Het vermogen om BFU-e en CFU-e voorlopers prospectief te isoleren rechtstreeks uit vers weefsel met hoge zuiverheid was eerder onderzoekers ontgaan. Onze nieuwe aanpak, gevalideerd met scRNAseq en cell fate assays 1,27, biedt nu de tools om dit te doen.
Er zijn een aantal belangrijke punten voor het succesvol uitvoeren van zowel de sorteer- als de analytische protocollen. Eerst moeten de cellen worden gesponnen bij 900 x g om ...

Erytropoëse kan worden onderverdeeld in twee hoofdfasen: vroege erytropoëse en erytroïde terminale differentiatie (figuur 1)1,2,3. In vroege erytropoëse binden hematopoëtische stamcellen zich aan de erytroïde afstamming en geven ze aanleiding tot vroege erytroïde voorlopers, die voor het eerst werden geïdentificeerd in de jaren 1970 op basis van hun kolonievormend potentieel in halfvast medium 4,5,6,7,8,9 . In grote lijnen zijn erytroïde voorlopers verdeeld in twee categorieën: eerdere voorlopercellen die elk aanleiding geven tot een "burst" (een groot aggregaat van kleinere erytroïde celclusters), genaamd "burst-forming unit erythroid" of BFU-e 4,5,6; en hun nakomelingen, die elk een enkele, kleine erytroïde celcluster of kolonie vormen, genaamd "kolonievormende eenheid erytroïde" of CFU-e 7,8,9. BFU-e en CFU-e brengen nog geen terminale erytroïde genen tot expressie en zijn morfologisch niet herkenbaar. Na een aantal zelfvernieuwings- of expansieceldelingen ondergaat de CFU-e een transcriptionele schakelaar waarbij erytroïde genen zoals globines worden geïnduceerd, waardoor deze overgaat in erytroïde terminale differentiatie (ETD)1,10. Tijdens ETD ondergaan erytroblasten drie tot vijf rijpingsceldelingen voordat ze enucleeren om reticulocyten te vormen, die rijpen tot rode bloedcellen.
Erytroblasten tijdens terminale differentiatie werden oorspronkelijk geclassificeerd op basis van hun morfologie in proerytroblasten, basofiele, polychromatische en orthochromatische. De komst van flowcytometrie maakte hun prospectieve sortering en isolatie mogelijk op basis van celgrootte (gemeten door voorwaartse spreiding, FSC) en twee celoppervlakmarkers, CD71 en Ter11911,12,13 (figuur 1). Deze en soortgelijke flowcytometrische benaderingen14 hebben een revolutie teweeggebracht in het onderzoek naar de moleculaire en cellulaire aspecten van ETD, waardoor ontwikkelingsstadiumspecifieke analyse van erytroblasten in vivo en in vitro 10,15,16,17,18,19,20 mogelijk is. De CD71/Ter119-benadering wordt nu routinematig gebruikt bij de analyse van erytroïde precursoren.
Tot voor kort is een vergelijkbare, toegankelijke flowcytometrische benadering voor directe, zeer zuivere prospectieve isolatie van CFU-e en BFU-e uit muizenweefsel onderzoekers ontgaan. In plaats daarvan hebben onderzoekers flowcytometrische strategieën gebruikt die slechts een fractie van deze voorlopers isoleren, vaak in de aanwezigheid van niet-erytroïde cellen die co-zuiveren binnen dezelfde flowcytometrische subsets21. Bijgevolg was het onderzoek van BFU-e en CFU-e beperkt tot in vitro differentiatiesystemen die BFU-e en CFU-e afleiden en versterken van eerdere beenmergvoorlopers. Het is dan mogelijk om flowcytometrische strategieën toe te passen die CFU-e onderscheiden van BFU-e in deze erytroïde voorloperverrijkte culturen22,23. Een alternatieve benadering maakt gebruik van foetale CFU-e en BFU-e, die sterk verrijkt zijn in de Ter119-negatieve fractie van de foetale lever van de muis halverwege de zwangerschap 10,24,25. Geen van deze benaderingen maakt echter het onderzoek van volwassen BFU-e en CFU-e in hun fysiologische toestand in vivo mogelijk. De omvang van de uitdaging kan worden gewaardeerd wanneer eraan wordt herinnerd dat, op basis van kolonievormingstests, deze cellen aanwezig zijn in het volwassen beenmerg met een frequentie van respectievelijk slechts 0,025% en 0,3%6.
