光学顕微鏡は、サンプルの微細構造を観察するために使用されます。光学イメージングは、他の技術よりも迅速で、侵襲性が低く、破壊的ではありません1。光学顕微鏡による生細胞イメージングは、培養生細胞の動態を長期間にわたって捉えるために開発されています2。異なるタイプの光学コントラストは、生物学的サンプルに関する異なる情報を提供します。例えば、光学位相顕微鏡は、試料3全体の屈折率の微妙な違いを示す。蛍光顕微鏡は、特定の生体分子や細胞小器官の画像化に広く使用されています。しかしながら、蛍光の広帯域励起スペクトルおよび発光スペクトルは、通常、多色イメージングが行われるときにスペクトルオーバーラップをもたらす4。蛍光分子は光に敏感であり、長期間の定期的な光曝露後に漂白することができます。さらに、蛍光標識は、細胞5における分子の生体分布を変化させ得る。SRS顕微鏡は、ラベルフリーの化学イメージング技術です6。SRSのコントラストは、特定の化学結合の振動遷移に依存しています。化学結合の振動周波数は狭いスペクトル帯域幅を示すことが多く、同じサンプル内の複数のラマンバンドを画像化することが可能になります7。SRS顕微鏡は、生細胞イメージングのためのユニークなツールであり、ラベルフリーの方法で複数の化学的コントラストを提供します8。
染色されていない細胞のSRSイメージングは多くの研究に使用されていますが、生細胞の長期タイムラプスSRSイメージングは広く採用されていません。その理由の1つは、市販のオープンチャンバーは厚さ9,10,11,12が大きいため、SRSイメージングに直接使用できないことです。ガラス蓋付きのこれらのチャンバーは、主に、後方検出スキームを備えた単一の高NA対物レンズを使用した明視野または蛍光イメージング用に設計されています。ただし、SRSイメージングでは、高NA対物レンズと高NAコンデンサーの両方を使用した透過検出が好まれるため、対物レンズとコンデンサーの間に非常に短いギャップ(通常は数ミリメートル)しか残されません。この問題を克服するために、我々は、正立顕微鏡フレームを使用して生細胞のタイムラプスSRSイメージングを可能にするために、柔らかい材料を使用して柔軟なチャンバーを設計しました。この設計では、水浸対物レンズはソフトチャンバーに封入されており、焦点合わせとイメージングの目的で3次元で自由に移動できます。
ほとんどの哺乳類細胞を培養するのに最適な温度は37°Cですが、室温は常にこれより10°C低くなっています。37°Cより高いまたは低い温度は、細胞増殖速度に劇的な影響を及ぼします13。そのため、生細胞イメージングシステムにおいては細胞培養環境の温度制御が求められている。温度の不安定性は、長期イメージング中に焦点ぼけの問題につながることが知られています14。37°Cの安定した環境を実現するため、顕微鏡下の断熱層を含む顕微鏡フレーム全体を覆う大型の筐体チャンバーを構築しました(図1)。大きな温度制御チャンバー内では、小さな柔軟なチャンバーは、5%CO2を補充した調整された空気の流れを介して生理学的湿度とpHを正確に維持するのに役立ちます(図2)。チャンバーの温度と湿度を測定し、ダブルチャンバー設計が長期の定期的なSRSイメージング下での細胞増殖に最適な細胞培養条件を提供することを確認しました(図3)。次に、SKOV3がん細胞のタイムラプスイメージングと脂肪滴(LD)の追跡へのシステムの応用を実証しました(図4、図5、および図6)。
