手术方法是使用最广泛的技术;该方法涉及通过视神经5固定眼睛，然后进行解剖。这种做法很艰巨，可能会损害任何脆弱的组织，因为球眼在解剖过程中继续移动。尽管该技术有利于分离视网膜层的各个部分，但该技术无法在解剖时划定组织的空间方向。

在我们的实验室中，该程序已在任何性别的 P28 的 CBA/J 或 rd1 小鼠品系上进行。该程序可以对动物的任何品系，年龄或性别进行，并且根据这些特征没有偏差。
这些动物是从商业来源采购的（见 材料表），并在国家免疫学研究所（NII）的小动物设施（SAF）进行维护。他们被关在单独的通风笼子（IVC）中， 并随意获得 酸化的高压灭菌水和食物。它们保持在21-23°C，光照/黑暗循环为14小时/ 10小时。
