De bovenstaande protocollen geven een gedetailleerde beschrijving van de assemblage van de nieuwe imager en de werking ervan in de microseconde FLIM/PLIM-modus. De TCSPC-gebaseerde nieuwe generatie Tpx3Cam-camera, gekoppeld door middel van de optomechanische adapter Cricket met de beeldversterker, emissiefilter en macrolens, produceert een stabiele, compacte en flexibele optische module die eenvoudig te bedienen is. O 2 is een van de belangrijkste omgevingsparameters voor levende systemen, en kennis van de verdeling van O 2 en zijn dynamiek is belangrijk voor veel biologische studies 1,2,3. De beoordeling van weefseloxygenatie door middel van fosforescerende sondes 4,5,6,7,8 en PLIM 9,10,11,12,13 wint aan populariteit in biologisch en medisch onderzoek 3,9,14,15,16, 17,18,19. Dit komt omdat PLIM, in tegenstelling tot fluorescentie- of fosforescentie-intensiteitsmetingen, niet wordt beïnvloed door externe factoren zoals sondeconcentratie, fotobleaching, excitatie-intensiteit, optische uitlijning, verstrooiing en autofluorescentie.
De huidige O2 PLIM-platforms worden echter beperkt door hun gevoeligheid, beeldacquisitiesnelheid, nauwkeurigheid en algemene bruikbaarheid. Time-correlated single photon counting (TCSPC), gecombineerd met een rasterscanprocedure, wordt vaak gebruikt in PLIM- en fluorescentie-lifetime imaging microscopie (FLIM) -apparaten20,21,22. Omdat PLIM echter een lange pixelverblijftijd vereist (in het millisecondebereik), is de tijd van beeldacquisitie veel langer dan wat vereist is voor FLIM-toepassingen20,22,23. Andere technieken, zoals gated CCD / CMOS-camera's, missen enkele fotongevoeligheid en hebben lage framesnelheden20,24,25,26. Bovendien worden de bestaande PLIM-systemen meestal gebruikt in het microscopische formaat, terwijl macroscopische systemen minder vaak voorkomen27.
De op TCSPC gebaseerde PLIM-macro-imager28 is opgezet om veel van deze beperkingen te overwinnen. Het ontwerp van de imager werd aanzienlijk vergemakkelijkt door het gebruik van een nieuwe opto-mechanische adapter, Cricket, die het volgende heeft: i) twee C-mount adapters, die een eenvoudige koppeling van de cameramodule aan de achterkant en een objectieflens aan de voorkant bieden; ii) een interne behuizing voor een beeldversterker en een stopcontact voor de laatste aan de buitenzijde van de Krekel; iii) een interne ruimte achter de C-montageadapter aan de voorzijde waar een standaard emissiefilter van 25 mm voor de versterker kan worden ondergebracht; en iv) een ingebouwde lichtbotsende optiek met ringregelaars, die optische uitlijning / scherpstelling tussen de lens en de camera mogelijk maken om scherpe beelden op de camerachip te produceren.
In de geassembleerde imager is de cameramodule gekoppeld aan de achterkant van de Cricket-adapter, die ook een beeldversterker herbergt die bestaat uit een fotokathode gevolgd door een microkanaalplaat (MCP), een versterker en een snelle scintillator, P47-fosfor. Een 760 nm ± 50 nm emissiefilter is gemonteerd in de Cricket en een objectieflens, NMV-50M11'', is bevestigd aan de C-mount adapter aan de voorzijde. Tot slot zijn de lens en de camera optisch uitgelijnd met ringregelaars.
De rol van de versterker is om inkomende fotonen te detecteren en om te zetten in snelle lichtflitsen op de camerachip, die worden geregistreerd en gebruikt om emissieverval en levensduurbeelden te genereren. De cameramodule bestaat uit een geavanceerde TCSPC-gebaseerde optische sensorarray (256 pixels x 256 pixels) en een nieuwe generatie uitleeschip 29,30,31,32,33, die de gelijktijdige registratie van de aankomsttijd (TOA) en de tijd over drempel (TOT) van fotonuitbarstingen bij elke pixel van de beeldchip mogelijk maakt met een tijdresolutie van 1,6 ns en een uitleessnelheid van 80 Mpixel / s.
