Time-lapse live-cell SRS-microscopie is een alternatieve beeldvormingstechniek voor het volgen van moleculen op een labelvrije manier. In vergelijking met fluorescentie-etikettering is SRS-beeldvorming vrij van fotobleken, waardoor moleculen op lange termijn kunnen worden gemonitord. Tot op heden is het live cell imaging-systeem op een rechtopstaande SRS-microscopie echter niet in de handel verkrijgbaar. Optische microscopie wordt gebruikt om de microstructuren van monsters te observeren. Optische beeldvorming is snel, minder invasief en minder destructief dan andere technologieën1. Live-cell imaging met optische microscopie is ontwikkeld om de dynamiek van gekweekte levende cellen over een lange periode vast te leggen2. Verschillende soorten optische contrasten bieden verschillende informatie over biologische monsters. Optische fasemicroscopie toont bijvoorbeeld het subtiele verschil in de brekingsindices in het monster3. Fluorescentiemicroscopie wordt veel gebruikt om specifieke biomoleculen of cellulaire organellen in beeld te brengen. De breedbandexcitatie- en emissiespectra van fluorescentie resulteren echter meestal in spectrale overlapping wanneer meerkleurige beeldvorming wordt uitgevoerd4. Fluorescerende moleculen zijn lichtgevoelig en kunnen worden gebleekt na langdurige, periodieke blootstelling aan licht. Bovendien kan fluorescentielabeling de biodistributie van de moleculen in cellen5 veranderen. SRS-microscopie is een labelvrije chemische beeldvormingstechnologie6. Het contrast van SRS is gebaseerd op de trillingsovergang van specifieke chemische bindingen. De trillingsfrequentie van een chemische binding vertoont vaak een smalle spectrale bandbreedte, waardoor het mogelijk is om meerdere Raman-banden in dezelfde monsters af te beelden7. SRS-microscopie is een uniek hulpmiddel voor beeldvorming van levende cellen en biedt meerdere chemische contrasten op een labelvrije manier8.
Hoewel SRS-beeldvorming van niet-gekleurde cellen voor veel studies is gebruikt, is SRS-beeldvorming van levende cellen op lange termijn niet op grote schaal toegepast. Een reden is dat commerciële open kamers niet direct kunnen worden gebruikt voor SRS-beeldvorming vanwege hun grote dikte 9,10,11,12. Deze kamers met een glazen deksel zijn meestal ontworpen voor brightfield- of fluorescentiebeeldvorming met behulp van een enkel hoog NA-objectief met een achterwaarts detectieschema. SRS-beeldvorming geeft echter de voorkeur aan verzonden detectie met behulp van zowel een hoog NA-objectief als een hoge NA-condensor, waardoor er slechts een zeer korte opening (meestal enkele millimeters) tussen het objectief en de condensor overblijft. Om dit probleem op te lossen, hebben we een flexibele kamer ontworpen met behulp van een zacht materiaal om time-lapse SRS-beeldvorming van levende cellen mogelijk te maken met behulp van een rechtopstaand microscoopframe. In dit ontwerp is het waterdompelobjectief ingesloten in de zachte kamer en kan het vrij bewegen in drie dimensies voor scherpstel- en beeldvormingsdoeleinden.
De optimale temperatuur voor het kweken van de meeste zoogdiercellen is 37 °C, terwijl de kamertemperatuur altijd 10° lager is dan dit. Temperatuur hoger of lager dan 37 °C heeft een dramatisch effect op de celgroeisnelheid13. Daarom is temperatuurregeling van de celkweekomgeving vereist in een live-cell imaging-systeem. Het is bekend dat temperatuurinstabiliteit zal leiden tot onscherpe problemen tijdens langdurige beeldvorming14. Om een stabiele omgeving van 37 °C te bereiken, hebben we een grote behuizingskamer gebouwd om het hele microscoopframe te bedekken, inclusief een thermische isolatielaag onder de microscoop (figuur 1). In de omvangrijke temperatuurregelkamer helpt de kleine flexibele kamer om de fysiologische vochtigheid en pH nauwkeurig te handhaven via de gereguleerde luchtstroom aangevuld met 5% CO 2 (figuur 2). De temperatuur en vochtigheid van de kamers werden gemeten om te bevestigen dat het ontwerp met dubbele kamer de optimale celkweekconditie bood voor celgroei onder langdurige, periodieke SRS-beeldvorming (figuur 3). Vervolgens demonstreerden we de toepassing van het systeem voor time-lapse beeldvorming en het volgen van lipidedruppels (LDs) in SKOV3-kankercellen (figuur 4, figuur 5 en figuur 6).