Het hier beschreven protocol is een nieuwe flowcytometrische benadering op basis van eencellige transcriptomische analyse van vers geoogste Kit+ beenmergcellen van muizen (Kit wordt uitgedrukt door alle vroege voorloperpopulaties van het beenmerg)1. Onze aanpak bevat enkele celoppervlakmarkers die al in gebruik waren door Pronk et al.21,26. Eencellige transcriptomen werden gebruikt om combinaties van celoppervlakmarkers te bepalen die erytroïde en andere vroege hematopoëtische voorlopercellen identificeren (figuur 2). In het bijzonder kan de CD55+ fractie van lineage-negatieve (Lin-) Kit+ cellen worden onderverdeeld in vijf populaties, waarvan er drie aaneengesloten segmenten van het erytroïde traject opleveren (figuur 2). De transcriptomische identiteiten van elk van deze populaties werden bevestigd door sortering, gevolgd door scRNAseq en projectie van de gesorteerde eencellige transcriptomen terug op de oorspronkelijke transcriptomische kaart (de genexpressie in elk van de vijf populaties en de volledige beenmergdataset kan worden onderzocht in https://kleintools.hms.harvard.edu/paper_websites/tusi_et_al/index.html)1 . Het potentieel van het lot van elk van de populaties werd bevestigd met behulp van traditionele kolonievormingstests (figuur 2), evenals een nieuwe high-throughput single-cell fate assay 1,27. Deze analyses tonen aan dat de nieuwe flowcytometrische benadering resulteert in een zeer zuivere isolatie van alle BFU-e en CFU-e voorlopers van vers volwassen beenmerg en milt. In het bijzonder bevat populatie 1 (P1) alleen CFU-e en geen andere hematopoëtische voorlopers, en populatie 2 (P2) bevat alle BFU-e-voorlopers van het beenmerg en een klein aantal CFU-e, maar geen andere voorlopers1. Het gedetailleerde protocol hieronder wordt verder geïllustreerd met een voorbeeldexperiment bij muizen die werden geïnjecteerd met een zoutoplossing of met het erytropoësestimulerend hormoon erytropoëtine (Epo).

identification and isolation of burst-forming unit and colony-forming unit erythroid progenitors from mouse tissue by flow cytometry

Vroege erytroïde voorlopers werden oorspronkelijk gedefinieerd door hun kolonievormende potentieel in vitro en ingedeeld in burst-vormende en kolonievormende "eenheden" bekend als BFU-e en CFU-e. Tot voor kort waren methoden voor de directe prospectieve en volledige isolatie van zuivere BFU-e en CFU-e voorlopers uit vers geïsoleerd volwassen muizenbeenmerg niet beschikbaar. Om deze kloof aan te pakken, werd een eencellige RNA-seq (scRNAseq) dataset van het beenmerg van muizen geanalyseerd op de expressie van genen die coderen voor celoppervlakmarkers. Deze analyse werd gecombineerd met cel lot assays, waardoor de ontwikkeling van een nieuwe flowcytometrische benadering mogelijk werd die volledige en zuivere subsets van BFU-e en CFU-e voorlopers in het beenmerg of de milt van muizen identificeert en mogelijk maakt. Deze benadering identificeert ook andere voorlopersubsets, waaronder subsets verrijkt voor basofielen / mestcellen en megakaryocytische potentialen. De methode bestaat uit het labelen van verse beenmerg- of miltcellen met antilichamen gericht op Kit en CD55. Voorlopers die beide markers uitdrukken, worden vervolgens onderverdeeld in vijf hoofdpopulaties. Populatie 1 (P1 of CFU-e, Kit+ CD55+ CD49fmed/low CD105med/high CD71med/high) bevat alle CFU-e voorlopers en kan verder worden onderverdeeld in respectievelijk P1-low (CD71med CD150high) en P1-hi (CD71high CD150low), overeenkomend met respectievelijk vroege en late CFU-e; Populatie 2 (P2 of BFU-e, Kit+ CD55+ CD49fmed/low CD105med/high CD71low CD150high) bevat alle BFU-e voorlopers; Populatie P3 (P3, Kit+ CD55+ CD49fmed/high CD105med/low CD150low CD41low) is verrijkt voor basofiele/mestcelvoorlopers; Populatie 4 (P4, Kit+ CD55+ CD49fmed/high CD105med/low CD150high CD41+) is verrijkt voor megakaryocytische voorlopers; en Populatie 5 (P5, Kit+ CD55+ CD49fmed/high CD105med/low CD150high CD41-) bevat voorlopers met erytroïde, basofiele/mestcel en megakaryocytisch potentieel (EBMP) en erytroïde/ megakaryocytische/ basofiel-bevooroordeelde multipotentiële voorlopers (MPP's). Deze nieuwe benadering maakt een grotere precisie mogelijk bij het analyseren van erytroïde en andere hematopoëtische voorlopers en maakt ook verwijzing naar transcriptoominformatie mogelijk voor elke flowcytometrisch gedefinieerde populatie.

