ヒト補助生殖技術の開発により、体外受精胚移植(IVF-ET)の現在の臨床妊娠率は自然妊娠のそれに達するか、それを超えています。それにもかかわらず、生殖補助医療の多くの患者は依然として複数の胚移植を受けていますが、希望する妊娠を達成できません。しかし、その具体的な分子メカニズムはまだ不明であるため、臨床介入は効果がなく、生殖医療が直面している重要な課題の1つです1,2。
子宮内膜因子は、体外受精不全の原因の約3分の2を占めています3。ヒト胚着床は、ポジショニング、接着、侵襲の3つの段階に分けられます4,5,6。母体の子宮内膜は、胚の到着に合わせて一連の変化を受けます。着床「ウィンドウ期間」を形成することは、胚着床に好ましい条件を提供する7,8。
ほとんどの哺乳類では、胚盤胞が子宮の管腔上皮に付着した後、胚盤胞を取り巻く間質細胞が急速に増殖および分化し始め、間葉の急速なリモデリングがその形状と機能を変化させ、胚着床につながります5,9,10。部位の体積と重量の急速な増加により、胚盤胞が子宮間質に埋め込まれるようになり、脱落膜化11として知られるプロセスが可能になります。子宮内膜間質は妊娠に備えて分化およびリモデリングしますが、間質細胞の移行は、脱落膜細胞がその機能を果たすためのスペースと新しいシグナル伝達接続を提供します12,13。間質細胞は脱落膜細胞に変化し、プロラクチン(PRL)、インスリン様成長因子結合タンパク質1(Igfbp1)などの多くの象徴的な因子を分泌します。研究によると、異常な脱落膜化が胚着床失敗の主な理由の1つであることが示されていますが、異常な脱落膜化の原因はまだ不明であり、さらに解明する必要があります1,14。
マウス人工脱落膜モデルは、脱落膜化の根底にある生理学的プロセスと分子メカニズムを研究するために不可欠です。人工脱落膜化(AD)は、主に妊娠または月経周期をシミュレートするための人工的な方法によって確立された子宮内膜脱落膜化のプロセスを指します。形態に関しては、妊娠脱落膜化と人工脱落膜化の間に全体的な違いはほとんどありません15,16。子宮腺は脱落膜が形成される前に子宮内膜に存在し、脱落膜化後に消えます。遺伝子発現に関しては、自然妊娠脱落膜化(NPD)とAD15の間にわずかな違いが確認されています。その結果、マウスの人工脱落膜化モデルは、妊娠脱落膜化をシミュレートして、未知の病因とヒト生殖疾患の新しい治療法を探索することができます。
NPD、AD、および in vitro 脱落膜化(IVD)は、マウス脱落膜化を達成するための3つの方法です。NPDモデルは自然妊娠に依存し、胚の影響を含む母体の生理学的状態に最も近い。移植部位と非移植部位の違いを比較することは、脱落膜化を研究するためのより生理学的で便利なアプローチです。ADモデルは、胚の影響を避けるために、精管切除された雄と交配した偽妊娠雌マウスに脱落膜を誘発するための刺激剤としてゴマ油の子宮内注射を使用することによって開発されました。NPDモデルとADモデルはどちらも異なる研究目的で重要な役割を果たしますが、母体ホルモン代謝の異なる活動によって引き起こされる交配の失敗やグループ内の違いを避けることはできません。IVDは、細胞レベルでのエストロゲンとプロゲステロンの併用治療に依存する方法であり、より厳しい実験条件と操作能力が必要です。しかし、 in vitro モデルは、生理学的条件下での脱落膜応答を完全にシミュレートすることはできません15。そこで我々は、脱落膜化に対する内因性ホルモンの影響を低減するために、従来のADから修正された簡便で改良された誘導法を提案する。脱落膜誘導の成功を確実にすることに基づいて、それは生理学的状態に近く、胚因子を排除する必要がある実験により適しています。