In deze configuratie heeft de camera met de versterker een enkele fotongevoeligheid. Het is datagedreven en gebaseerd op het speedy pixel detector readout (SPIDR) systeem34. De ruimtelijke resolutie van de imager werd eerder gekarakteriseerd met vlakke fosforescerende O2-sensoren en een resolutieplaatmasker. De instrumentresponsfunctie (IRF) werd gemeten door de beeldvorming van een vlakke fluorescentiesensor onder dezelfde instellingen als voor alle andere metingen. De levensduur van de kleurstof van ongeveer 2,6 ns was kort genoeg om te worden gebruikt voor de IRF-meting in PLIM-modus. De imager kan objecten van maximaal 18 mm x 18 mm groot maken met ruimtelijke en temporele resoluties van respectievelijk 39,4 μm en 30,6 ns (volledige breedte bij half-maximum),28.
De volgende protocollen beschrijven de assemblage van de macro-imager en het daaropvolgende gebruik ervan voor het in kaart brengen van de O 2-concentratie in biologische monsters die zijn gekleurd met de eerder gekarakteriseerde nabij-infrarode O2-sonde, NanO2-IR35. De sonde is een heldere, fotostabiele, celdoorlatende O2-sensing sonde op basis van platina (II) benzoporfyrine (PtBP) kleurstof. Het is prikkelbaar bij 625 nm, zendt uit bij 760 nm en biedt een robuuste optische respons op O 2 in het fysiologische bereik (0% -21% of 0-210 μM van O2). De imager blijkt ook verschillende sensormaterialen te karakteriseren op basis van Pt(II)-porfyrinekleurstoffen. Over het algemeen is de imager compact en flexibel, vergelijkbaar met een gewone fotocamera. In de huidige opzet is de imager geschikt voor verschillende breedveld PLIM-toepassingen. Het vervangen van de LED door een snelle laserbron zal de prestaties van de imager verder verbeteren en kan mogelijk nanoseconde FLIM-toepassingen mogelijk maken.

fluorescence lifetime macro imager for biomedical applications

Dit artikel presenteert een nieuwe fotoluminescentie lifetime imager ontworpen om de moleculaire zuurstof (O 2) concentratie in verschillende fosforescerende monsters in kaart te brengen, variërend van solid-state, O 2-gevoelige coatings tot levende dierlijke weefselmonsters gekleurd met oplosbare O 2-gevoelige sondes. In het bijzonder werd de op nanodeeltjes gebaseerde nabij-infrarode sonde NanO2-IR gebruikt, die prikkelbaar is met een 625 nm light-emitting diode (LED) en uitzendt bij 760 nm. Het beeldsysteem is gebaseerd op de Timepix3-camera (Tpx3Cam) en de optomechanische adapter, die ook een beeldversterker herbergt. O2 fosforescentie lifetime imaging microscopie (PLIM) is vaak vereist voor verschillende studies, maar de huidige platforms hebben beperkingen in hun nauwkeurigheid, algemene flexibiliteit en bruikbaarheid.
Het hier gepresenteerde systeem is een snelle en zeer gevoelige imager, die is gebouwd op een geïntegreerde optische sensor en uitleeschipmodule, Tpx3Cam. Het is aangetoond dat het fosforescentiesignalen met hoge intensiteit en stabiele levensduurwaarden produceert uit oppervlaktegekleurde darmweefselmonsters of intraluminaal gekleurde fragmenten van de dikke darm en maakt het mogelijk om weefsel O2-niveaus gedetailleerd in kaart te brengen in ongeveer 20 s of minder. Eerste experimenten met de beeldvorming van hypoxie in getransplanteerde tumoren bij bewusteloze dieren worden ook gepresenteerd. We beschrijven ook hoe de imager opnieuw kan worden geconfigureerd voor gebruik met O2-gevoelige materialen op basis van Pt-porfyrinekleurstoffen met behulp van een 390 nm LED voor de excitatie en een bandpass 650 nm filter voor emissie. Over het algemeen bleek de PLIM-imager nauwkeurige kwantitatieve metingen van levensduurwaarden voor de gebruikte sondes en respectieve tweedimensionale kaarten van de O2-concentratie te produceren. Het is ook nuttig voor de metabole beeldvorming van ex vivo weefselmodellen en levende dieren.

Voor ex vivo beeldvormingstoepassingen werden fragmenten van darmweefsels gekleurd door de topische toepassing van de NanO2-IR-sonde aan de serosale zijde van het weefsel. Voor diepere kleuring werd 1 μL van de sonde in het lumen geïnjecteerd. In het laatste geval schermde de 0,2-0,25 mm dikke darmwand de sonde af van de camera. De twee kleuringsprocessen worden gedemonstreerd in figuur 2A.