Stimulated Raman scattering (SRS) microscopie is een labelvrije chemische beeldvormingstechnologie. Live-cell beeldvorming met SRS is aangetoond voor vele biologische en biomedische toepassingen. Langdurige time-lapse SRS-beeldvorming van levende cellen is echter niet op grote schaal toegepast. SRS-microscopie maakt vaak gebruik van een waterdompelobjectief met hoog numeriek diafragma (NA) en een hoge NA-oliedompelcondensor om beeldvorming met hoge resolutie te bereiken. In dit geval is de opening tussen het objectief en de condensor slechts enkele millimeters. Daarom kunnen de meeste commerciële podiumomgevingskamers niet worden gebruikt voor SRS-beeldvorming vanwege hun grote dikte met een stijve glazen afdekking. Dit artikel beschrijft het ontwerp en de fabricage van een flexibele kamer die kan worden gebruikt voor time-lapse live-cell imaging met verzonden SRS-signaaldetectie op een rechtopstaand microscoopframe. De flexibiliteit van de kamer wordt bereikt door een zacht materiaal te gebruiken - een dunne natuurlijke rubberen film. De nieuwe behuizing en het ontwerp van de kamer kunnen eenvoudig worden toegevoegd aan een bestaande SRS-beeldvormingsopstelling. De tests en voorlopige resultaten tonen aan dat het flexibele kamersysteem stabiele, langdurige, time-lapse SRS-beeldvorming van levende cellen mogelijk maakt, die in de toekomst voor verschillende bioimaging-toepassingen kan worden gebruikt.

We fabriceerden en assembleerden het flexibele kamersysteem voor time-lapse SRS-beeldvorming (figuur 1 en figuur 2) en evalueerden vervolgens de prestaties van het systeem. De temperatuur in de omgevingsruimte van de microscoop bereikte binnen 1 uur de verwachte 37 °C, wat de kamertemperatuur niet significant beïnvloedde (figuur 3A). A Flexible Chamber for Time-Lapse Live-Cell Imaging with Stimulated Raman Scattering Microscopy

We rapporteren een stage-top, flexibele omgevingskamer voor time-lapse beeldvorming van levende cellen met behulp van rechtop gestimuleerde Raman-verstrooiingsmicroscopie met verzonden signaaldetectie. De flexibele omgevingskamertechniek maakt time-lapse mogelijk als SRS-beeldvorming met verzonden signaaldetectie op een bedieningsmicroscoopframe. SRS-microscopie gebruikte een objectief met een hoog numeriek diafragma en een high-end condensor, waardoor er slechts een paar millimeter opening overbleef waar een conventionele kamer niet in past. Een flexibele kamer lost dit probleem op.Onderzoekers die SRS-microscopie gebruiken, kunnen deze techniek gemakkelijk toepassen. De flexibele kamer kan ook worden gebruikt op een ontleed- of een steriele microscoop om het monster onder optimale fysiologische omstandigheden te houden. Markeer om te beginnen de locaties van de voeten van het SRS-microscoopframe en het gemotoriseerde podium met behulp van een markeerpen op de optische tafel.Nadat u het microscoopframe en het podium van de optische tafel hebt verwijderd, legt u het siliconenrubberen vel op de tafel. Snijd het siliconenrubber langs de markeringen met een mes. Verwijder kleine rubberen stukjes en plaats de vierkante mica keramische vellen op dezelfde locaties.Beweeg vervolgens het microscoopframe en het podium terug naar de optische tafel en lijn de voeten voorzichtig uit op de mica-keramische vellen langs de markeringslijnen. Nadat u het optische pad van SRS opnieuw hebt uitgelijnd, monteert u de omgevingsbehuizing bovenop de thermische isolatiefundering om het hele microscoopframe te bedekken, met behulp van vijf stukken grote polycarbonaatplaten. Sluit de randen en interfaces van de doos af met aluminiumfolietape.Sluit de flexibele kanaalslang aan op de inlaat en de uitlaatpoorten van de behuizingsdoos om een circulaire, warme luchtstroom mogelijk te maken die door de verwarmingsmodule wordt gepompt en geregeld. Monteer het bewerkte holle cilindrische aluminium stuk 1 met drie stelschroeven op het neusstuk van het objectief. Monteer vervolgens het bewerkte holle cilindrische aluminium stuk 2 met vier schroeven op de monsterhouder.Monteer de monsterhouder met aluminium stuk 2 op het gemotoriseerde podium en monteer deze met schroeven. Plaats de natuurrubberen filmhuls tussen de twee bewerkte aluminium stukken en monteer deze met behulp van elastiekjes aan elk uiteinde. Sluit de gecomprimeerde CO2-cilinder aan op de gasmengermodule met behulp van de juiste buizen en connectoren.Stel de CO2-ingangsdruk in op 20 tot 25 psi. Gebruik de