Het protocol beschrijft een flowcytometrische benadering om BFU-es en CFU-es te identificeren in vers geoogst beenmerg- en miltcellen. Het begint met het oogsten van verse BM en milt van muizen en het onmiddellijk op ijs leggen van het weefsel. Identification and Isolation of Burst-Forming Unit and Colony-Forming Unit Erythroid Progenitors from Mouse Tissue by Flow Cytometry

Hier beschrijven we een nieuwe flowcytometrische methode voor prospectieve isolatie van vroege burst-forming unit erythroid (BFU-e) en kolonievormende unit erythroid (CFU-e) voorlopers rechtstreeks uit vers beenmerg en milt van muizen. Dit protocol, ontwikkeld op basis van eencellige transcriptomische gegevens, is het eerste dat alle erytroïde voorlopercellen van het weefsel met een hoge zuiverheid isoleert.

Dit protocol maakt de identificatie en isolatie van BFU-E en CFU-E erytroïde voorlopers rechtstreeks mogelijk uit vers geoogst muizenweefsel zoals beenmerg en milt. Met behulp van deze techniek kunnen we zuivere populaties van erytroïde voorlopers isoleren, wat niet mogelijk was met de vorige stroomprotocollen. Deze methode verbetert ons vermogen om muismodellen met erytroïde ziekte te bestuderen aanzienlijk door voorloperisolatie mogelijk te maken voor veel downstream-experimenten.Plaats om te beginnen de vers ontlede dijbenen en het scheenbeen direct in de koude kleuringsbuffer of SB-5 en houd de buizen op ijs totdat ze klaar zijn om het beenmerg te extraheren. Plaats een schone gesteriliseerde mortel op ijs in een emmer en breng de botten over op de mortel. Knip met behulp van de dissectieschaar ongeveer 1 millimeter uiteinden van elk bot af.Gebruik een spuit van 3 milliliter uitgerust met een naald van 26 gauge met SB-5 om het merg uit de botten te spoelen en in de mortel te verzamelen en herhaal het proces 3 tot 5 keer met behulp van een verse buffer elke keer totdat de botten kleurloos lijken. Filter het gespoelde merg door een gaas van 100 micrometer en verzamel de cellen in een centrifugebuis van 50 milliliter die op ijs is geplaatst. Gebruik vervolgens de vijzel en stamper om de gespoelde botten in 5 tot 10 milliliter SB-5 te verpletteren.Filter het mengsel van gemalen botten en buffer door de 100-micrometer celzeef om zich te poolen met de eerder verzamelde cellen. Plaats een gaasfilter van 100 micrometer op een lege centrifugebuis van 50 milliliter die op ijs is geplaatst. Plaats een enkele milt op het gaasfilter en voeg 0,5 tot 1 milliliter SB-5 toe aan de milt om ervoor te zorgen dat deze niet droogt.Gebruik de rubberen kant van een 3- of 5-milliliter spuitzuiger om de milt door het gaas te persen. Voeg meer SB-5 toe, 5 milliliter per keer, en blijf pureren totdat alle cellen in de buis zijn verzameld. Bereid een eencellige suspensie voor door de verzamelde cellen met 2 milliliter SB-5 op en neer te pipetteren met behulp van een grote openingpunt.Kleur de fluorescentie minus één of FMO-controles door alle antilichamen behalve het gelijknamige antilichaam van de FMO toe te voegen aan de celsuspensie in de FACS-buis. Om de controles met één kleur te kleuren, voegt u een enkel antilichaam toe aan de celsuspensie in de FACS-buis. Vortex elke controlebuis gedurende 2 tot 3 seconden bij 3000 RPM en incubeer vervolgens