地方分権化の障害は、死亡、再発流産などの子宮内膜障害のウイルスにつながります。この方法は、子宮内膜の分散化の研究のための優れた信頼性の高い動物モデルを提供します。卵巣摘出術を伴うこの人工分散化モデルは、グループ内のわずかな違いで再現性の高い結果をもたらすことが多いホルモンに対するアンジェリーナの影響を回避できます。手術の1日前に高温高圧滅菌器で手術器具の滅菌を開始する。麻酔中の乾燥を防ぐために、眼球に保護眼軟膏を塗布します。マウスが完全に麻酔をかけられたら手術を開始します。マウスを手術面の腹臥位に置き、石鹸水で濡らした後、背中の髪を剃ります。70%エタノールで皮膚を消毒します。脊椎に平行な両後肢の大腿根の上端にある0.5センチメートルの手術切開部位を見つけます。切開部位を中心に、ヨードフォアで切開部を3回消毒します。ハサミを使用して約0.5〜1センチメートルの縦方向の切開を行います。筋膜を切り、ピンセットで筋肉を受動的に分離します。薄い筋肉層を通して腎臓の下極に近い切開野で白い脂肪パッドの一部を見つけます。脂肪塊を止血クリップで固定し、脂肪パッドで包まれた卵巣を切開部から引き出します。卵巣と卵管の接合部を湾曲したピンセットで固定し、ハサミで卵巣茎に沿って卵巣を取り除きます。電気凝固ペンまたはアルコールランプで加熱された1ミリリットルの注射器の針を使用して出血を止めます。組織の癒着を防ぐために、外科的切開部を生理食塩水ですすいでください。4-0縫合糸を使用して筋肉と皮膚をそれぞれ播種し、ヨードフォアで外科的切開を再度消毒します。マウスをケージの腹臥位に置き、摂氏25度のインキュベーターで蘇生させます。光源と換気システムを約2時間装備。手術後のマウスに注意してください。完全に回復するまで、元の給餌場所に一人で戻し、十分な水と餌を与えます。50マイクロリットルのゴマ油に100ナノグラムのエストロゲンを各マウスに3日間皮下注射し、その後2日間休息させます。50マイクロリットルのゴマ油に1ミリグラムのプロゲステロンと10ナノグラムのエストロゲンをさらに3日間各マウスに注射します。マウスを操作して、3回目のエストロゲンプロゲステロン併用注射の6時間後に人工残差モデルを誘導する。卵管の下端にある子宮角を見つけ、子宮角に沿って20マイクロリットルのゴマ油をゆっくりと注入します。組織を押し戻し、切開部を縫合し、マウスを回復させます。パラフィンに埋め込まれた10%ホルマリンで3〜5ミリリットルの子宮組織を固定し、それらを切片化します。切片をヘマトキシリンとエオジンで染色して、形態学的変化を観察します。子宮組織のさらに3〜5ミリメートルを最適な切断温度に埋め込み、液体窒素で凍結し、摂氏マイナス80度の冷凍庫に保管します。この組織を10マイクロメートルで凍結切片し、アゾカップリング法により子宮組織中のアルカリホスファターゼ活性を検出した。油によって誘発されたマウスの人工分散型子宮の一般的な形態は、妊娠中の子宮の形態に近い。子宮体は厚くなり、子宮腔は非誘発側よりも小さくなります。誘発された子宮角の体積と重量は、非誘発側の体積と重量よりも有意に高い。子宮組織のヘマトキシリンおよびエオジン染色は、腺が消失し、誘導された引き裂かれた上の子宮内膜末間質細胞が、細胞境界が不明瞭な大きな丸い細胞質および多核脱落膜細胞に分化したことを示した。非誘導側切片は、正常な生理学的状態下での子宮内膜組織形態を示す。アゾカップリング法の結果から,油誘導側のアルカリホスファターゼ含量は非誘導側のアルカリホスファターゼ含量が有意に高かった。QPCRの結果は、誘導された角におけるPRL-8-A-2、ALPL、およびIGFBP-1のmRNA発現が非誘導角のそれよりも有意に高いことを示し、油がマウスの人工的な分散化を誘発できることを示唆しています。ホモの補給は、成功への鍵であるホモ間の汚染に特別な注意を必要とする長いプロセスです。このモデルに基づいて、アデノウイルスの子宮角注射を行うことができ、残留を調節する特定の分子の関連メカニズムを研究することができます。
