Traditioneel zijn celoppervlakmarker-gedefinieerde HSC's voorbereid om de functies van HSC's te bestuderen, zoals zelfvernieuwingscapaciteit en multi-potentie 19,20,21. De immunofenotypisch gedefinieerde (Lineage−c-Kit+Sca-1+CD150+CD34−/loFlk2−) HSC-fractie bevat echter twee discrete HSC-populaties: LT-HSCs en ST-HSCs 9,10<sup class="xr...

Hematopoietische stamcellen (HSC's), die zelfvernieuwingsvermogen en multipotentie bezitten, bevinden zich aan de top van de hematopoietische hiërarchie 1,2. In 1988 toonden Weissman en collega's voor het eerst aan dat de isolatie van HSC's van muizen kon worden bereikt met behulp van flowcytometrie3. Vervolgens werd gemeld dat een fractie gedefinieerd door een combinatie van celoppervlakmarkers, Lineage−c-Kit+Sca-1+CD150+CD34−/loFlk2−, alle HSC's in muizen 4,5,6,7,8 bevatte.
Immunofenotypisch gedefinieerde (Lineage−c-Kit+Sca-1+CD150+CD34−/loFlk2−) HSC's (hierna pHSC's) werden voorheen als functioneel homogeen beschouwd. Recente eencellige analyses hebben echter aangetoond dat pHSC's nog steeds heterogeniteit vertonen met betrekking tot hun zelfvernieuwingscapaciteit9,10 en multipotentie11,12. In het bijzonder lijken er twee populaties te bestaan in de pHSC-fractie met betrekking tot hun zelfvernieuwingscapaciteit: hematopoëtische stamcellen op lange termijn (LT-HSC's), die een continu zelfvernieuwingsvermogen hebben, en kortdurende hematopoëtische stamcellen (ST-HSC's), die een tijdelijk zelfvernieuwingsvermogen hebben 9,10.
Tot op heden blijven de moleculaire mechanismen van zelfvernieuwingscapaciteit die LT-HSC's en ST-HSC's onderscheiden slecht begrepen. Het is cruciaal om beide celpopulaties te isoleren op basis van hun zelfvernieuwingscapaciteiten en om onderliggende moleculaire mechanismen te ontdekken. Er zijn ook verschillende reportersystemen geïntroduceerd om LT-HSC's13,14,15 te zuiveren; de LT-HSC-zuiverheid die door elk reportersysteem wordt gedefinieerd, is echter variabel en exclusieve LT-HSC-zuivering is tot op heden niet bereikt.
Daarom zal de ontwikkeling van een isolatiesysteem voor LT-HSC's en ST-HSC's het onderzoek naar zelfvernieuwingscapaciteit in de pHSC-fractie versnellen. In de isolatie van LT-HSC's en ST-HSC's identificeerde een studie met behulp van meerstaps, onbevooroordeelde screening een enkel gen, Hoxb5, dat heterogeen tot expressie komt in de pHSC-fractie16. Bovendien bleek uit beenmerganalyse van de Hoxb5-reportermuizen dat ongeveer 20% -25% van de pHSC-fractie bestaat uit Hoxb5-pos-cellen. Een competitieve transplantatietest met Hoxb5pos pHSCs en Hoxb5 neg pHSCs toonde aan dat alleen Hoxb5pos pHSCs over zelfvernieuwingscapaciteit op lange termijn beschikken, terwijl Hoxb5neg pHSCs hun zelfvernieuwingscapaciteit binnen een korte periode verliezen, wat aangeeft dat Hoxb5 LT-HSC's identificeert in de pHSC-fractie16. 
Hier demonstreren we een stapsgewijs protocol om LT-HSC's en ST-HSC's te isoleren met behulp van het Hoxb5-reportersysteem . Daarnaast presenteren we een competitieve transplantatietest om de zelfvernieuwingscapaciteit van Hoxb5pos/neg pHSCs te beoordelen (figuur 1). Dit Hoxb5 reporter systeem stelt ons in staat om LT-HSC's en ST-HSCs prospectief te isoleren en draagt bij aan het begrip van LT-HSC-specifieke kenmerken.