bij 4 graden Celsius schommelen gedurende 2,5 uur in het donker.Was na de incubatie de cellen met SB-5 en vul het volume van de celsuspensie aan tot 4 milliliter met SB-5. Draai de cellen op 900 g gedurende 10 minuten bij 4 graden Celsius en adem het supernatant op. Schorste de niet-gebeitste en alle eenkleurige bedieningselementen opnieuw in 300 microliter SB-5 en filter door een filtergaas van 40 micrometer in nieuwe FACS-buizen.Maak een DAPI SB-5-oplossing door de DAPI-voorraad te verdunnen in een verhouding van 1 tot 10.000 in SB-5. Suspendeer de FMOS opnieuw in 1,8 milliliter DAPI SB-5 en DAPI single-color control in 300 microliter DAPI SB-5 en filter ze vervolgens door een filtergaas van 40 micrometer in nieuwe FACS-buizen. Na het toevoegen van de antilichaam master mix aan elk monster, vortex de buizen gedurende 2 tot 3 seconden bij 3000 rpm gevolgd door incubatie bij 4 graden Celsius in het donker gedurende 2,5 uur in de schommelende toestand.Na het wassen van de cellen zoals eerder aangetoond, suspensie de monsters opnieuw in 1,8 milliliter DAPI SB-5 en filter door een 40-micrometer filtergaas in een nieuwe FACS-buis en analyseer de monsters met behulp van een cystometer. Gebruik de voorwaartse verstrooiing om het vuil te verwijderen op basis van de grootte en gebruik vervolgens de voorwaartse verstrooiingshoogte versus het histogram van het voorwaartse spreidingsgebied om celaggregaten uit te sluiten en afzonderlijke cellen te selecteren. Herhaal de uitsluiting met de zijverstrooiingshoogte versus het histogram van het zijverstrooiingsgebied.Sluit de dode cellen uit door de positieve cellen uit te splitsen voor DAPI. Selecteer de afstamming negatieve TUR één 19 negatieve kit positieve cellen. En selecteer hieruit een subpopulatie die CD 55 uitdrukt.Verdeel de afstamming negatieve TUR één 19, negatieve kit, positieve, CD 55 positieve cellen in 2 principepopulaties, P zes en P zeven op basis van de expressie van CD 49 F en CD 1 0 5. Nu gebaseerd op de uitdrukking van CD 150 en CD 41, onderverdelen P 6 verder in P 3, P 4 en P 5. Op dezelfde manier, op basis van de expressie van CD 150 en CD 71, onderverdelen P 7 verder in BFUE, vroege CFUE en late CFUE.In deze studie werden burst- en kolonievormende eenheden geïdentificeerd uit de vers geoogste beenmerg- en miltcellen. Na 72 uur erytropoëtine of voertuiginjectie bij de muizen, toonde erytropoëtinestimulatie uitbreiding van de vroege en late kolonievormende eenheidspopulaties P 1 laag en P 1 hoog in de geoogste beenmerg- en miltcellen. De respons van beenmerg- en miltvoorlopers op erytropoëtine in vivo toonde ook een hoger aantal cellen in de vroege en late kolonievormende eenheidspopulaties P 1 laag en P 1 hoog, vergeleken met voertuigcontrole.Het is het belangrijkst om een eencellige suspensie te genereren voordat de cellen met antilichamen worden geëtiketteerd. Dit kan worden bereikt door herhaaldelijk op en neer te pipetteren en door EDTA in de buffer op te nemen. De gezuiverde voorlopers kunnen worden gebruikt voor elke stroomafwaartse cellulaire en moleculaire analyse.Bijvoorbeeld single cell transcriptomics, atesque, colony assays en biochemie. Deze techniek hielp ons bij het testen van de voorspellingen van single cell RNA-seq experimenten.
