MI는 일반적으로 취약한 플라크의 파열로 인한 관상동맥의 혈전성 폐색에 의해 발생한다3. 결과적인 허혈은 영향을받는 심근의 심오한 이온 및 대사 변화뿐만 아니라 수축기 기능의 급격한 감소를 유발합니다. 심근경색은 심근 세포의 사멸을 초래하며, 이는 심실 기능 장애와 심부전을 유발할 수 있다4.
MI 환자에서 MI에 대한 연구는 MI5 환자로부터 얻은 조직의 부족으로 인해 제한적입니다. 이와 같이, MI의 쥐 모델은 질병 기전을 연구하고 잠재적인 치료 표적을 개발하는 데 유용합니다. MI의 현재 이용 가능한 쥐 모델에는 비가역적 허혈 모델(LCA 및 절제 방법) 및 재관류 모델(허혈/재관류, I/R)이 포함됩니다6. 마우스에서 좌관상동맥(LCA)의 영구 결찰이 가장 많이 사용되는 방법이며, 환자에서 MI의 병태생리학 및 면역학을 모방한다 7,8,9. 영구적인 MI는 전기적 손상이나 냉동 손상을 수반하는 절제 방법에 의해서도 유도될 수 있습니다. 절제 방법은 정확한 위치(10)에서 균일한 크기의 경색을 발생시킬 수 있다. 반면에, 흉터 형성, 경색 형태 및 분자 신호 전달 메커니즘은 절제 방법10,11에 따라 다를 수 있다. 쥐 I/R 방법은 재관류 요법의 임상적 시나리오를 나타내기 때문에 또 다른 중요한 MI 모델이다12. I/R 모델은 다양한 경색 크기, 초기 손상의 반응을 구별하는 어려움, 재관류6와 같은 문제와 관련이 있다.
LCA 결찰법은 널리 사용되고 있지만, 낮은 생존율과 수술 후 통증과 관련이 있다13. 이 프로토콜은 마우스의 준비 및 삽관, LCA 결찰, 수술 후 관리 및 MI의 검증을 포함하는 LCA 결찰의 쥐 수술 MI 모델을 입증한다. 침습적 기관 절개술14을 사용하는 대신, 이 방법은 기관내 삽관을 사용한다. 동물은 후두경을 사용하여 구강 인두를 비추어 삽관하므로 절차가 더 쉽고 안전하며 외상이 적습니다15. 마우스는 시술 내내 인공호흡기 지지대와 이소플루란 마취 하에 유지됩니다. 또한, 심초음파와 Masson's trichrome 염색을 수행하여 MI 후 심장 기능과 심장 섬유증을 각각 평가합니다. 전반적으로, 이 방법은 MI 후 병태생리학 및 염증을 연구하는 데 사용할 수 있는 MI의 신뢰할 수 있고 재현 가능한 외과적 쥐 모델을 제공합니다.

허혈성 심장 질환 및 그에 따른 심근 경색(MI)은 미국 및 전 세계에서 사망의 주요 원인 중 하나입니다. 심근경색 후 병태생리학적 변화를 탐색하고 향후 치료법을 설계하기 위해서는 MI의 연구 모델이 필요합니다. 마우스에서 좌관상동맥(LCA)의 영구 결찰은 MI 후 심장 기능 및 심실 리모델링을 조사하는 데 널리 사용되는 모델입니다. 여기에서 우리는 LCA의 영구 결찰에 의한 덜 침습적이고 신뢰할 수 있으며 재현 가능한 외과적 쥐 MI 모델을 설명합니다. 우리의 수술 모델은 쉽게 가역적인 전신 마취, 기관 절개술이 필요하지 않은 기관내 삽관 및 개흉술로 구성됩니다.
