二次代謝産物として、ピロリジジンアルカロイド(PA)は草食動物、昆虫、および病原体から植物を保護します1,2。これまでに、異なる構造を持つ660を超えるPAおよびPA-N酸化物(PANO)が、世界中の6,000を超える植物種で発見されています3,4。PA生産植物は、主にキク科、ボラギナ科、ブナ科、およびキク科5,6に見られます。PAは不安定なデヒドロピロリジジンアルカロイドに容易に酸化され、強い求電子性を持ち、DNAやタンパク質などの求核剤を攻撃し、肝細胞の壊死、静脈閉塞、肝硬変、腹水、およびその他の症状を引き起こす可能性があります7,8。PA毒性の主な標的器官は肝臓です。PAはまた、肺、腎臓、およびその他の臓器毒性および変異原性、発がん性、および発生毒性を引き起こす可能性があります9,10。
PAを含む伝統的なハーブ、サプリメント、お茶の摂取、または牛乳、蜂蜜、肉などの食品の間接的な汚染(PAを含む放牧の摂取による毒性)から、人間と動物の中毒の症例が多くの国で報告されています11,12,13。欧州食品安全機関(EFSA)の調査結果は、(ハーブ)ティーなどの物質がPA / PANOへの人間の曝露の重要な原因であることを示しています14。茶サンプルはPAを生成しませんが、PA産生植物は茶園で一般的に見られます(例:エミリアソンチフォリア、セネシオアングラタス、およびアゲラタムコニゾイデス)15。以前は、お茶がピッキングおよび加工中に生産工場からのPAで汚染されている可能性があると疑われていました。しかし、PAはいくつかの手摘み茶葉(すなわち、PA生産プラントがない)でも検出され、他の経路または汚染源があるに違いないことを示唆しています16。ラグワート(セネシオ・ジャコバエア)とメリッサ(メリッサ・オフィシナリス)、ペパーミント(ハッカ・ピペリタ)、パセリ(ペトロセリナム・クリスプム)、カモミール(マトリカリア・レキューティタ)、キンレンカ(トロパエオラム・マジュス)の共栽培実験を行い、その結果、これらすべての植物でPAが検出されました17。PAは実際に土壌18,19を介して生きている植物間で移動および交換されることが確認されています。Van Wykら20は、ルイボスティー(Aspalathus linearis)が雑草が豊富な場所でひどく汚染されており、同じ種類と比率のPAを含んでいることを発見しました。しかし、雑草のない場所のルイボスティーからPAは検出されませんでした。
現在、高い選択性と感度を備えた超高速液体クロマトグラフィータンデム質量分析(UPLC-MS / MS)は、農産物および食品中のPAの定性的および定量的分析に広く使用されています21,22。サンプル処理方法は通常、固相抽出(SPE)または複雑な食品マトリックス抽出物のQuEChERS(クイックイージーで安価で効果的な頑丈な安全)クリーンアップのいずれかで構成され、可能な限り最高の感度を得ることができます12,19。しかし、土壌、雑草、新鮮な茶葉などの複雑なマトリックス中のPAの検出と定量を可能にする堅牢な分析方法はまだ不足しています。
本研究では、乾燥茶試料、生茶葉、雑草、雑草根圏土壌試料中の15種類のPAを、UPLC-MS/MSと吸着剤精製法を組み合わせて分析した。さらに、中国安徽省金寨茶園の5つのサンプリングサイトから15対の雑草および雑草根圏土壌サンプル15ペアと60生茶葉サンプルを収集し、15のPAについて分析しました。これらの結果は、お茶の品質と安全性を確保するために、お茶のサンプル中のPA(汚染)の発生源と経路に関する調査方法といくつかの情報を提供することができます。

有毒なピロリジジンアルカロイド(PA)はお茶のサンプルに含まれており、人間の健康に脅威をもたらします。しかし、茶サンプル中のPA汚染の原因と経路は不明のままです。本研究では、UPLC-MS/MSと組み合わせた吸着剤法を開発し、雑草Ageratum conyzoides L.、A. conyzoides 根粒球土壌、新鮮な茶葉、および乾燥茶サンプル中の15のPAを測定しました。平均回収率は78%〜111%の範囲であり、相対標準偏差は0.33%〜14.8%であった。中国安徽省の金寨茶園から15組のコニゾイデスとコニゾイデスの根圏土壌サンプル60枚を採取し、15個のPAについて分析しました。インターメジン-N-オキシド(ImNO)とセネシオニン(Sn)を除いて、15のPAすべてが新鮮な茶葉で検出されたわけではありません。ImNO(34.7μg/kg)の含有量はSn(9.69μg/kg)よりも多かった。また,ImNOとSnはいずれも茶樹の若葉に集中していたが,その含有量は古い葉では低かった。その結果,茶園ではPA生成雑草-土壌-生茶葉の経路を通って茶類中のPAが移動することが示された。