isolation method for long-term and short-term hematopoietic stem cells

Zelfvernieuwingscapaciteit en multi-lineage differentiatiepotentiaal worden over het algemeen beschouwd als de bepalende kenmerken van hematopoietische stamcellen (HSC's). Talrijke studies hebben echter gesuggereerd dat functionele heterogeniteit bestaat in het HSC-compartiment. Recente eencellige analyses hebben HSC-klonen gemeld met verschillende celloten binnen het HSC-compartiment, die bevooroordeelde HSC-klonen worden genoemd. De mechanismen die ten grondslag liggen aan heterogene of slecht reproduceerbare resultaten zijn weinig begrepen, vooral met betrekking tot de lengte van zelfvernieuwing wanneer gezuiverde HSC-fracties worden getransplanteerd door conventionele immunostaining. Daarom is het van cruciaal belang om dit probleem op te lossen door een reproduceerbare isolatiemethode vast te stellen voor HSC's op lange termijn (LT-HSC's) en HSC's op korte termijn (ST-HSC's), gedefinieerd door de lengte van hun zelfvernieuwing. Met behulp van onbevooroordeelde meerstapsscreening identificeerden we een transcriptiefactor, Hoxb5, die een exclusieve marker van LT-HSC's in het hematopoietische systeem van de muis kan zijn. Op basis van deze bevinding hebben we een Hoxb5-reportermuislijn opgezet en met succes LT-HSC's en ST-HSC's geïsoleerd. Hier beschrijven we een gedetailleerd protocol voor de isolatie van LT-HSC's en ST-HSC's met behulp van het Hoxb5-reportersysteem . Deze isolatiemethode zal onderzoekers helpen de mechanismen van zelfvernieuwing en de biologische basis voor dergelijke heterogeniteit in het HSC-compartiment beter te begrijpen.

Eerder werd de zelfvernieuwingscapaciteit gemeten met behulp van competitieve transplantatietests, waarbij wordt aangenomen dat donor-HSC's hun zelfvernieuwingscapaciteit alleen behouden als multi-lineage donorcellen in het perifere bloed van de ontvanger worden waargenomen17. Bovendien definiëren verschillende rapporten LT-HSC's als cellen die enkele maanden na de tweede beenmergtransplantatie perifere bloedcellen blijven produceren10,18...

We presenteren een stapsgewijs protocol voor de isolatie van lange termijn hematopoëtische stamcellen (LT-HSCs) en korte termijn HSC's (ST-HSCs) met behulp van het Hoxb5 reporter systeem.

