マウスの左動脈の永久結紮の外科モデルは、心筋梗塞後の病態生理学および関連する炎症反応を調べるために使用することができる。この方法は、MIの信頼性が高く生産可能なマウスモデルを提供し、簡単で安全で侵襲性が低い。まず、ビーズ滅菌器を摂氏250度に予熱し、オートクレーブ滅菌した手術器具を数分間入れます。しっかりとしたつま先のつまみに対する反応をチェックして、マウスの麻酔の深さを確認してください。マウスの体重を量って、術前の鎮痛薬ブプレノルフィンの投与量を推定し、腹腔内に注射します。電気かみそりを使用して胸部の左側の毛皮を整えます。手術部位を消毒した後、マウスを傾斜したボード上の仰臥位に置きます。マウスの頭と手足を、それぞれ上顎切歯と粘着テープに取り付けられたゴムバンドを使用して固定します。麻酔下での乾燥を防ぐために、滅菌眼科用潤滑剤を目に塗布します。顎を開き、舌を口腔からそっと引き出します。喉頭鏡を使用して口腔咽頭を照らすことによって喉頭の開口部を識別します。24ゲージのカテーテル針から約0.5センチメートルを切り取り、鈍い針をプラスチックシールドに挿入します。プラスチック製のシールドが付いた鈍い針を気管に向けます。プラスチックシールドを気管に残して、針を取り出します。人工呼吸器を毎分137拍の呼吸数に設定し、この研究で使用したマウスと一回換気量0.18の閉鎖能力に最適化します。人工呼吸器チューブをカテーテルシールドに接続し、人工呼吸器と同期した胸の動きを探して、正しい挿管を確認します。呼吸用保護チューブをカテーテルシールドから外し、予熱された温度制御された手術用ボード上で動物を仰臥位に置きます。マウスを人工呼吸器に再接続します。手術部位を消毒した後、鉗子を使用して皮膚をそっと持ち上げ、手術用ハサミを使用して左大胸筋と小胸筋の間の線に沿って小さな皮膚横切開を行います。下にある胸筋を鉗子と解剖ハサミで分離します。3番目の肋間腔を切開した後、開創器を使用して肋骨を静かに伸ばし、左心室を露出させます。心膜脂肪を脇に動かし、左心房の端から心臓の頂点に向かって走る左冠状動脈を見つけます。パス 8-0針ホルダーの助けを借りて左冠状動脈の下にナイロン縫合糸。左冠状動脈を二重結び目とそれに続く2番目の結び目でレゲートします。開創器を取り外し、22ゲージのカテーテル針を胸腔に挿入します。プラスチックシールドの先端を胸腔に残して、針を取り外します。4-0ナイロン縫合糸を使用して胸郭を閉じます。シリンジを22ゲージのプラスチックシールドに接続し、胸部を軽く押して負圧を確立し、胸腔に閉じ込められた余分な空気をゆっくりと取り除きます。プラスチックシールドを取り外します。イソフルランの供給をオフにし、酸素を供給する人工呼吸器にマウスを置きます。マウスの自発呼吸が1回始まり、人工呼吸器の電源を切ります。マウスをヒートランプの下に置き、起動するまで監視します。動物は、胸骨横臥を維持するのに十分な意識を回復するまで放置しないでください。痛みや不快感の兆候がないかマウスを毎日監視します。手術後さらに2日間、6〜8時間ごとにブプレノルフィンの腹腔内注射を継続します。.心エコーデータを取得する前に、アクティブなECGおよび呼吸信号を検証しました。心臓機能パラメータの心エコー測定は、左冠動脈結紮の28日後に認められました。SHAMおよびMI心臓の腓骨短軸図のMモード画像が観察された。MI画像は、左冠動脈結紮後の心臓壁運動の欠陥を示しました。左心室質量の増加、駆出率の減少、心拍出量の減少などの心不全の指標は、SHAM群と比較してMI群で観察されました。マッソンの下脳室、中脳室、上心室のトリクローム染色は、梗塞した心臓のコラーゲン染色の増加を示し、線維症の増強を示しました。ほとんどの場合、動脈は実行可能ではありません。しかし、左心房の縁から2〜4ミリメートル下の心筋組織を結紮すると、動脈の効率的な結紮が得られます。
