가장 중요한 공중 보건 혁신 중 하나인 백신은 전 세계적인 인간 질병 부담과 싸우는 데 핵심적인 역할을 합니다1. 예를 들어, 현재 COVID-19 질병에 대한 120개 이상의 후보 백신이 테스트되고 있으며 그 중 일부는 많은 국가에서 승인되었습니다2. 최근 보고에 따르면 암 백신은 암 환자의 면역체계가 항원을 체내에 이물질로 인식하도록 지시하기 때문에 임상 암 치료의 진행을 효과적으로 개선했다고한다 3. 더욱이, 종양 세포 내부 또는 외부에 위치한 다중 T 세포 에피토프는 펩티드 백신을 설계하는 데 사용될 수 있으며, 이는 방사선 요법 및 화학 요법과 관련된 상당한 독성이 없기 때문에 전이성 암 치료에 이점을 보여주었습니다 4,5. 1990년대 중반 이래로 종양 치료를 위한 전임상 및 임상 시험이 주로 항원 펩티드 백신을 사용하여 수행되었으나 암 환자에게 적절한 치료 효과를 나타내는 백신은 거의 없다6. 또한, 펩타이드 에피토프를 이용한 암 백신은 면역원성이 낮고 전달 효율이 불충분한데, 이는 투여 부위에서 빠르게 확산되는 세포외 펩타이드의 급속한 분해로 인해 면역세포에 의한 항원 흡수가 불충분하기 때문일 수 있다7. 따라서 백신 전달 기술로 이러한 장애물을 극복할 필요가 있다.
융합 단백질로서 발현되는 MHC 클래스 I-결합 257-264 에피토프인 OVA257-264는 빈번하게 사용되는 모델 에피토프8이다. 또한, OVA257-264는 세포독성 T 림프구 (CTL) 반응에 의존하는 종양에 대한 적응 면역 반응에 결정적이다. 세포-조직 및 세포-세포 상호 작용의 에피토프를 결합하기 위해 여러 합성 생체 재료가 고안되었습니다. Ile-Lys-Val-Ala-Val (IKVAV)과 같은 특정 생리활성 단백질이 하이드로겔 구조의 일부로 도입되어 생체 활성을 부여한다12. 이 펩타이드는 세포 부착, 이동 및 성장에 기여할 가능성이 높으며13 인테그린α 3β1 및 α6β1에 결합하여 다른 암세포 유형과 상호 작용합니다. W805EC 및 MF59와 같이 최근에 보고된 에멀젼 시스템도 불활화 인플루엔자 백신 또는 재조합 B형 간염 표면 항원의 비강 전달을 위해 합성되었으며 점막 및 전신 면역을 모두 유발하는 것으로 설명되었다15. 나노에멀젼(NEs)은 미립자 점막 전달 시스템에 비해 투여가 용이하고 효과적인 보조제와의 편리한 공형성의 장점이 있다16. 나노에멀젼 백신은 기존의 탈감작과는 다른 지속적인 방식으로 알레르기 표현형을 변화시켜 장기적인 억제 효과를 가져오는 것으로 보고되었다17. 다른 연구에서는 MTB 특이적 면역우성 항원과 결합된 나노에멀젼이 강력한 점막 세포 반응을 유도하고 상당한 보호를 제공할 수 있다고 보고했다18. 따라서, 합성 펩타이드 IKVAV-OVA257-264(I-OVA ,OVA 257-264에 결합된 IKVAV로 구성된 펩타이드)를 이용한 신규한 비강내 자가조립 나노백신을 설계하였다. 이 새로운 나노백신을 체계적으로 평가하는 것이 중요합니다.
