Traditionnellement, les CSH définies par marqueurs de surface cellulaire ont été préparées pour étudier les fonctions des CSH, telles que la capacité d’auto-renouvellement et la multipuissance 19,20,21. Cependant, la fraction de CSH définie immunophénotypiquement (Lineage−c-Kit+Sca-1+CD150+CD34−/loFlk2−) contient deux populations distin...

Les cellules souches hématopoïétiques (CSH), qui possèdent une capacité d’auto-renouvellement et de multipotence, résident au sommet de la hiérarchie hématopoïétique 1,2. En 1988, Weissman et ses collègues ont démontré pour la première fois que l’isolement des CSH de souris pouvait être réalisé en utilisant la cytométriede flux 3. Par la suite, une fraction définie par une combinaison de marqueurs de surface cellulaire, Lineage−c-Kit+Sca-1+CD150+CD34−/loFlk2−, a été signalée comme contenant toutes les CSH chez les souris 4,5,6,7,8.
Les CSH définies immunophénotypiquement (Lineage−c-Kit+Sca-1+CD150+CD34−/loFlk2−) (ci-après, les CSHp) étaient auparavant considérées comme fonctionnellement homogènes. Cependant, des analyses récentes sur une seule cellule ont révélé que les CSHp présentent toujours une hétérogénéité en ce qui concerne leur capacité d’auto-renouvellement 9,10 et leur multipotence11,12. Plus précisément, deux populations semblent exister dans la fraction des CSHp en ce qui concerne leur capacité d’auto-renouvellement : les cellules souches hématopoïétiques à long terme (CSH-LT), qui ont une capacité d’auto-renouvellement continue, et les cellules souches hématopoïétiques à court terme (CSS-ST), qui ont une capacité d’auto-renouvellement transitoire 9,10.
À ce jour, les mécanismes moléculaires de la capacité d’auto-renouvellement qui distinguent les CSL-LT et les CST-ST restent mal compris. Il est crucial d’isoler les deux populations cellulaires en fonction de leurs capacités d’auto-renouvellement et de découvrir les mécanismes moléculaires sous-jacents. Plusieurs systèmes rapporteurs ont également été introduits pour purifier les LT-HSC13,14,15; cependant, la pureté LT-HSC définie par chaque système rapporteur est variable, et la purification exclusive LT-HSC n’a pas été atteinte à ce jour.
Par conséquent, le développement d’un système d’isolation pour les CSL-LT et les CSS-ST accélérera la recherche sur la capacité d’auto-renouvellement dans la fraction des CSHp. Dans l’isolement des CSH-LT et des CSS-ST, une étude utilisant un criblage non biaisé en plusieurs étapes a identifié un seul gène, Hoxb5, qui est exprimé de manière hétérogène dans la fraction16 des CSp. De plus, l’analyse de la moelle osseuse des souris rapporteures Hoxb5 a révélé qu’environ 20% à 25% de la fraction pHSC est constituée de cellulesPos Hoxb5. Un test de transplantation compétitif utilisant des CSp Hoxb5 pos et des CSp négatives Hoxb5 a révélé que seuls les CSp Hoxb5 pos possèdent une capacité d’auto-renouvellement à long terme, tandis que les CSPp Hoxb5neg perdent leur capacité d’auto-renouvellement en peu de temps, ce qui indique que Hoxb5 identifie les CSH-LT dans la fraction16 des CSPp. 
Ici, nous démontrons un protocole étape par étape pour isoler les LT-HSC et les ST-HSC à l’aide du système de rapporteur Hoxb5 . De plus, nous présentons un test de transplantation compétitif pour évaluer la capacité d’auto-renouvellement des CSp Hoxb5pos/neg (Figure 1). Ce système de rapporteur Hoxb5 nous permet d’isoler de manière prospective les LT-HSC et les ST-HSC et contribue à la compréhension des caractéristiques spécifiques aux LT-HSC.

La capacité d’auto-renouvellement et le potentiel de différenciation multi-lignée sont généralement considérés comme les caractéristiques déterminantes des cellules souches hématopoïétiques (CSH). Cependant, de nombreuses études ont suggéré qu’une hétérogénéité fonctionnelle existe dans le compartiment HSC. Des analyses récentes de cellules uniques ont rapporté des clones de CSH avec des destins cellulaires différents dans le compartiment des CSH, appelés clones de CSH biaisés. Les mécanismes sous-jacents aux résultats hétérogènes ou peu reproductibles sont peu compris, notamment en ce qui concerne la durée de l’auto-renouvellement lorsque des fractions de CSH purifiées sont transplantées par immunomarquage classique. Par conséquent, l’établissement d’une méthode d’isolation reproductible pour les CSH à long terme (CSH-LT) et les CSH à court terme (CSS-ST), définies par la durée de leur auto-renouvellement, est crucial pour surmonter ce problème. En utilisant un dépistage non biaisé en plusieurs étapes, nous avons identifié un facteur de transcription, Hoxb5, qui pourrait être un marqueur exclusif des CSL-LT dans le système hématopoïétique de la souris. Sur la base de cette découverte, nous avons établi une lignée de souris rapporteures Hoxb5 et isolé avec succès les LT-HSC et les ST-HSC. Nous décrivons ici un protocole détaillé pour l’isolation des LT-HSC et des ST-HSC à l’aide du système de rapporteur Hoxb5 . Cette méthode d’isolement aidera les chercheurs à mieux comprendre les mécanismes d’auto-renouvellement et la base biologique d’une telle hétérogénéité dans le compartiment HSC.

Nous présentons un protocole étape par étape pour l’isolement des cellules souches hématopoïétiques à long terme (LT-HSC) et des CSH à court terme (ST-HSC) à l’aide du système rapporteur Hoxb5.