Ons protocol biedt gedetailleerde 2D-mapping van fosforescentielevensduur en zuurstofconcentratie in macroscopische objecten, met name levend dierlijk weefsel en hele dieren, door middel van speciale sondes en detectiematerialen. Deze informatie is belangrijk voor veel onderzoeksgebieden. Ons protocol maakt gebruik van een geïntegreerde en compacte beeldvormingsmodule die werkt in TCSPC-modus en biedt eenvoudige en nauwkeurige visualisatie van levensduur en zuurstofverdeling in complexe biologische monsters, zoals levensweefsel.Neem de Cricket adapter en inspecteer deze van verschillende kanten. Verwijder de C-mount adapter aan de voorzijde om de Photonis PP0360EF versterker in de Cricket te laten zien en plaats vervolgens de C-mount adapter terug. Verwijder de C-mount adapter aan de voorzijde van de Cricket en plaats het 650 plus/minus 50 nanometer emissiefilter.Repareer het door de C-mount terug te plaatsen. Sluit vervolgens de TPX-cameramodule met drie nokken aan op de achterkant van de Cricket-module via hun ringadapters. Sluit de lens aan op de voorkant van de Cricket-module via de C-mount adapter.Sluit de LED aan op een voeding en een pulsgenerator. Monteer nu de 390-nanometer superheldere LED op een paal die is bevestigd aan een breadboard in de zwarte doos. Schakel de LED in en focus deze om een effectieve en uniforme excitatie van het afgebeelde monster te garanderen.Monteer de camera-assemblage bovenop de optische zwarte doos naar beneden gericht naar het podium waar de monsters worden afgebeeld. Sluit de camera en LED aan op generatoren met twee pulsen en een vierkanaals oscilloscoop. Met behulp van instellingen op de oscilloscoop en pulsgeneratoren synchroniseert u de pulsen die naar de camera en de LED worden verzonden.Gebruik de speciale kabel en aansluiting op de Cricket-eenheid om de versterker aan te sluiten op een standaardvoeding en stel de versterking in op 2,7 volt. Plaats het monster voor de cameralens. Schakel vervolgens de camera en alle elektrische modules in, behalve de versterker.Activeer de SOFI-software om de operationele parameters af te stemmen, zoals scherpstellen en sample-uitlijning. Stel in de modules de framebelichting in op 0,01 seconden. Selecteer Oneindige frames en stel de pixelbewerkingsmodus in op tijd boven drempel.Ga vervolgens naar Voorvertoning en selecteer Actieve module om het venster Medipix / Timepix-frames te openen. Pas in de volgende stap, na het starten van de opname, de kleurenschaal aan en roteer de afbeelding naar de gewenste oriëntatie. Schakel daarna de lichten in de kamer uit en schakel de versterker in.Focus de camera-optiek op het voorbeeldpodium met de scherpstelmogelijkheden van de lens en Cricket-adapter om heldere beelden van monsters met een goed contrast en helderheid te genereren. Zodra de focus is voltooid, sluit u de SOFI-software. Ga vervolgens naar de terminal, voer de opdrachten van de op maat gemaakte ontwerpsoftware uit om de onbewerkte gegevens in het binaire formaat te verkrijgen en sluit de terminal.Open een nieuwe terminal om de verkregen gegevens te verwerken. Laad het gegevensbestand en voer het gegevensreducer uit. Analyseer de nabewerkingsgegevens met een speciaal programma geschreven in C-taal dat de gegevens in een ICS-afbeeldingsbestand schrijft.Open vervolgens de ICS-afbeeldingsbestanden met behulp van de vrij beschikbare Time Resolved Imaging-software en gebruik twee exponentiële functies om het fosforescentieverval aan te passen. Open ten slotte de Fitted. ICS-afbeeldingsbestanden met de beschikbare beeldanalysesoftware.Genereer fosforescentielevensduurafbeeldingen met behulp van opzoektabellen en codeer ze in pseudo-kleurenschaal. Gebruik de functie Meten om de gemiddelde levensduurwaarden voor de gehele afbeelding of interessegebieden te berekenen. Fosforescentie-intensiteit en fosforescentie lifetime imaging microscopiebeelden van de platina octaethylporfyrine kleurstofsensorvlek in de zuurstofrijke en zuurstofarme toestanden worden getoond.Zorg ervoor dat het juiste emissiefilter in de Cricket is gemonteerd en dat de juiste LED, kabelverbindingen en pulsinstellingen worden gebruikt. Momenteel is dit protocol beperkt tot diermodellen, maar kan mogelijk worden toegepast om pathologische toestanden geassocieerd met weefselhypoxie, zoals kanker, beroerte en ontsteking, te diagnosticeren en te behandelen.