De hematopoëtische stamcel- of HSC-isolatiemethode op lange en korte termijn zal onderzoekers helpen de celvernieuwingsmechanismen en biologische basis van heterogeniteit in het HSC-compartiment beter te begrijpen. Een transcriptiefactor, Hoxb5, die een exclusieve marker van HSC's op lange termijn kan zijn, werd geïdentificeerd. Op basis hiervan werd een Hoxb5-reporter muizenlijn opgezet en werden de lange termijn en de korte termijn HSC's met succes geïsoleerd.Om te beginnen met het verzamelen van de donorbeenmergcellen, steriliseer een vijzel en stamper met 70% ethanol en laat ze volledig drogen. Evenwicht die met celkleuringsbuffer bestaande uit calcium- en magnesiumvrije PBS-bevattende supplementen. Doe de botten geïsoleerd van de donormannetje Hoxb5-tri-mCherry-muis in de mortel en voeg drie milliliter van de celkleuringsbuffer toe.Borstel de botten open met de stamper, splits vervolgens de celklonten door voorzichtig pipetteren en breng de celsuspensie door een celzeef van 100 micron over in een buis van 50 milliliter. Herhaal het proces met een verse celkleuringsbuffer totdat de oplossing helder wordt. Meestal is drie keer herhalen voldoende om een duidelijke oplossing op te leveren.Na het isoleren van de dijbenen en het scheenbeen van de CD45.1.2 congene muis, knipt u beide uiteinden van de botten met een scherpe steriele schaar. Met behulp van een 23-gauge naald uitgerust met een spuit van vijf milliliter gevuld met ijskoude celsuspensiebuffer, spoelt u het beenmerg uit in een steriele celkweekschaal met celsuspensiebuffer. Splits de celklonten uit elkaar door voorzichtig pipetteren en filter vervolgens de celsuspensie door een celzeef van 40 micron met behulp van een P1000-pipet.Voor de gating-opstelling bereidt u het monster voor volgens de instructies in de tekst en brengt u 400 microliter ervan over naar een polystyreenreageerbuis met ronde bodem met een 35-micron celzeef snap cap. Bereid het dodecelpersende reagens vóór de analyse voor en voeg het toe aan het monster, volgens de instructies van de fabrikant. Schakel vervolgens de flowcytometer in en start de analysesoftware volgens de instructies van de fabrikant.Druk vervolgens op Laden om te beginnen met het ophalen van de gegevens. Na het uitsluiten van doubletten, dode cellen en afstammingspositieve cellen, gate de afstammingsnegatieve, de c-kit-positieve, Sca-1-positieve fractie. Sluit vervolgens de Flk2-positieve fractie af.Gate vervolgens Hoxb5-positieve of Hoxb5-negatieve pHSC's in de CD150-positieve, CD34-negatieve of lage fractie. Bereid vervolgens een 1,5 millimeter eiwitarme bindingsbuis met 600 microliter calcium en magnesiumvrije PBS, aangevuld met 10% warmte-geïnactiveerde FBS. Om de Hoxb5-positieve of Hoxb5-negatieve pHSC-sortering uit te voeren, stelt u de voorbereide 1,5-milliliter eiwitarme bindingsbuis in op een sorteerapparaat en sorteert u de Hoxb5-positieve of Hoxb5-negatieve pHSC's in de buis met behulp van de eerder genoemde gating-strategie.Gebruik bij de eerste sortering de opbrengst van de sorteerprecisiemodus. Plaats de 96-putplaat met de eerder voorbereide ondersteunende cellen op het stadium van de geautomatiseerde celdepositie-eenheid. Stel vervolgens de 1,5 milliliter eiwitarme bindingsbuis in op de laadpoort van de flowcytometer.Sorteer de Hoxb5-positieve of Hoxb5-negatieve pHSC's in de 96-putplaat met de ondersteunende cellen met behulp van de eerder genoemde gating-strategie. Sorteer 10 Hoxb5-positieve of Hoxb5-negatieve pHSC's om hun zelfvernieuwingscapaciteiten te testen. De flowcytometrieplots van de beenmerganalyse van de donor Hoxb5-tri-mCherry-muizen onthulden dat ongeveer 20 tot 25% van de cellen in de pHSC-fractie gedefinieerd door afstammingsnegatieve, c-kit-positieve, Sca-1-positieve, CD150-positieve, CD34-negatieve of lage, Flk2-negatieve Hoxb5-positieve pHSC's waren.Hier worden de representatieve flowcytometrieplots van de perifere bloedanalyse van de ontvangende muizen weergegeven. Verder onthulde de perifere bloedanalyse van de ontvangende muizen na primaire transplantatie dat hoewel Hoxb5-positieve en Hoxb5-negatieve pHSC's vier weken na transplantatie vergelijkbaar donorchimerisme vertoonden, continue hematopoiese werd waargenomen bij alleen de Hoxb5-positieve pHSC-ontvangers. De Hoxb5-negatieve HSC's begonnen acht weken na transplantatie het vermogen te verliezen om hematopoëtische cellen te produceren.In de secundaire transplantatieanalyse vertoonden alleen de Hoxb5-positieve pHSC-ontvangers een robuuste hematopoiese. Daarentegen werden donorcellen nauwelijks waargenomen in de Hoxb5-negatieve pHSC-ontvangende muizen, wat suggereert dat Hoxb5-negatieve pHSC's het zelfvernieuwingsvermogen verloren binnen 16 weken na primaire transplantatie. Dit protocol duurt meestal negen tot 12 uur, dus het is belangrijk om het monster zoveel mogelijk op vier graden Celsius te houden tijdens de procedures om de levensvatbaarheid van de cel te behouden.
