1. 昆虫准备

最后，孵化器中有足够的空间来添加外部光或湿度源，而不会影响笼子中的昆虫。根据昆虫种类或实验设计，一个或两个培养箱内的笼子顶部可以很容易地将 1 cm 厚的 LED 灯放置在笼子顶部。对于某些光敏实验设计，为两者提供光并提供温度选择可能是更现实的方案，或者仅向一个腔室提供光（或湿度）是对评估光/湿度选择的方案的可能修改。

协议中的全尺寸设备明显小于温度梯度设置，如上所述，可以更轻松地安装在BSL 3手套箱内。此外，昆虫更容易控制，因为它们可以在实验结束时用CO2 击倒，并且在与桥分离后可以快速重新密封笼子。这些密闭优势是BSL 3工作的理想选择。
然而，我们承认，我们的设备只允许二元决策，而不是沿着梯度自由选择，根据应用的不同，可能需要额外的运行来确定最佳温度。

生物体只能在其热耐受范围内生活和繁殖。由于季节变化和全球变暖，环境温度会发生变化，物种必须相应地适应和应对，以确保其生存。这包括变温，其中体温与环境平衡1。因此，每只昆虫都有自己的最佳环境温度范围，它们试图保持在2 以内。
温度是预测昆虫分布和范围的重要因素之一3，4，5，观察病原虫关系6，7以及外部因素对变温动物适应性的影响，如成虫寿命，繁殖力和摄食率8，9。
以前的研究已经调查了具有不同设置的变温管的首选温度。最常见的是使用带有冷却或热水浴10，冰浴和可编程加热器元件11，冷板和热板12，13，热调节板14，15或热袋和冰袋16在两端的大型铝块以创建温度梯度。此外，其他研究还使用温度梯度培养箱来研究所选细菌17的生长，并将铝棒安装在热电装置上（在末端加热和冷却）以观察黑腹果蝇18，19的热偏好。
