さらに、別の考えられるシナリオは、ダニ刺されおよび高レベルの抗α-Gal IgE抗体に対して強いアレルギー反応を示すが、哺乳類の肉の消費に耐性がある患者です12。したがって、哺乳類の肉アレルギーは、特定のタイプのダニ咬傷関連アレルギーである可能性があります。AGSに関連する主なダニ種には、Amblyomma americanum(米国)、Amblyomma sculptum(ブラジル)、Amblyomma testudinariumおよびHaemaphysalis longicornis(日本)、Ixodes holocyclus(オーストラリア)、およびIxodes ricinus(ヨーロッパにおけるライムボレリア症の主なベクター)が含まれます11,24。
マダニ刺されに関連するIgE産生を評価するために使用された唯一のモデルは、他の哺乳類と同様に、マウスもタンパク質および脂質上にα-Galを発現し、α-GalにIgEを産生しないため、α−1,3−ガラクトース転移酵素ノックアウト(α−Gal KO)マウス25,26の遺伝子で遺伝子改変されたマウスモデルである。しかし、ゼブラフィッシュ(Danio rerio)は、哺乳類と解剖学的に多くの類似点を共有し、人間と同様にα-Galを合成することができないため、哺乳類に適用される生物医学研究に有用なモデルです。α-Galはゼブラフィッシュで自然に生成されないため、手頃な価格のモデルであり、操作が容易で、α-Gal関連のアレルギー反応の研究に高いサンプルサイズを可能にします。
この研究では、ゼブラフィッシュをモデル生物として使用して、局所アレルギー反応、行動パターン、およびダニ唾液26,27およびその後の哺乳類肉消費に対する経皮感作への反応に関連する分子メカニズムを特徴付け、説明しています。この目的のために、魚は皮内注射によってダニの唾液にさらされ、次にα-Gal27を含む動物の使用に適した哺乳類の肉由来製品を含む犬の飼料を与え、次に関連するアレルギー反応の可能性を評価します。この方法は、アレルギープロセスに関連する他の生体分子、特にAGSに関連する生体分子の研究に適用できます。

問題は、ダニ刺されにさらされた一部の個人だけがアレルギー反応を発症する理由です。私たちにとって、マダニの唾液中の生体分子とそれに伴う免疫機構を解明することは、この問いに取り組むための鍵となります。α-Gal症候群のゼブラフィッシュ動物モデルは、ダニの唾液に反応して人間の行動パターンとアレルギー反応を再現します。この実験的アプローチは、α-Gal症候群に関連する葛藤と、病原体感染に対する防御であるα-Galに対する抗体応答に関連する協力を伴うダニ-宿主分子相互作用の理解を進めます。ダニの唾液で魚を処理するには、成体のゼブラフィッシュを性別のバランスのとれた3つのグループに分配します。市販のGala1-3Gal-BSA 3またはalpha-Galをポジティブコントロールとして使用し、PBSをネガティブコントロールとして使用します。唾液の抽出には、モルモットに6〜7日間給餌された半充血した病原体のない雌ダニを使用します。0.33ミリメートルの針を有する50マイクロリットルの注射器を使用して、pH 7.4のPBS中の塩酸ピロカルピンの2%溶液5マイクロリットルでダニをヘモコエルに処理する。マイクロピペットに取り付けられた10マイクロリットルのチップを使用して、ダニのハイポストームから唾液を採取します。氷の上の1.5ミリリットルのチューブに唾液を入れ、摂氏マイナス80度で保存します。唾液タンパク質濃度を決定し、製造元の推奨に従って、BCAタンパク質アッセイキットを使用して魚に注入するタンパク質の量を確立します。ゼブラフィッシュを摂氏27度のフロースルー水システムで14〜10時間の明暗サイクルで維持します。次に、雌と雄の比率が似ており、注射用の体重が似ているグループごとに10匹の魚を選択します。0.02%トリカインメタンスルホン酸塩に浸して魚を短時間麻酔します。.麻酔をかけた魚を鉗子または手を使用して慎重に、濡れたスポンジの上に置き、尾びれを右側にして、病変を制御するために同じ方向に化合物を注入します。尾びれに対して5ミリメートル、魚の体に対して45度の角度で1センチメートル、29ゲージの針を取り付けた100マイクロリットルの注射器を使用して、魚のグループを筋肉の皮内に注入します。注射後、処理した魚を麻酔なしで淡水タンクに戻して回復させます。午前9時30分に毎日2回魚に餌をやる午後1時30分、2日目まで魚1匹あたり50〜70マイクログラムの乾燥魚の餌を与えます。次に、ドッグフードを乳鉢と乳棒でマッシュアップして、治療注射後2日目から実験終了8日目まで魚に餌を与えます。サンプル収集のために、安楽死させた魚をピンでパラフィンプレートに固定します。えらがまだ血液で洗浄されているときに、1センチメートル、29ゲージの針を取り付けた0.5ミリリットルの注射器を使用して、魚のえら血管から血清を収集します。採取した血清は、使用するまで20°Cで1.5ミリリットルのチューブに保管してください。メスの刃で魚を矢状に切り、内部病変を評価します。それから各魚の腸と腎臓を別々の1.5ミリリットルのチューブに集めます。RNA精製キットを使用して、腸と腎臓からトータルRNAを抽出します。製造元の指示に従って、免疫応答に関連する遺伝子の発現を分析し、RT-qPCR用の逆転写ミックスを使用して定量的逆転写ポリメラーゼ連鎖反応、またはRT-qPCRを実行します。ゼブラフィッシュGAPDHに対してmRNA cT値を正規化し、不等分散のスチューデントt検定を使用してグループ間で比較します。血清サンプル中のα-Galを認識するIgM抗体価をELISA法で測定します。プレートリーダーを用いて抗体価を光学濃度450ナノメートルの値として記録し、不等分散のスチューデントt検定を用いて群間で比較する。このゼブラフィッシュモデルは、ダニの唾液に対する宿主の反応によるさまざまなアレルギー反応の特性評価と評価を可能にし、アルファギャル症候群(AGS)におけるそれらの意味合いを可能にします。また、対照魚と比較して、ダニ唾液処理魚では、ゆっくり泳ぐ、水槽の底に横たわる、食べない、振動する、ジグザグに動くなどの行動の変化が観察されました。ダニの唾液で処理された魚ではアレルギー反応の有意な発生率が観察され、急速な脱感作と耐性を示しました。行動の変化は、α-Galのみよりもダニの唾液で処理された魚でより顕著でした。代表的な免疫マーカーの発現のさらなる分析をRT−PCRによって行った。結果は、腎臓のゼブラフィッシュ群間の違いを示し、対照群と比較して、唾液とα-Galで処理された魚で免疫応答マーカーがダウンレギュレーションされました。さらに、唾液とα-Galで処理したゼブラフィッシュは、PBSで処理した魚よりも高いレベルを示すα-Galに対するIgM抗体を開発しました。治療の注射、アレルギー反応と行動の評価、アレルギー反応による死んだ魚の数の報告は、プロトコルの最も重要な側面です。mRNA、タンパク質、さらにはマイクロバイオームレベルでオミクス技術を使用してダニの唾液に対する応答を特徴付けることで、このプロセスに関与する主要な生体分子を特定することができます。
