細胞はさまざまなサイズの細胞外小胞(EV)を放出し、細胞間コミュニケーションに重要なシグナル伝達分子と膜封入体をもたらします1。異なるサイズのEVも異なる生物学的役割を果たし、50〜200nmのsEVはRNA、DNA、およびタンパク質を正しい細胞外位置に正確に分配することができます。sEVはまた、免疫監視、幹細胞維持、血液凝固、組織修復などの正常な生理学的プロセスの調節だけでなく、腫瘍の進行や転移などのいくつかの疾患の根底にある病状を含む、それらの分泌メカニズムを決定するのに役立ちます2,3。sEVの効果的な分離と分析は、バイオマーカーを特定し、将来の薬物介入を設計するために重要です。
sEVの臨床応用に関する継続的な研究により、sEVの分離方法はより高い要件を提唱しています。sEVのサイズ、ソース、内容の不均一性、および物理化学的および生化学的特性における他のEVとの類似性により、sEVを分離するための標準的な方法はありません4,5。現在、超遠心分離、サイズ排除クロマトグラフィー(SEC)、ポリマー沈殿、および免疫親和性捕捉が最も一般的なsEV分離方法です6。超遠心分離は、時間がかかるにもかかわらず、研究におけるsEV分離のゴールドスタンダードであり、その結果、40〜500 nmの広いサイズ分布で低純度になり、長期遠心分離後のsEVに重大な機械的損傷が生じます7,8,9。通常ポリエチレングリコール(PEG)を使用するポリマー沈殿は、細胞外タンパク質凝集体の沈殿とポリマー汚染を伴うその後の機能分析では許容できない純度に悩まされています10,11。免疫親和性捕捉ベースの方法は、様々な特異性を有する高コストの抗体を必要とし、また、処理量および収率が低いという問題を有する12,13,14。粒子径は、単離されたsEVの純度を評価するための主要な指標の1つです。sEVの純度は達成不可能な目標のままですが、SECはかなりの量の媒体成分を除去し、SEC法によって抽出されたsEV粒子は主に50〜200nmの範囲にあります15。既存の技術には、時間がかかる、純度が低い、収率が低い、再現性が低い、サンプルのスループットが低い、sEVの潜在的な損傷など、いくつかの欠点があり、臨床利用と両立しません16。したがって、多様な生体液に適用可能な迅速で安価でサイズ指定のsEV分離方法は、いくつかの研究および臨床状況で不可欠なニーズです。
フローサイトメトリーでは、単一粒子がハイスループット、マルチパラメトリック方式で分析され、サブセットが選別されます17。EVの不均一性のために、単一粒子フローサイトメトリー測定が理想的であり、これは細胞分析のパラダイムに従ってEVを調査するために使用されており、光散乱および蛍光標識を使用して生理学関連の特徴およびタンパク質成分を特定するために使用されている18,19,20。それにもかかわらず、従来のフローサイトメトリーは、sEVのサイズが小さく、表面バイオマーカーの存在量が少ないという課題があります。フローサイトメトリーの感度は、前方散乱(FSC)、側方散乱(SSC)、または蛍光閾値トリガーパラメータ19の使用に関係なく、バックグラウンドノイズとsEVを区別するように検出パラメータを最適化することで改善できます。
本プロトコルでは、蛍光ビーズを標準として高分解能フローサイトメトリーのソート設定を最適化しました。適切なノズルサイズ、シース液圧力、しきい値トリガーパラメータ、および散乱光強度を制御する電圧を選択することにより、複雑な混合物からsEVの特定のサブセットを分離することができました。