PROCÉDÉ D’ISOLEMENT DE CELLULES SOUCHES HÉMATOPOÏÉTIQUES À LONG ET À COURT TERME

La méthode d’isolement des cellules souches hématopoïétiques à long et à court terme ou des CSH aidera les chercheurs à mieux comprendre les mécanismes de renouvellement cellulaire et la base biologique de l’hétérogénéité dans le compartiment des CSH. Un facteur de transcription, Hoxb5, qui peut être un marqueur exclusif des CSH à long terme, a été identifié. Sur cette base, une lignée de souris rapporteures Hoxb5 a été établie et les CSH à long terme et à court terme ont été isolées avec succès.Pour commencer la collecte des cellules de moelle osseuse du donneur, stérilisez un mortier et un pilon avec de l’éthanol à 70% et laissez-les sécher complètement. Équilibrez ceux qui ont un tampon de coloration cellulaire composé de suppléments de PBS sans calcium et sans magnésium. Mettez les os isolés de la souris mâle donneuse Hoxb5-tri-mCherry dans le mortier et ajoutez trois millilitres du tampon de coloration cellulaire.Brossez les os avec le pilon, puis désagrégez les amas de cellules par pipetage doux et transférez la suspension cellulaire à travers une passoire cellulaire de 100 microns dans un tube de 50 millilitres. Répétez le processus avec un tampon de coloration cellulaire frais jusqu’à ce que la solution devienne claire. Habituellement, répéter trois fois suffit pour obtenir une solution claire.Après avoir isolé les fémurs et les tibias de la souris congénique CD45.1.2, couper les deux extrémités des os avec des ciseaux stériles tranchants. À l’aide d’une aiguille de calibre 23 munie d’une seringue de cinq millilitres remplie d’un tampon de suspension cellulaire glacé, rincer la moelle osseuse dans une boîte de culture cellulaire stérile contenant un tampon de suspension cellulaire. Désagrégez les amas de cellules par pipetage doux, puis filtrez la suspension cellulaire à travers une crépine cellulaire de 40 microns à l’aide d’une pipette P1000.Pour la configuration du déclenchement, préparez l’échantillon en suivant les instructions fournies dans le texte et transférez-en 400 microlitres dans un tube à essai en polystyrène à fond rond avec un capuchon de crépine de 35 microns. Préparer et ajouter un réactif de contrainte des cellules mortes à l’échantillon avant l’analyse, en suivant les instructions du fabricant. Ensuite, allumez le cytomètre en flux et lancez le logiciel d’analyse en suivant les instructions du fabricant.Ensuite, appuyez sur Charger pour commencer à acquérir les données. Après avoir exclu les doublets, les cellules mortes et les cellules de lignée positive, la fraction positive de la lignée négative, la fraction positive c-kit, Sca-1-positive. Ensuite, sortez la fraction positive Flk2.Ensuite, les CSPp Hoxb5-positives ou Hoxb5-négatives dans la fraction CD150-positive, CD34-négative ou faible. Ensuite, préparez un tube de liaison à faible teneur en protéines de 1,5 millimètre avec 600 microlitres de PBS sans calcium et magnésium, complété par 10% de FBS inactivé par la chaleur. Pour effectuer le tri pHSC Hoxb5-positif ou Hoxb5-négatif, placez le tube de liaison à faible teneur en protéines de 1,5 millilitre préparé sur un dispositif de collecte de tri et triez les pHSC Hoxb5-positifs ou Hoxb5-négatifs dans le tube en utilisant la stratégie de gating mentionnée précédemment.Dans le premier tri, utilisez le rendement du mode de précision de tri. Placez la plaque à 96 puits avec les cellules de support préalablement préparées sur l’étage unitaire de dépôt automatisé de cellules. Ensuite, réglez le tube de liaison à faible teneur en protéines de 1,5 millilitre sur le port de chargement du cytomètre en flux.Triez les CSPp Hoxb5-positives ou Hoxb5-négatives dans la plaque à 96 puits avec les cellules de support en utilisant la stratégie de gating mentionnée précédemment. Trier 10 CSPp Hoxb5-positives ou Hoxb5-négatives pour tester leurs capacités d’auto-renouvellement. Les diagrammes de cytométrie en flux de l’analyse de la moelle osseuse des souris donneuses Hoxb5-tri-mCherry ont révélé environ 20 à 25% des cellules de la fraction pHSC définie par lignée négative, c-kit-positive, Sca-1-positive, CD150-positive, CD34-négative ou faible, Flk2-négatif étaient des pHSCp Hoxb5-positifs.Voici les diagrammes de cytométrie en flux représentatifs de l’analyse du sang périphérique des souris receveuses. De plus, l’analyse du sang périphérique des souris receveuses après la primotransplantation a révélé que, bien que les CSPp Hoxb5 positives et Hoxb5 négatives présentaient un chimérisme de donneur similaire quatre semaines après la transplantation, une hématopoïèse continue n’a été observée que chez les receveurs de CSPp Hoxb5 positifs. Les CSH Hoxb5 négatives ont commencé à perdre la capacité de produire des cellules hématopoïétiques huit semaines après la transplantation.Dans l’analyse de transplantation secondaire, seuls les receveurs de CSPp Hoxb5-positifs présentaient une hématopoïèse robuste. En revanche, les cellules de donneurs ont été à peine observées chez les souris receveuses de CSPp Hoxb5 négatives, ce qui suggère que les CSPp Hoxb5 négatives ont perdu leur capacité d’auto-renouvellement dans les 16 semaines suivant la transplantation primaire. Ce protocole prend habituellement de neuf à 12 heures, il est donc important de maintenir l’échantillon à quatre degrés Celsius tout au long des procédures autant que possible pour maintenir la viabilité cellulaire.
