞移動と浸潤の測定が可能になり、上部チャンバーは細胞外マトリックス(ECM)タンパク質でコーティングされ、侵入が可能になります10。
以前、腫瘍細胞の移動と浸潤のインピーダンスベースのリアルタイム測定は、実験全体を通してリアルタイムデータを提供し、さまざまな実験条件下で即座に比較および定量化できることが実証されました11。その方法の論文では、腫瘍細胞の遊走と浸潤における目的のタンパク質の役割をテストするために、遺伝子ノックダウンが誘導されました。試験された実験条件下での本格的な遺伝子ノックダウン効果は、低分子干渉RNA(siRNA)によるエレクトロポレーション後3〜4日を要したため8、エレクトロポレーション後に細胞を再プレーティングし、3日後に再採取して、腫瘍細胞の移動と浸潤のインピーダンスベースのリアルタイム測定を行いました。
キナーゼ(Crk)およびCrk様(CrkL)のCT10調節因子は、様々な成長因子受容体キナーゼ経路および非受容体チロシンキナーゼ経路の下流でタンパク質間相互作用を媒介するアダプタータンパク質である12。CrkおよびCrkLタンパク質のレベルの上昇は、神経膠芽腫を含むいくつかのヒト癌の予後不良に寄与する13。しかし、CrkおよびCrkLタンパク質の上昇がどのようにして予後不良につながるかは不明です。したがって、CrkおよびCrkLの過剰発現が腫瘍細胞機能に及ぼす影響を定義することが重要です。以前、遺伝子ノックダウン研究が実施され、膠芽腫細胞の遊走と浸潤には内因性のCrkおよびCrkLタンパク質が必要であることが実証されました8。ここでは、腫瘍細胞の遊走および浸潤に対するCrkおよびCrkLの過剰発現の影響に対処するために、修正されたアッセイシステムが開発されました。
近年、SARS-CoV-2に対するmRNAワクチンの開発により、mRNAのin vitro合成とその治療応用が改めて注目されています(Verbekeらによるレビュー14)。さらに、がんやその他の疾患における合成mRNAの使用において、目覚ましい進歩が見られました15,16。細胞のエレクトロポレーションは、合成mRNAを送達し、一過性の遺伝子改変を誘導する効果的な方法であり(Campillo-Davoらによるレビュー17)、合成mRNAの使用により、不死化線維芽細胞における迅速かつ効率的な遺伝子発現が可能になります18。この方法論文では、合成mRNAを用いた遺伝子の過剰発現とリアルタイムの細胞解析を組み合わせて、腫瘍細胞の遊走と浸潤を研究しています。しかし、siRNAに用いられる実験スキームは、外因性タンパク質のレベルが合成mRNAトランスフェクションで急激に増加し、合成mRNAトランスフェクション時に徐々に減少するため、合成mRNAトランスフェクションでは機能しない18。そのため、トランスフェクション直後の細胞遊走や浸潤を、細胞を追加培養することなくリアルタイムに解析できるように改良しました。
