관절 연골은 연골 세포와 세포 외 기질1로 구성된 고도로 분화되고 조밀 한 조직입니다. 관절 연골의 표층은 유리질 연골의 한 형태로, 표면이 매끄럽고 마찰이 적으며 강도와 탄성이 좋으며 기계적 응력 내성이 우수합니다2. 세포외 기질은 콜라겐 프로테오글리칸과 물을 포함하며, 타입 II 콜라겐은 전체 콜라겐3의 약 90%를 차지하기 때문에 콜라겐의 주요 구조 성분이다. 연골 조직에는 혈관이나 신경이 존재하지 않기 때문에 손상 후 자가 복구 능력이 부족합니다4. 따라서 외상으로 인한 연골 결손은 항상 클리닉에서 다루기 힘든 관절 질환이었습니다. 또한, 이 관절 질환은 젊은이들을 공격하는 경향이 있으며 전 세계적으로 발병률이 증가하고 있습니다 5,6. 무릎 관절은 연골 결손의 가장 흔한 부위이며, 여기서 결손은 관절 통증, 관절 기능 장애 및 관절 연골 변성을 동반하여 결국 무릎 골관절염(kOA)으로 이어집니다7. 무릎 관절의 연골 결손은 환자에게 경제적, 생리적 부담을 주고 환자의 삶의 질에 심각한 영향을 미친다8. 이 질병은 임박한 해결책이 없는 중대하고 시급한 임상적 문제를 제기합니다. 현재 수술은 연골 결손 치료의 중심이지만 장기적인 결과는 여전히 만족스럽지 않다9.
임상적 연골 결손은 결국 kOA를 유발하고, 따라서, kOA 동물 모델은 연골 결손의 병리학적 연구 및 약물 개발에 통상적으로 사용된다. 동물모델의 확립은 연골 결손 복구의 병태생리학적 과정을 이해하는 데 중요하며, 이는 연골 재생 및 섬유연골과 유리질 연골 사이의 변화를 관찰하는 데 사용할 수 있다10. 그러나, 전방십자인대 절개술(ACLT), 내측 반월판의 불안정화(DMM), 난소절제술(OVX) 및 헐스(Hulth)와 같이 일반적으로 사용되는 kOA 동물 모델은 일반적으로 장기 모델링이 필요하고 병리학적 및 통증 평가만 허용하므로 약물 개발의 효율성에 한계가 있다11. 수술 모델 외에도 모노요오드아세테이트(monoiodoacetate, MIA) 및 파파인 주사와 같은 화학적 모델도 연골 결손을 초래하지만, 결손의 정도를 잘 관리할 수 없고, 임상 현실과는 거리가 멀다11. 충돌(Collision)은 큰 동물의 연골 결손을 모델링하는 또 다른 접근법이지만, 이 방법은 특정 기구의 사용에 따라 달라지며 거의 적용되지 않는다12.