ingebouwde CO2-sensor en een controller om ervoor te zorgen dat de luchtmixermodule 5% CO2 in de luchtstroom kan regelen en mengen. Gebruik inline filters om de luchtstroom op te ruimen.Gebruik de juiste slangen en connectoren om de gemengde lucht naar de voorgesteriliseerde waterfles op de kookplaat te leiden en vervolgens de bevochtigde lucht naar de flexibele kamer te leiden. Zet de kookplaat op 37 graden Celsius. Bel de luchtstroom in het warme water om de luchtvochtigheid van de luchtstroom te verhogen.Koppel het aluminium stuk 2 van de machine los van de monsterhouder door de schroeven te verwijderen. Veeg alle delen van de flexibele kamer schoon met 70% ethanol, inclusief het neusstuk, de monsterhouder en het waterdompelobject. Ontsmet het hele behuizingssysteem met behulp van een UV-lamp die gedurende 20 tot 30 minuten in de behuizing is geplaatst.Laad vervolgens de celkweekschaal. Verwijder de deksel van de schaal en immobiliseer deze met behulp van de klemmen. Verlaag het objectief in de celkweekmedia voor grof scherpstellen.Laat het aluminium stuk 2 zakken om de flexibele kamer te omsluiten en monteer het met schroeven op de monsterhouder. Plaats vervolgens de sensor. Stel de luchttoevoer in met 5% CO2 en 19% O2 voor normale celcultuur met een luchtdebiet van 200 kubieke centimeter per minuut en een temperatuur van 37 graden Celsius.Stem de lasergolflengte af op 805 nanometer om de 2.854 centimeter inverse Raman-verschuiving te richten, die wordt toegeschreven aan de trilling van CH2 chemische bindingen. Gebruik een laag laservermogen om fotoschade aan de cellen te verminderen. Pas het doel aan en focus het om een goede SRS-beeldvorming van de cellen te bereiken met behulp van de focusknop op het hoofdbedieningspaneel van de software.Als u snel wilt scherpstellen, stelt u het pixelnummer in op 512 bij 512 pixels met een pixelverblijftijd van 4,8 microseconden op het configuratiepaneel. Na het bereiken van een goede scherpstelling, stelt u de beeldresolutie in op 2048 bij 2048 pixels voor een gezichtsveld van 175 micrometer in het kwadraat voor het verkrijgen van afbeeldingen van hoge kwaliteit. Controleer de opslagfunctie op het hoofdbedieningspaneel en controleer het SRS-kanaal op het kanalenpaneel.Stel de intervaltijd tussen twee frames in op 180 seconden op het hoofdbesturingskanaal. Stel het acquisitienummer in op 480 voor time-lapse-beeldvorming gedurende 24 uur. Start geautomatiseerde beeldacquisitie met behulp van de lusfunctie op het hoofdbedieningspaneel.De prestaties van het flexibele kamersysteem voor time-lapse SRS-beeldvorming werden geëvalueerd. De temperatuur in de omgevingsbehuizing van de microscoop bereikte de verwachte 37 graden Celsius en had geen significante invloed op de kamertemperatuur. De temperatuur in de flexibele kamer bereikte 37 graden Celsius in 1,5 uur en werd stabiel gehouden gedurende ten minste 24 uur.De relatieve luchtvochtigheid in de flexibele kamer bereikte 85% in één uur en kon minstens 24 uur worden gehandhaafd. De time-lapse SRS-beeldvorming van levende SKOV3-cellen toonde de snelle en actieve beweging van intracellulaire lipidedruppeltjes met een temporele resolutie van drie minuten. Tegen het einde van de 24-uurs beeldvormingssessie vertoonden de cellen nog steeds een normale morfologie en dichtheid, wat aangeeft dat de cellen gezond waren.Vervolgens werden SKOV3-cellen behandeld met oliezuur in beeld gebracht en werd de accumulatie van lipidedruppels gedurende 10 uur gevolgd. De hoeveelheden lipidedruppels werden gekwantificeerd met behulp van de verhouding tussen lipidedruppel en lichaamsoppervlak en de totale SRS-intensiteit van de lipidedruppels. De resultaten geven aan dat de hoeveelheid lipidedruppeltjes gedurende 10 uur bleef toenemen.Gelijktijdige voorwaartse SRS-beeldvorming van lipidedruppels en achterwaartse twee fotonenfluorescentiebeeldvorming van lysosomen gelabeld met een fluorescentiekleurstof DND-189 werd ook aangetoond. Een zeer lage mate van colocalisatie van lipidedruppels en lysosomen werd waargenomen. Houd er bij het proberen van dit protocol rekening mee dat focale drift een veel voorkomend probleem is bij beeldvorming met levende cellen.Heroriëntatie kan nodig zijn na ongeveer twee uur time-lapse beeldvorming. Na de ontwikkeling is time-lapse SRS-beeldvorming gebruikt voor verschillende onderzoeken om labelvrije moleculen of gelabelde moleculen met Raman-tekst in levende cellen te volgen.
