自己複製能と多能性を持つ造血幹細胞(HSC)は、造血ヒエラルキーの頂点に存在します1,2。1988年、Weissmanらは、フローサイトメトリー3を用いてマウス造血幹細胞の単離を達成できることを初めて実証した。その後、細胞表面マーカーの組み合わせによって定義される画分、系統-c-キット+Sca-1+CD150+CD34-/loFlk2-が、マウス4,5,6,7,8のすべての造血幹細胞を含むことが報告されました。
免疫表現型で定義された(系統-c-キット+Sca-1+CD150+CD34-/loFlk2-)造血幹細胞(以下、pHSC)は、以前は機能的に均質であると考えられていました。しかし、最近の単一細胞解析により、pHSCは自己複製能力9,10および多能性11,12に関して依然として不均一性を示すことが明らかになりました。具体的には、自己複製能に関して、pHSC画分には、継続的な自己複製能を有する長期造血幹細胞(LT-HSC)と、一過性の自己複製能を有する短期造血幹細胞(ST-HSC)の2つの集団が存在するようである9,10。
今日まで、LT-HSCとST-HSCを区別する自己複製能力の分子メカニズムはよくわかっていません。自己複製能力に基づいて両方の細胞集団を分離し、その根底にある分子メカニズムを発見することが重要です。LT-HSCを精製するために、いくつかのレポーターシステムも導入されています13,14,15;しかし、各レポーター系で規定されるLT-HSC純度は可変であり、これまで排他的なLT-HSC精製は達成されていません。
したがって、LT-HSCおよびST-HSCの分離システムを開発することで、pHSC画分の自己複製能力に関する研究が加速します。LT-HSCおよびST-HSCの単離において、多段階の偏りのないスクリーニングを用いた研究により、pHSC画分16に不均一に発現する単一の遺伝子Hoxb5が同定された。さらに、Hoxb5レポーターマウスの骨髄分析により、pHSC画分の約20%〜25%がHoxb5pos細胞からなることが明らかになりました。Hoxb5pos pHSCとHoxb5neg pHSCを用いた競合移植アッセイでは、Hoxb5pos pHSCのみが長期自己複製能を有するのに対し、Hoxb5neg pHSCは短期間で自己複製能を失うことがわかり、Hoxb5がpHSC画分16でLT-HSCを同定することが示された。
ここでは、 Hoxb5 レポーターシステムを使用してLT-HSCとST-HSCを分離するための段階的なプロトコルを示します。さらに、Hoxb5pos/neg pHSCの自己複製能力を評価するための競合移植アッセイを提示します(図1)。この Hoxb5 レポーターシステムは、LT-HSCとST-HSCを前向きに分離することを可能にし、LT-HSC特有の特性の理解に貢献します。
