赤血球形成は、早期赤血球形成と赤血球終末分化の2つの主要な段階に分けることができます(図1)1,2,3。初期の赤血球形成では、造血幹細胞は赤血球系にコミットし、半固体培地でのコロニー形成の可能性に基づいて1970年代に最初に同定された初期の赤血球前駆細胞を生じさせます4,5,6,7,8,9 .大まかに言えば、赤血球前駆細胞は2つのカテゴリーに分けられます:それぞれが「バースト形成ユニットエリスロイド」またはBFU-e 4,5,6と呼ばれる「バースト」(より小さな赤血球細胞クラスターの大きな集合体)を引き起こす初期の前駆細胞。そしてそれらの子孫は、それぞれが単一の小さな赤血球細胞クラスターまたはコロニーを形成し、「コロニー形成ユニット赤血球」またはCFU-e 7,8,9と名付けられる。BFU-eおよびCFU-eはまだ末端赤血球遺伝子を発現しておらず、形態学的に認識できません。CFU-eは、自己複製または増殖細胞分裂を数回行った後、グロビンなどの赤血球遺伝子が誘導される転写スイッチを受け、赤血球末端分化(ETD)に移行します1,10。ETDの間、赤芽球は網状赤血球を形成するために核形成する前に3〜5回の成熟細胞分裂を受け、赤血球に成熟します。
末端分化中の赤芽球は、もともとその形態に基づいて、前赤芽球、好塩基球、多色、およびオルソクロマティックに分類されていました。フローサイトメトリーの出現により、細胞サイズ(前方散乱、FSCで測定)と2つの細胞表面マーカーCD71およびTer11911、12、13に基づく将来の選別と分離が可能になりました(図1)。これおよび同様のフローサイトメトリーアプローチ14は、ETDの分子的および細胞的側面の研究に革命をもたらし、in vivoおよびin vitroでの赤芽球の発生段階特異的分析を可能にしました10、15、16、17、18、19、20。CD71/Ter119アプローチは現在、赤血球前駆体の分析に日常的に使用されています。
最近まで、マウス組織からCFU-eおよびBFU-eを直接高純度で前向きに単離するための同様のアクセス可能なフローサイトメトリーアプローチは、研究者には知られていませんでした。代わりに、研究者らは、多くの場合、同じフローサイトメトリーサブセット内で共精製する非赤血球細胞の存在下で、これらの前駆細胞のほんの一部のみを単離するフローサイトメトリー戦略を使用してきました21。その結果、BFU-eおよびCFU-eの調査は、初期の骨髄前駆細胞からBFU-eおよびCFU-eを誘導および増幅するin vitro分化系に限定されていました。次いで、CFU-eをBFU-eから区別するフローサイトメトリー戦略をこれらの赤血球前駆細胞富化培養物に適用することができる22,23。別のアプローチでは、妊娠中期にマウス胎児肝臓のTer119陰性画分が非常に濃縮されている胎児CFU-eおよびBFU-eを利用します10,24,25。しかしながら、これらのアプローチのいずれも、in vivoでの生理学的状態における成人のBFU-eおよびCFU-eの調査を可能にしない。コロニー形成アッセイに基づいて、これらの細胞がそれぞれわずか0.025%および0.3%の頻度で成体骨髄に存在することを思い出すと、課題の大きさが理解されるかもしれません6。
ここで説明するプロトコルは、採取したばかりのKit+マウス骨髄細胞のシングルセルトランスクリプトミクス解析に基づく新しいフローサイトメトリーアプローチです(Kitは骨髄の初期前駆細胞集団のすべてによって発現されます)1。私たちのアプローチには、Pronkらによってすでに使用されていたいくつかの細胞表面マーカーが含まれています21,26。シングルセルトランスクリプトームを使用して、赤血球および他の初期造血前駆細胞を同定する細胞表面マーカーの組み合わせを決定しました(図2)。具体的には、系統陰性(Lin-)Kit+細胞のCD55+画分を5つの集団に細分することができ、そのうち3つは赤血球軌道の連続したセグメントを生成します(図2)。これらの各集団のトランスクリプトーム同一性は、ソート、続いてscRNAseq、およびソートされた単一細胞トランスクリプトームの元のトランスクリプトームマップへの投影によって確認されました(5つの集団のそれぞれおよび骨髄データセット全体の遺伝子発現は https://kleintools.hms.harvard.edu/paper_websites/tusi_et_al/index.html で探索できます)1.各集団の細胞運命の可能性は、従来のコロニー形成アッセイ(図2)および新規のハイスループット単一細胞運命アッセイ1,27を用いて確認された。これらの解析は、新しいフローサイトメトリーアプローチにより、新鮮な成人骨髄および脾臓のすべてのBFU-eおよびCFU-e前駆細胞の高純度分離が得られることを示しています。具体的には、集団1(P1)にはCFU-eのみが含まれ、他の造血前駆細胞は含まれず、集団2(P2)には骨髄のBFU-e前駆細胞と少数のCFU-eがすべて含まれ、他の前駆細胞は含まれていません1。以下の詳細なプロトコルは、生理食塩水または赤血球生成刺激ホルモンエリスロポエチン(Epo)のいずれかを注射したマウスにおける実験例でさらに説明される。
