많은 염증성 질환은 부종이나 부상 부위에 호중구가 침윤하는 것이 특징이다1. 이러한 조직에 침투하기 위해 호중구는 내피에 대한 체포, 혈관 벽을 통한 유출 및 조직 내로의 모집을 포함하는 호중구 모집 캐스케이드를 완료해야 합니다2. 순환 호중구는 특히 정지 단계에서 이 캐스케이드를 완료하기 위해 β2 인테그린 활성화가 필요합니다. 따라서 호중구 부착, 외래 및 모집을 감소시키는 인테그린 억제 약물은 염증성 질환을 효과적으로 치료할 수 있습니다 3,4.
β2 인테그린은 이전에도 염증성 질환의 표적이 된 적이 있습니다. 인테그린 αLβ2를 직접 표적으로 하는 단클론 항체인 에팔리주맙(Efalizumab)은 건선5를 치료하기 위해 개발되었습니다. 그러나 에팔리주맙은 JC 바이러스 재활성화로 인한 진행성 다발성 백질뇌병증이라는 치명적인 부작용으로 인해 철회되었다 6,7. 새로운 항염증 인테그린 기반 요법은 부작용을 최소화하기 위해 백혈구의 항감염 기능을 유지하는 것을 고려해야 합니다. 에팔리주맙의 부작용은 혈류 내 단일클론항체의 장기적 순환으로 인해 발생할 수 있으며, 이는 장기적으로 면역 기능을 저해할 수 있다8. 최근 연구에 따르면 에팔리주맙은 αLβ2 교차결합과 α4 인테그린의 원치 않는 내재화를 매개하여 부작용에 대한 대안적 설명을 제공한다9. 따라서 수명이 짧은 저분자 길항제는 이 문제를 피할 수 있습니다.
인간 호중구를 사용하여 저분자 β2 인테그린 길항제를 스크리닝하는 고처리량 분석법이 여기에 제시되어 있습니다. β2 인테그린 활성화는 리간드에 접근하고 결합 친화도를 높이기 위해 인테그린 엑토도메인의 구조적 변화를 필요로 합니다. 표준 스위치블레이드 모델에서, 벤트-클로즈드 인테그린 엑토도메인(benent-closed integrin ectodomain)은 먼저 확장-폐쇄 형태(extended-closed conformation)로 연장된 다음, 완전히 활성화된 확장-개방 형태(10,11,12,13)로 그 헤드피스를 개방한다. 구부러진 닫힘에서 구부러진 열림 및 확장 열림으로 시작하여 결국 14,15,16,17,18,19로 시작하는 대체 경로도 있습니다. 형태 특이적 항체 mAb24는 엑토도메인의 헤드피스가 열려 있을 때 인간 β2-I 유사 도메인의 에피토프에 결합합니다(20,21,22,23).

A Flow Cytometry-Based High-Throughput Technique for Screening Integrin-Inhibitory Drugs
