精巣と卵巣は、性ホルモンの産生に関与する主要な器官です。テストステロンとエストロゲンの産生につながるホルモン伝達のカスケードは、視床下部でゴナドトロピン放出ホルモン(GnRH)の放出とともに始まる、よく特徴付けられたプロセスです1。GnRHの放出は、下垂体からの黄体形成ホルモン(LH)と卵胞刺激ホルモン(FSH)の放出を引き起こします。これらのホルモンが血流に入ると、体内の他の組織に影響を与えます。LHの主な標的は、精巣(男性)と卵巣(女性)です2。LHに反応して、精巣はテストステロン3を産生し、放出します。同様に、卵巣はエストロゲン4を産生します。これらのホルモンの意図された効果は、受精のために細胞と体を準備し、生殖器系の機能を確保することですが、他の多くの身体システムが影響を受ける可能性があります。
性ホルモンは、いくつかの生理学的機能に関連しています。例えば、エストロゲンは破骨細胞による骨の吸収を防ぐことで、骨の恒常性を維持するのに役立ちます。このため、卵巣摘出マウスモデルを用いて、骨粗鬆症などの骨疾患の生理学を研究することができます5,6,7。テストステロンとエストロゲンは、多くの心血管疾患や神経変性疾患の研究対象でもあります。最近では、テストステロン産生の上昇と高脂肪食が血管の酸化ストレスと関連していることがわかっています8。脳では、卵巣摘出術後のLHの変化が空間記憶の変化を引き起こしている9。卵巣摘出術後のエストロゲンの減少は、アポトーシスを誘発し、記憶障害を引き起こす可能性があるため、海馬の細胞死を研究するためのモデルシステムにもなっています10。テストステロンは、腎臓移植後のマウスモデルとヒトの両方で腎臓の成長にも関与していることが示されています11。
ホルモン欠乏マウスモデルの作成により、性ホルモンとそのホルモンカスケードがさまざまな疾患や組織に蓄積される様子を研究することができます。これは、精巣の外科的切除(精巣摘出術)または卵巣切除術(卵巣摘出術)によって達成できます。この手順は、離乳年齢(21日)または成人年齢の若いマウスで実行できます。卵巣摘出術は雌マウスで行われ、精巣摘出術は雄マウスで行われます。これらの器官を取り除くことにより、エストロゲンとテストステロンのレベル、およびプロゲステロンなどのそれらの誘導体の多くは、大幅に減少させることができます12,13。マウスで精巣摘出術または卵巣摘出術を行うプロセスは、適切な技術で迅速かつ低侵襲にすることができます。これらの臓器を安全かつ効率的に迅速に切除することで、正しく行えば100%の生存率が得られ、マウスの数を最小限に抑えながら、迅速な外科的処置が可能になります。ここでは、精巣と卵巣の迅速な切除のためのプロトコルを詳述し、研究者がこの手術を迅速かつ安全に実施できるようにするための適切な術後モニタリングを示します。また、この手術を行う際に外科医に解剖学的ランドマークを提供するために、性器や周囲の組織の視覚的な例も含まれています。
